Untersuchungen zur pH-Regulation am Blättermagenepithel des Schafes mit H+-sensitiven Mikroelektroden | Investigations on the pH regulation in the omasum epithelium of the sheep with H+-sensitive microelectrodes

Eine Hauptfunktion des Blättermagens liegt in der Absorption von kurzkettigen Fettsäuren und Bikarbonat. Diese Aufnahme bedingt effektive Mechanismen zur Regulation des intrazellulären pH-Wertes. Zur Charakterisierung der beteiligten Mechanismen ist im Rahmen dieser Arbeit eine neue Methode etabliert worden, mit der es möglich ist, den pH-Wert sowohl supraapikal (direkt an der apikalen Oberfläche des Epithels) als auch subapikal (in der Epithelzelle) zu messen. Diese double-barreled H+-sensitiven Mikroelektroden wurden in Kombination mit einer modifizierten Ussing-Kammer eingesetzt. Auf diese Weise konnten die Auswirkungen von veränderten Ionenkonzentrationen und von Hemmstoffen verschiedener Transportmechanismen auf den supra- und subapikal pH-Wert und auf die elektrophysiologischen Parameter aufgezeichnet werden. Mit Hilfe der Natriumreduktionsversuche, die zum Teil mit einer erhöhten Kaliumkonzentration kombiniert wurden, konnte bekräftigt werden, dass der transepitheliale Strom hauptsächlich von Natrium getragen wird. Außerdem wurde damit die Hypothese von Schultheiss (1995) bestätigt, der in dem apikalen Natriumkanal einen nicht-selektiven Kationenkanal sieht. Die hemmende Wirkung auf diesen Kanal wird von Calcium bzw. Magnesium durch eine Depolarisation (in diesem Fall bedingt durch eine luminale Kaliumerhöhung) aufgehoben. Auch die von Schultheiss postulierte Anwesenheit eines apikalen Kaliumkanals konnte mit diesen Versuchen bestätigt werden. Der parazelluläre Fluss von Natrium und Chlorid scheint hingegen gering zu sein. Die double- barreled H+-sensitiven Mikroelektrode wurde zur Messung des pH-Wertes etabliert. Die Elektroden hatten pro pH-Einheit eine Spannungsabweichung von 57,91 mV ± 2,65 (n = 58), die nahezu mit dem wünschenswerten Nernst-Potential übereinstimmten. Die routinemäßige Herstellung von Elektroden mit diesen idealen Eigenschaften erwies sich jedoch als zeitaufwendig und schwierig. Die Hypothese eines supraapikalen Mikroklimas konnte mittels dieser Mikroelektroden bestätigt werden. Der während der Standardinkubation gemessene pH-Wert direkt an der apikalen Oberfläche des Epithels war mit einem Wert von 7,23 ± 0,02 signifikant geringer als der pH-Wert (7,4) in der mukosalen Perfusionslösung (bulksolution). Die Messung des pH-Wertes in der Psalterepithelzelle ergab mit 7,30 ± 0,02 ebenfalls einen signifikant saureren pH-Wert im Vergleich zu dem in der bulksolution (Standardpuffer, pHWert 7,4). Wurde das Epithel mukosal mit einem fettsäurehaltigen Puffer (pH 7,4) inkubiert, fiel der pH-Wert in der Zelle unter dem des apikalen Puffers. Hatte der Fettsäurepuffer einen pH-Wert von 6,4, so konnte intrazellulär stets ein weitaus höherer Wert als im apikalen Medium gemessenen werden. Durch den Einsatz von Hemmstoffen bestimmter Transportmechanismen konnte gezeigt werden, dass die pH-Regulation der Psalterepithelzelle aus einem komplexen Zusammenspiel mehrerer Transportmechanismen besteht. So wird der Ausfall eines Mechanismus durch verstärkte Aktivität der Übrigen kompensiert. Zu diesen Transportern gehören in erster Linie die Natrium-Protonen-Austauscher, insbesondere der apikale NHE3. Dieser Transporter sorgt für die Ausschleusung der Protonen, die hauptsächlich über die Diffusion der protonierten kurzkettigen Fettsäuren in die Zelle gelangen. Zusätzlich zu dem NHE sind die Chlorid-Bikarbonat-Austauscher an der pHRegulation beteiligt. Der apikale Transporter (DRA) befördert Bikarbonat in die Zelle und Chlorid hinaus. Dies wird ermöglicht durch den transepithelialen Chloridgradienten und durch die Aktivität eines apikalen Natrium-Chlorid-Cotransporters, der für eine hohe (lokale) intrazelluläre Chloridkonzentration sorgt. Durch die Aufnahme von Bikarbonat können die Protonen in der Zelle neutralisier werden. Zusätzlich wird Natrium aufgenommen und Chlorid rezirkuliert. Serosal schleust der AE2 Transporter ein Bikarbonat-Ion im Austausch gegen ein Chlorid-Ion aus der Zelle heraus. Der Transporter arbeitet entsprechend des Chloridgradienten. Es finden sich auch funktionelle Hinweise auf die Aktivität einer apikalen vH+-ATPase, die ebenfalls Protonen aus der Zelle herausschleust. Sie scheint jedoch im Wesentlichen nach dem Ausfall anderer regulatorischer Mechanismen als „Notbremse“ zum Einsatz zu kommen. Eine zu starke Hyperpolarisation der apikalen Membran dürfte dabei durch einen verstärkten Einstrom von Natrium über den apikalen nicht-selektiven Kationenkanal verhindert werden. Die im Rahmen dieser Arbeit gewonnen Ergebnisse sprechen somit für eine rigorose Regulation der intrazellulären Protonenkonzentration des Psalterepithels. Fettsäuren gelangen apikal mit Protonen in das Epithel und werden serosal über verschiedene Mechanismen wieder ausgeschleust. Das im Austausch für Protonen aufgenommene Natrium wird basolateral über die Natrium-Kalium-ATPase aus der Zelle heraustransportiert. Der serosale Efflux der Fettsäureanionen wird vermutlich über den am Pansenepithel beschriebenen hochleitenden Anionenkanal erfolgen. Dieser Transport wird vom basolateralen Potential getrieben und ist somit funktionell an den Transport von Natrium gekoppelt.

A major function of the omasum is the absorption of short chain fatty acids and bicarbonate. This admission requires effective mechanisms for regulation of the intracellular pH. For the characterisation of the involved mechanisms a new method has been established within the framework of this research. With such method it is possible to measure the supraapical (directly on the apical surface of the epithelium) as well as the subapical (in the epithelium cell) pH value. These double barreled H+-sensitive microelectrodes were used in combination with a modified Ussing chamber. Thus the effects of changed ion concentrations and of inhibitors of different transport mechanisms could be noted on the supra- and subapical pH value and on the electrophysiological parameters. By means of the sodium reduction experiments, which were partly combined with a raised potassium concentration, it could be affirmed that the transepithelial current is mainly carried by sodium. Furthermore, the hypothesis of Schultheiss (1995) was confirmed in whose opinion the apical sodium channel as a non-selective cation channel. The restraining effect on this channel is abolished by calcium or magnesium caused through a depolarization (in this case due to a luminal potassium increase). The presence of an apical potassium channel, which Schultheiss postulated, could be also confirmed with these trials. However, the paracellular flux of sodium and chloride seems to be low. The double-barreled H+-sensitive microelectrode was established to measure the pH value. The electrodes had a voltage deviation of 57.91 ± 2.65 mV (n = 58) per pH unit, which almost agreed with the desirable Nernst-potential. The routinely manufacturing of electrodes with these ideal characteristics, however, proved to be time-consuming and difficult. The hypothesis of a supraapical microclimate could be confirmed by the application of these microelectrodes. Directly at the apical surface of the epithelium, during the standardincubation, the measured pH value was significantly lower (7,23 ± 0.02) than the pH value (7,4) in the mucosal perfusion solution (bulksolution). The results of the measurement of the pH value in the epithelium cell of the omasum were with 7.30 ± 0.02 also more acidic in comparison to the pH of the bulksolution (standard buffer, pH 7.4). If the mucosal epithelium was incubated with a fatty-acid buffer (pH 7,4), the pH value in the cell dropped under that of the apical buffer. If the fatty acid buffer had a pH value of 6,4, it was always possible to measure a much higher value in the cell than in the apical buffer. By using inhibitors of specific transport mechanisms it could be shown that the pH regulation of the psalter epithelium cell is represented by a complex interaction of several transport mechanisms. Thus the failure of one mechanism is compensated by reinforced activities of the remaining. These transporters are primarily the sodium-proton-exchanger, in particular the apical NHE3. Such transporter provides for the ejection of protons, which reach the cell mainly by the diffusion of the protonated short chain fatty acids. In addition to the NHE, the chloride bicarbonate exchangers are involved in the pHregulation. The apical transporter (DRA) carries bicarbonate into the cell and chloride out of the cell. This is enabled by the transepithelial chloride gradient and by the activity of an apical sodium-chloride-cotransporter, which ensures a high (local) intracellular chloride concentration. The protons in the cell can be neutralized by the admission of bicarbonate. Furthermore sodium is taken up, whereby chloride is recirculated. On the serosal side the AE2 transporter channels one bicarbonate ion in the exchange against one chloride ion out of the cell. The transporter works corresponded to the chloride gradient. There are also functional references to the activity of an apical vH+-ATPase, which also channels protons out of the cell. It seems to be used, however, basically after the failure of other regulatory mechanisms as an "emergency brake". An excessive hyperpolarization of the apical membrane might be prevented thereby by an increased influx of sodium across the apical non-selective cation channel. Therefore results gaindes within the scope of this thesis argue for a rigorous regulation of the intracellular proton concentration of the omasal epithelium. Fatty acids reach the epithelium apically together with protons and were discharged serosal again through different mechanisms. The sodium taken up in the exchange for protons is transported out of the cell basolateral over the sodium-potassium ATPase. The serosal efflux of the fatty acid anion probably occurs by the highly conductive anion channel described in the rumen.This transport is driven by the basolateral potential and is thus functionally linked to the transport of sodium.
