Untersuchungen zum Resistenzverhalten von Riemerella anatipestifer und Ornithobacterium rhinotracheale des Wirtschaftsgeflügels | Investigations on the antibiotic resistance of Riemerella anatipestifer and Ornithobacterium rhinotracheale of the commercial poultry

Ein großes Problem der bakteriellen Atemwegserkrankungen von Geflügel sind die Anforderungen an eine wirksame Behandlung der Herde, um die Gesundheit und den Tierschutz zu gewährleisten und wirtschaftliche Verluste zu verhindern. Die Verwendung und der Missbrauch von antimikrobiellen Substanzen ist derzeit ein großes Problem, da dies meist zu einer erhöhten Inzidenz von Antibiotika- resistenten Bakterien führt. Ziel dieser Arbeit war es daher, geeignete Testmethoden zur Bestimmung der antibiotischen Sensitivität von R. anatipestifer und O. rhinotracheale gegenüber derzeit verwendeten Wirkstoffen zu entwickeln und zu etablieren. Im ersten Teil der Arbeit wurden zur Identifizierung und Charakterisierung von R. anatipestifer- und O. rhinotracheale-Isolaten, die in den Jahren 2007 bis 2012 aus verschiedenen Geflügelarten isoliert wurden, biochemische und molekularbiologische Methoden angewandt. Insgesamt konnten 108 Feldisolate eindeutig als R. anatipestifer identifiziert werden, ebenso wie 100 Feldisolate als O. rhinotracheale. Das Ergebnis der Makro-Restriktionsanalyse zeigte eine große Vielfalt von DNA- Fingerabdrücken sowohl bei R. anatipestifer als auch bei O. rhinotracheale. Im Falle von R. anatipestifer konnte eine große Vielfalt von Fragmentmustern innerhalb der Entenisolate nachgewiesen werden. Während Isolate von Puten 100% genetische Ähnlichkeit innerhalb der Untergruppe zeigten. Im Falle von O. rhinotracheale wurden die Isolate der Serotypen A, B und E in gemeinsamen Subclustern gefunden. Die Bestimmung der antibiotischen Sensitivität von R. anatipestifer und O. rhinotracheale ist aufgrund der komplexen Kultivierungsansprüche und des ungewöhnlich häufigen Auftretens von Resistenz schwierig. Daher wurde im zweiten Teil der vorliegenden Arbeit die Wirkung verschiedener Parameter untersucht (Testmedium, Inokulationsdichte, Inkubationszeit und Inkubationsatmosphäre). Ziel war es die Bouillon- Mikrodilution als Standard-Prüfmethode für R. anatipestifer zu etablieren und validieren. Die erhaltenen Ergebnisse zeigten, dass Müller-Hinton-Bouillon II (MHB II) suppelentiert mit 2% lysiertem Pferdeblut (lPb) ein geeignetes Kulturmedium ist. Die Inkubation bei 37 ° C für 24 Stunden unter aeroben Bedingungen ergab visuell gut ablesbare Ergebnisse in den kommerziellen Sensititre-Mikrotiterplatten. Aufgrund fehlender Breakpoints wurden die epidemiologischen Cut-off-Werte sowie die minimale Hemmkonzentration MHK50 und MHK90-Werte mit Hilfe eines statistischen Programms bestimmt. Tiamulin und alle Substanzen, die zu den Klassen der Cephalosporine und Carbapeneme gehören, haben sich als wirksam erwiesen. Die multimodale Verteilung der R. anatipestifer-Isolate gegenüber den Fluorchinolonen zeigt die Aufteilung in eine empfindliche und eine unempfindliche Population und zeigt die Notwendigkeit quantitativer Empfindlichkeitstests. Bezüglich der Aminoglycoside und Tetracycline wurde der Großteil der MHK-Werte in den hohen Konzentrationsbereichen bestimmt. Für O. rhinotracheale konnte keine Kombination von Testparametern ermittelt werden, die zu visuell gut ablesbaren Ergebnissen in den Mikrotiterplatten führte. Stattdessen konnte Mueller- Hinton-Agar mit 5% lysiertem Pferdeblut (lPb) als Medium für den E-Test sowie für den Agar-Dilutionstest hergestellt werden. Nach 24 Stunden Inkubation bei 37 ° C war ein visuell gutes Ablesen der Platten möglich. Bei beiden Methoden können O. rhinotracheale-Isolate nur für wenige antimikrobielle Substanzen getestet werden. Die Verfügbarkeit von E-Teststreifen mit tiermedizinisch relevanten Wirkstoffen ist begrenzt und mit hohen Kosten verbunden. Das Agar- Dilutionsverfahren hingegen führt zu einem hohen Arbeitsaufwand und wird nur in speziellen Labors durchgeführt. Jedoch können beide Verfahren eine Alternative zu herkömmlichen Diffusionstests mit der Möglichkeit einer quantitativen MHK-Bestimmung bieten.

A major problem of bacterial respiratory diseases of poultry are the requirments for an effective treatment of the flock, in order to ensure the health, the animal welfare as well as to prevent economic losses. Use and misuse of antimicrobial substance is mostly lead to increase the incidence of antibiotic resistant bacteria and this is currently a huge problem. Therefore the aim of this thesis was focus on the development and establishment of suitable test methods to determine antibiotic sensitivity of R. anatipestifer and O. rhinotracheale to commen used drugs. The first part was the identification and characterization of R. anatipestifer and O. rhinotracheale isolates, which were isolated from different poultry species during the period 2007 to 2012 by using biochemical and molecular biological methods. A total of 108 field isolates could be clearly identified as R. anatipestifer, as well as 100 field isolates as O. rhinotracheale. The result of the macro-restriction analysis showed a large variety of DNA fingerprints in both, R. anatipestifer and O. rhinotracheale. In the case of R. anatipestifer, a large diversity of fragment patterns could be detected within the duck isolates, while isolates from turkeys showed 100% genetic similarity within the subgroup. In the case of O. rhinotracheale the isolates of serotypes A, B and E were found in common subclusters. The determination of the antibiotic sensitivity of R. anatipestifer and O. rhinotracheale is difficult, because of the complex cultivation requirements and the unusually frequent occurrence of resistance. Therefore, in the second part of the present work was aimed to study the effect of different parameters (Test medium, Inoculation density, Incubation time and Incubation atmosphere). The aim was to establish and validate Bouillon microdilution as a standard test method for R. anatipestifer. The obtained results proved that Mueller-Hinton broth II (MHB II) with 2% lysed horse blood (lPb) supplementation to be a suitable medium for cultivation. Incubation at 37 ° C for 24 hours under aerobic conditions gave visually good readable results in the commercial sensititre microtiter plates. Since, lack of Breakpoints the epidemiological cut-off values as well as the Minimum inhibitory concentration MIC50 and MIC90 values were determined with the help of a statistical program. Tiamulin and all substances belonging to the classes of cephalosporins and carbapenemes have been proved to be effective. The multimodal distribution of the R. anatipestifer isolates against the fluoroquinolones shows the division into a sensitive and as well as resistance population and shows the necessity of quantitative sensitivity tests. Concerning aminoglycosides and tetracyclins, the majority of the MIC values were determined in the high concentration ranges. For O. rhinotracheale no combination of test parameters could be established, which leads to visually clearly readable results in the microtiter plates. Instead, it was possible to establish a Mueller-Hinton-Agar with 5% lysed horse blood (lPb) as the medium for the E-test as well as for the agar dilution test. A visually well reading of the plates was possible after 24 hours of incubation at 37 ° C. With both methods, O. rhinotracheale isolates can only be tested for a few antimicrobial substances. The availability of E-test strips with veterinary relevant active substances is limited and connected with high costs. The agar dilution method, on the other hand, entails a high amount of work and will only be carried out in special laboratories. However, both methods can provide an alternative to conventional diffusion tests with the possibility of quantitative MIC deterrnination.
