Untersuchung von Einflussfaktoren auf die Qualität und Menge des Erstkolostrums unter Verwendung verschiedener Feldmethoden | Investigation of factors influencing the quality and quantity of first milking colostrum using different field methods

Das Erstkolostrum wird vom Drüsengewebe des Euters bereits vor der Kalbung gebildet und ist das Sekret, welches nach der Geburt beim ersten Melk- oder Saugvorgang gewonnen bzw. vom Neugeborenen getrunken wird. Im Unterschied zur reifen Milch zeichnet es sich vor allem durch eine vielfach höhere Konzentration der Immunglobuline (Ig) aus. Diese ist das entscheidende Kriterium für die Qualität des Erstkolostrums. Der Gastrointestinaltrakt des Neugeborenen ist unmittelbar nach der Kalbung für einen begrenzten Zeitraum in der Lage, die enthaltenen Ig zu absorbieren. Da beim Wiederkäuer während der Trächtigkeit keine maternalen Antikörper über das Blut auf den Fetus übertragen werden, ist dieser Weg die einzige Möglichkeit, das neugeborene Kalb mit den überlebensnotwendigen Abwehrstoffen zu versorgen. Diese passive Immunisierung schützt das Kalb bis zur vollständigen Ausreifung des eigenen Immunsystems. Nur wenn es unmittelbar nach der Geburt mit Erstkolostrum mit adäquater Ig-Konzentration in ausreichender Menge versorgt wird, kann eine belastbare passive Immunisierung erreicht werden. Somit stellen die Qualität und Menge des Erstkolostrums einen wesentlichen Faktor für die Kälbergesundheit dar, welche wiederum von entscheidender Bedeutung für die Wirtschaftlichkeit eines Milchviehbetriebes ist. Daher war die Zielstellung dieser Arbeit, das Kolostrometer und Brix-Refraktometer als geeignete Feldmethoden zur Beurteilung der Erstkolostrumqualität zu verifizieren. Durch die Erhebung zahlreicher Tier- sowie Stoffwechselparameter mit anschließender Überprüfung auf einen Zusammenhang zur Qualität und Menge des Erstkolostrums sollten zudem die Einflussfaktoren aufgezeigt werden. Zu diesem Zweck wurden von April 2013 bis Februar 2014 die Herden-, Fruchtbarkeits- sowie Abkalbedaten von 1949 Rindern der Rasse Holstein-Friesian erfasst. Zudem wurden im peripartalen Zeitraum Blutuntersuchungen und Rückenfettdickemessungen durchgeführt. Es wurde die Menge des Erstkolostrums bestimmt und die Qualität mit einem Kolostrometer sowie Brix-Refraktometer ermittelt. Als Referenzparameter für die Qualität wurde die TP-Konzentration im Labor bestimmt. Sowohl das Kolostrometer als auch das Brix-Refraktometer erwiesen sich als geeignete Feldmethoden, um die Qualität des Erstkolostrums unmittelbar im Milchviehbetrieb überprüfen zu können. Die Vorteile lagen jedoch beim Refraktometer in Hinblick auf Genauigkeit, Handling, Robustheit sowie der Temperaturunabhängigkeit, welche die bedeutendste Überlegenheit darstellt. Der Anteil von 22,4% an Erstkolostrum von schlechter Qualität mit weniger als 22 %Brix macht deutlich, wie wichtig die Kontrolle mithilfe einer der beiden Feldmethoden noch vor dem Verfüttern an das Kalb ist, um eine ausreichende IgG-Erstversorgung sicherzustellen und die Bildung einer belastbaren Immunkompetenz beim Neugeborenen zu ermöglichen. Bereits die separaten Regressionsmodelle für die primi- und pluriparen Tiere verdeutlichen den Unterschied in der Qualität und Menge des Erstkolostrums zwischen den verschiedenen Altersgruppen. Die Tiere der vierten Laktation bildeten dabei das Erstkolostrum mit der höchsten Qualität. Zudem erwiesen sich, im Falle der pluriparen Kühe, die Trockenstehzeit sowie der Kalbemonat als signifikante Einflussfaktoren und es zeigte sich ein Zusammenhang der Qualität des Erstkolostrums zur Ca-Serumkonzentration des Muttertieres zum Zeitpunkt der Kalbung. Im Modell der primiparen Rinder verblieb dagegen einzig der Kalbemonat als erklärende Variable. Wie sich zeigte, wird die Menge des Erstkolostrums der Pluriparen von der Gesamttrockenstehzeit, dem Kälbergeburtsgewicht sowie dem Kalbemonat und vom Mitarbeiter, welcher die Kuh als erstes melkt, signifikant beeinflusst. Auch wurde ein Zusammenhang zwischen der Menge und der NEFA-Serumkonzentration zum Zeitpunkt der Geburt deutlich. Die Tiere der vierten Laktation bildeten am meisten Erstkolostrum. Im Regressionsmodell der Primiparen verblieben hingegen die Vorbereitungszeit, das Geburtsgewicht der Kälber sowie der Monat der Kalbung als signifikante Einflussfaktoren und es wurde ein Zusammenhang der Menge zur Ca- Serumkonzentration deutlich. Auffällig war, dass sich für die pluriparen Kühe zudem ein negativer Zusammenhang zwischen der Qualität und Menge des Erstkolostrums ergab. Dennoch bildeten die Tiere der vierten Laktation sowohl am meisten als auch das hochwertigste Erstgemelk. Diese Ergebnisse sowie die Vielzahl der Einflussfaktoren auf die Qualität und Menge des Erstkolostrums verdeutlichen die Komplexität der Zusammenhänge und unterstreichen, wie wichtig die Kontrolle eines adäquaten Ig-Gehalts mithilfe einer geeigneten Feldmethode ist. Die Überprüfung der Qualität des Kolostrums beim ersten Melken sollte daher fester Bestandteil im Betriebsablauf sein, um die Kälbergesundheit zu gewährleisten und zu optimieren.

First milking colostrum is already formed in the udder’s glandular tissue before calving and is secreted after parturition at the first milking or first suctioning of the neonatal calf. It is distinguished from mature milk above all by the much higher concentration of immunoglobulins (Ig). This concentration is the decisive criterion for the colostrum’s quality. The newborn calf’s gastrointestinal tract for a short time immediately after calving is capable of absorbing the Ig present in the colostrum. Since pregnant ruminants do not transfer maternal antibodies through blood to the fetus, this is the only opportunity to supply the newborn calf with these antibodies essential to its survival. This passive immunization protects the calf until its own immune system has matured completely. A reliable passive immunization will only be achieved if calves receive a sufficient quantity of colostrum immediately after parturition, containing an adequate concentration of Ig. Consequently, the quality and quantity of colostrum at first milking represent crucial factors for calf health, which, in turn, is decisive for a dairy farm’s economic viability. Therefore the objective of this study was to verify the colostrometer and Brix refractometer as suitable field methods for evaluating colostrum quality. Additionally the aim was to demonstrate the influencing factors by surveying numerous animal and metabolism parameters, followed by analyzing the relationship of these parameters to the quality and amount of first milking colostrum. To this end, herd, fertility and calving data were collected from April 2013 to February 2014 of 1949 cows of the Holstein-Friesian breed. In addition, during the antepartum timeframe, blood tests were carried out and backfat thickness (RFD) was measured. The amount of colostrum was determined and its quality detected with a colostrometer and a Brix refractometer. Total protein (TP) concentration was established in the laboratory as the quality reference parameter. Both the colostrometer and the Brix refractometer proved to be suitable field methods for evaluation of colostrum quality on site in the dairy farm. However, the refractometer had a clear perk when it came to precision, handling, robustness and temperature compensation, which represents its most significant advantage. The 22.4% share of low-quality colostrum with less than 22 %Brix clearly shows the importance of testing with one of the field methods even before the calf is fed, to ensure a sufficient first supply with Ig and make possible the building up of a reliable immune competence in the neonatal calf. Separate regression models for the primi- and pluriparous animals readily point up the difference in quality and quantity of colostrum between different age groups. Animals in the fourth lactation produced the best quality colostrum. It also became clear that length of dry (non-lactating) period as well as the month of calving were significant influence factors for the pluriparous cows and furthermore the quality correlated with the calcium (Ca) concentration in serum of the dam at the time of calving. However, for the model of the primiparous cows only the month of calving remained the explanatory variable. It was demonstrated that the amount of colostrum of the pluripara was significantly influenced by the length of total dry period, the calving birth weight, the calving month and the worker that milked the cow first. A link also emerged between the amount and the concentration of non-esterified fatty acids (NEFA) in the serum of the dam at the time of birth. The animals of the fourth lactation produced the most colostrum. For the primipara regression model, on the other hand, significant influencing factors remained length of preparation period, calf birthweight, calving month and a correlation between amount and serum concentration of Ca at the time of birth was established. Of interest is that, in the case of the pluriparous cows, there was shown a negative correlation between the quality and the quantity of first milking colostrum. Nonetheless, the animals in their fourth lactation produced the most as well as the highest-quality colostrum. These findings, and the multiplicity of factors influencing quality and quantity point up the complexity of the relationships and underline how crucial testing of an adequate Ig content with a suitable field method is. Checking the colostrum quality at the first milking should therefore be a regular feature in dairy farm operations flow if calf health is to be ensured and optimized.
