Estresse oxidativo em resposta ao alumínio em aveia branca | Oxidative stress in response to aluminum in oat

A toxidez por alumínio (Al) é um dos fatores limitantes para a produção agrícola em solos ácidos, os quais abrangem aproximadamente 60% do território brasileiro. Evidências têm relacionado à tolerância ao Al com mecanismos de detoxificação de espécies reativas de oxigênio, uma vez que, além de inibir o desenvolvimento normal da raiz, o excesso de alumínio interfere nas reações enzimáticas. Sua absorção através da parece celular contribui para indução do estresse oxidativo, levando à oxidação de biomoléculas como lipídios, pigmentos e ácidos nucléicos. Com o objetivo de avaliar o efeito da exposição ao Al sobre o metabolismo oxidativo dos genótipos tolerantes UFRGS 17 e UPF 91Al-100-1-4 e sensível UFRGS 930598 de aveia branca, foram feitas análises fisiológicas, genéticas e bioquímicas em plântulas sob condição hidropônica após sete dias de tratamento com 740 μM de AlCl3. Foram analisadas as enzimas superóxido dismutase (SOD), ascorbato peroxidase (APX), catalase (CAT) e glutationa redutase (GR); o conteúdo de tióis não protéicos (NPSH) e ascorbato (AsA), assim como os níveis de clorofila A , B e carotenóides. O estresse oxidativo foi estimado através do teor de MDA, que avalia os níveis de peroxidação de lipídeos e pelo teor de H2O2. Constatou-se que o Al induz estresse oxidativo em aveia e, que os genótipos tolerantes UFRGS 17 e UPF 91Al100-1-4 apresentam mecanismos eficientes de detoxificação. A peroxidação de lipídeos é significativa apenas no genótipo sensível, há um decréscimo na produção de H2O2 na presença do Al, o mesmo é verificado para as atividades enzimáticas da CAT e APX. Os genótipos tolerantes aumentam a atividade de SOD e GR, assim como o teor de AsA e NPSH. Os genótipos tolerantes apresentam maior crescimento radicular comparado ao sensível e a tolerância ao Al nestes genótipos é governada pelo mesmo gene. A diferente deposição de Al nos tecidos da raiz entre os genótipos tolerantes sugere que o mecanismo de tolerância não seja de detoxificação externa, mas, possivelmente, relacionado à redução do estresse oxidativo causado pelo Al.

The toxicity of aluminum (Al) is one of the limiting factors to agriculture in acid soils, which cover approximately 60% of Brazilian territory. It is suggested that Al tolerance mechanisms is related to reactive oxygen species detoxification. Once Al not only inhibits the normal root development but also interferes with enzymatic reactions. Al uptake contributes to oxidative stress induction, leading to oxidation of biomolecules such as lipids, pigments and nucleic acids. In order to evaluate the effect of Al on the oxidative metabolism of tolerant oat genotypes (UFRGS 17 and UPF 91Al-100-1-4), and sensitive UFRGS 930598 oat, physiological, genetic and biochemical analysis were conducted under hydroponics after seven days of treatment with 740μM AlCl3. We analyzed the enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT) and glutathione reductase (GR), the content of non-protein thiols (NPSH) and ascorbate (AsA), and the levels of chlorophyll A, B and carotenoids. Oxidative stress was estimated by H2O2 levels and MDA content, which assesses the level of lipid peroxidation. It was found that Al induces oxidative stress in oat and the tolerant genotypes UFRGS 17 and UPF 91Al100-1-4 have efficient mechanisms of detoxification. The lipid peroxidation is significant only in the sensitive genotype. There is also a decrease in H2O2 production in the presence of Al. The activities of CAT and APX are reduced upon Al exposure. The tolerant genotypes increase the activity of SOD and GR, as well as the content of AsA and NPSH. The tolerant genotypes have longer roots compared to the sensitive one. Al tolerance in these two genotypes is governed by the same gene. The different Al deposition in the root tissues of tolerant genotypes suggests that the mechanism of tolerance is not external detoxification, but possibly related to reduction of the oxidative stress caused by Al.
