Detecção de Burkholderia spp. associadas a sementes de arroz nacionais e importadas | Detection of Burkholderia spp. associated to national and imported rice seeds

Burkholderia gladioli, B. glumae e B. plantarii causam o crestamento bacteriano da panícula e a podridão das plântulas do arroz e são transmitidas pelas sementes, mas não têm registro no Brasil. Considerando os registros do aumento do potencial de dano destas doenças à cultura do arroz no mundo e o intenso comércio nacional e internacional, esta pesquisa foi conduzida com o intuito de detectar, isolar e determinar as possíveis espécies presentes nas sementes de arroz. Utilizou-se 342 amostras, 290 de sementes importadas, onde 283 da Argentina, seis dos EUA,uma do Uruguai, e 52 amostras nacionais do RS, recebidas pelo Laboratório Agronômica. Para o isolamento e PCR, 5 g de cada amostra de sementes foram agitados em 15 ml de PBS e Tween 20. Alíquotas (200 μl) foram incubadas em meio líquido para o enriquecimento e o restante, após adicionar 300 mg de PVPP e 150 mg de Na2SO3, foi agitado, sonicado e filtrado, sendo utilizado diretamente na qPCR com sonda para o gênero Burkholderia, qPCR com SYBR® Green para detectar a espécie B. glumae e PCR multiplex para as espécies B. glumae, B. gladioli e B. plantarii, conforme protocolo de extração proposto para Xoo/Xoc (Vera Cruz et al., 2012). Colônias translúcidas, elevadas, com margens definidas, brancas acizentadas ou amarelas, Gram -, catalase +, oxidase +/-, RH + em tabaco, foram submetidas à qPCR e aos sistemas GN2 e GEN lll (Biolog Inc., CA, EUA). A presença de B. glumae foi detectada em duas amostras de sementes importadas dos EUA. B. vietnamiensis, não fitopatogênica, foi detectada e confirmada em uma amostra nacional e duas importadas da Argentina. Embora os resultados positivos com as amostras de sementes nacionais na qPCR e PCR para B. glumae, isolados bacterianos não foram obtidos, não permitindo a confirmação da presença desta praga no Brasil. Em virtude da relevância da cultura do arroz no país e a confirmação de B. glumae em sementes importadas, a utilização do método de isolamento adaptado do IRRI, associado à qPCR, deve ser adotado nas análises de sementes importadas.

Burkholderia gladioli, B. glumae and B. plantarii cause bacterial blight and rot panicle of rice seedlings and are transmitted by seeds, but do not have registration in Brazil. Considering the records of the increase in damage potential of these diseases to the rice crop in the world and the intense national and international trade, this research was conducted in order to detect, isolate and determine the possible species present in rice seeds. We used 342 samples, 290 of imported seed, where 283 of Argentina, six USA and a sample of Uruguay, and 52 national samples of RS received by the Laboratory Agronômica. For the isolation and PCR, 5 g of each sample seeds were stirred in 15 ml of PBS and Tween 20. Aliquots (200 μl) were incubated in liquid medium for the enrichment and the remainder, after adding 300 mg of PVPP and 150 mg Na2SO3, stirred, sonicated and filtered, and used directly in qPCR probe for the genus Burkholderia, qPCR with SYBR® Green to detect the species B. glumae and PCR multiplex for the species B. glumae, B. gladioli and B . plantarii as extraction protocol proposed for Xoo / Xoc (Vera Cruz et al., 2012). Translucent, elevated colonies, with greyish or yellow, Gram -, catalase +, oxidase +/-, HR + in tobacco were submitted to qPCR and GN2 and GEN III systems (Biolog Inc., CA, USA) . The presence of B. glumae was detected and confirmed two samples of seeds imported from the USA. B. vietnamiensis, not plant pathogen, was detected and confirmed in a national sample and two imported from Argentina. Although the positive results with the national seed samples in qPCR and PCR for B. glumae, bacterial isolates were not obtained, not allowing for confirmation of the presence of this pest in Brazil. Based on the relevance of rice in the country and the confirmation of B. glumae in imported seeds, the method of isolation adapted from IRRI, associated with qPCR, should be adopted in the analysis of imported seeds.
