Therapeutic doses of carbon tetrachloride produced pathological changes in biopsy specimens of liver, although biochemical methods of detection of liver damage were inconsistent. The evidence supported the hypothesis that fasciolocidal drugs depend for their action on producing some damage to the liver. 1. Bromsulpthalein clearance (B.S.P.), plasma alkali ne. phosphatase, plasma proteins, serum calcium, serum magnesium, inorganic phosphorus, blood urea, blood sugar, and liver biopsy were studied in seven poorly fed and three well fed sheep, both before and after treatment with carbon tetrachloride. 2. The blood sugar rose in three out of five poorly fed sheep, and fell in all the well fed sheep. 3. The plasma alkaline phosphatase fell in five of the poorly fed sheep and in two well fed sheep. 4. All sheep except one showed lowered B.S.P. clearance after carbon tetrachloride. 5. Serum calcium fell in three poorly fed sheep and in two out of three well fed sheep rose. The serum magnesium fell in each of three poorly fed and three well fed sheep. The inorganic phosphate fell in three poorly fed sheep and rose in three well fed sheep. 6. Blood urea fell in four poorly fed sheep and rose in two of the well fed group. 7. Some of the protein fractions separated by serum electrophoresis rose and some fell. The albumin globulin distribution in fluke infected sheep was markedly different from normal, the gamma globulin fraction particularly being elevated. 8. Biopsy at eighteen hours showed early necrosis in every case usually accompanied by nellui nr infiltration. Glycogen decreased in all cases, alkaline phosphatase activity increased as did periportal and centre lobular fat. The control sections of poorly and well fed sheep were found to be different.
