Немецкий. Deutsch: Erhöhte IL-10-Konzentrationen im Plasma von HIV-positiven Patienten legen nahe, dass HIV das immunsuppressive Zytokin IL-10 zur Immunevasion induziert. Zusätzlich wurde eine hoch konservierte immunsuppressive Domäne, die für die Produktion von IL-10 verantwortlich ist, im retroviralen Transmembran-Env und in Syncytinen gefunden. Es konnte jedoch nicht geklärt werden, welche Zellen für die IL-10-Produktion verantwortlich sind und ob auch retrovirale Oberflächen-Env einen immun-suppressiven Effekt haben. Um solche Zusammenhänge zwischen retroviralen Infektionen und dem Immunsystem besser zu verstehen, wird unter anderem das FV‑Modell genutzt. Um diese Zusammenhänge zu verstehen, ist ein detailliertes Verständnis der Zielzellen und der Kinetik der Infektion durch F-MuLV und SFFV in der akuten Phase der FV-Infektion bei suszeptiblen Mäusen erforderlich. In dieser Arbeit wurden detaillierte Einblicke gewonnen in die Kinetik von F-MuLV und SFFV in der akuten Phase der FV-Infektion im Knochenmark, in der Milz und in Lymphknoten durch die Infektion von suszeptiblen Mäusen mit dem doppelt fluoreszierend markierten FV-Katushka-mTagBFP. Zudem wurden IL-10-Reportermäuse mit FV-Katushka-mTagBFP infiziert, um die infektionsinduzierte IL-10-Produktion in FV‑Zielzellen zu ermitteln. In frühen Stadien der akuten FV-Infektion war ein signifikanter Anstieg der IL-10-Produktion in fast allen infizierten FV-Zielzellen im Knochenmark, in der Milz und in Lymphknoten, festzustellen. Des Weiteren konnte gezeigt werden, dass die Steigerung der IL-10-Produktion nicht auf F-MuLV infizierte Zellen beschränkt ist, sondern auch das replikationsdefiziente, pathogene SFFV zur Immunsuppression beiträgt. Die Infektion von T-Zell-IL-10-Knockout-Mäusen zeigte, dass eine fehlende IL-10-Produktion durch T-Zellen zu einer geringeren Viruslast in Mäusen führte, was mit einer signifikant höheren Anzahl aktivierter CD4+ und CD8+ T‑Zellen in Zusammenhang gebracht werden kann. Auch bei Mäusen, die eine DNA-Immunisierung mit Plasmiden, die für das gesamte F‑MuLV-Env codieren, erhalten hatten, ging eine vermehrte IL-10-Produktion von CD4+ T-Zellen, aber nicht von myeloiden Zellen aus. Weitere DNA-Immunisierungsversuche mit Plasmiden, die für retrovirale Oberflächen-Env codieren, zeigten, dass immunsuppressive Eigenschaften nicht nur auf das Transmembran-Env beschränkt sind, sondern auch im Oberflächen-Env zu finden sind. DNA- Immunisierungen mit unterschiedlichen retroviralen Oberflächen-Env-codierenden Plasmiden deuteten auf eine Konservierung der für den immunsuppressiven Effekt verantwortlichen Sequenz hin. Um darzulegen, dass der immunsuppressive Effekt nicht nur auf die Co-Immunisierung mit einem CD8+ T-Zell-Immunogen beschränkt ist, sondern auch einen negativen Einfluss auf das Tumorwachstum und die Tumorkontrolle in vivo hat, wurden Mäuse mit Env-codierenden Plasmiden geimpft und anschließend Tumorzellen injiziert. Diese Mäuse zeigten ein deutlich stärkeres Tumorwachstum im Vergleich mit unbehandelten Mäusen. Die Injektion von einem blockierenden anti-IL-10-Atnikörper konnte den Effekt aufheben, was die mechanistische Rolle von IL-10 in der Suppression der Tumorkontrolle unterstreicht. Mit diesen Ergebnissen konnten neue Einblicke in die Kinetik der FV-Infektion, vor allem des SFFV, erlangt werden. Des Weiteren wurde dargelegt, dass die Env-induzierte IL-10-Produktion in FV-Zielzellen, einen signifikanten Einfluss auf die antivirale und Antitumor-Immunantwort in vivo hat und diese nicht nur durch die bekannte immunsuppressive Domäne, sondern auch von dem Oberflächen-Env induziert wird. In addition, a highly conserved immunosuppressive domain responsible for IL-10-production was found in retroviral transmembrane Env and syncytins. However, it could not be clarified which cells are responsible for IL-10-production and whether retroviral surface Env also has an immunosuppressive effect. To better understand such interactions between retroviral infections and the immune system, the FV infection is often used as a model infection. A detailed understanding of the target cells and kinetics of infection by F-MuLV and SFFV in the acute phase of FV infection in susceptible mice is necessary for understanding of the anti-retroviral immune response. In this work, detailed insights into the kinetics of F-MuLV and SFFV in the acute phase of FV infection in bone marrow, spleen, and lymph nodes were obtained by infecting susceptible mice with the double fluorescently labeled FV-Katushka-mTagBFP. In addition, IL-10-reporter mice were infected with FV-Katushka-mTagBFP to determine infection-induced IL-10-production in FV target cells. In early stages of acute FV infection, a significant increase in IL-10-production was seen in almost all FV-infected target cells in bone marrow, spleen, and lymph nodes. Furthermore, it was shown that the increase in IL-10-production was not limited to F-MuLV infected cells, but that the replication-deficient pathogenic SFFV contributed to immunosuppression. Infection of T cell IL-10 knockout mice showed that lack of IL-10-production by T cells resulted in lower viral loads in mice, which can be associated with significantly higher numbers of activated CD4+ and CD8+ T cells. Similarly, in mice that received DNA immunization with plasmids encoding the entire F‑MuLV-Env, increased IL-10-production originated from CD4+ T cells but not from myeloid cells. Further DNA immunization experiments with plasmids encoding retroviral surface Env showed that immunosuppressive properties were not limited to the transmembrane Env but can also be found in the surface Env. DNA immunization with different retroviral surface Env-encoding plasmids indicated conservation of the gene sequence responsible for the immunosuppressive effect. To demonstrate that the immunosuppressive effect is not limited to co-immunization with a CD8+ T cell immunogen but also has a negative impact on tumor growth and tumor control in vivo, mice were immunized with Env-encoding plasmids and subsequently injected with tumors. These mice showed significantly increased tumor growth compared with untreated mice. Injection of a blocking anti-IL-10 antibody was able to abolish the effect, highlighting the mechanistic role of IL-10 in suppression of tumor control. These results provided new insights into the kinetics of FV infection, especially of SFFV. Furthermore, it was outlined that Env-induced IL-10-production in FV target cells has a significant impact on the antiviral and antitumor immune response in vivo and this is induced not only by the known ISD but also by the surface Env.
