Extraction, characterization, and valorization of biomolecules issued from coffee and cacao transformation | Extraction, caractérisation et valorisation de biomolécules issues de la transformation du café et du cacao

The processing of coffee and cocoa generates abundant by- and co-products, such as coffee pulp (CP), cocoa pod husk (CH), and cocoa husk (CH), which represent an interesting, source of biomolecules. However, very often, these residues are bad managed and provoque social, economic, and environmental problems. In this context, the main objective of this research work was to offer new perspectives for their valorization by: (i) obtaining extracts rich in biomolecules (e.g. phenolic compounds (PC)) from CP, CPH, CH, and green coffee, using green solvents (Deep Eutectic solvents - DES) ; (ii) evaluating the biological activity in vivo (rat model; green coffee extract) and the antioxidant activity in vitro (CP, CPH, and CH extracts), using the VesiCAT (Vesicle Congugated Autoxidizable Triene) assay,, and (iii) completing the actual portfolio of molecules with interesting physicochemical and/or biological activities through the research of isoprotanoids (phytoprostanes (PhytoPs) and phytofuranes (PhytoFs)) and thiol precursors, which have never been reported on coffee and cocoa residues.DES result from the mixture of an organic salt (ammonium of phosphonium) that acts as a hydrogen bond acceptor (HBA) and a hydrogen bond donor (HBD). The obtained mixture has a melting point inferior to those of the separated compounds. Six DES made of cholinium chloride or betain (HBA) and lactic acid, glycerol or 1,4-butanediol (HBD) and EtOH 70 % (v/v) (control) were tested, and extraction was assisted by heating-stirring (HSE) or by a ultrasounds probe (UPAE). Total phenolic compounds (PC) was determined by Folin-Ciocalteu method and by HPLC, and extracted compounds were characterized by UPLC-MS. The best extraction PC rates were obtained with DES CCLA (cholinium chloride:lactic acid:water, 1:2:1.5 mole ratio) and UPAE. Three compounds were extracted and identified from the residues: chlorogenic acid (CP), caffeine (CP, CH), and theobromine (CH). No compound was neither extracted nor identified from CPH under the tested conditions. During this extraction, furfural was formed as a result of the acidic deshydration of pentoses liberated from the hemicelluloses hydrolysis catalyzed by DES in CP, CPH, and CH.The study of the antioxidant capacity of CP, CPH, and CH ethanolic extracts, showed that the VesiCAT assay could represent a useful tool to examine the antioxidant performance of natural extracts.The effect of a DES formulation rich in chlorogenic acids, obtained from green coffee beans, was studied in vivo in rats. Even though results are preliminary and complementary work is needed, this study is pioneer, and would contribute to understand the factors and risks associated with in vivo DES application.The isoprostanoids and thiols precursors identification for the very first time in coffee and cocoa by- and co-products widened the portfolio of molecules reported in CP, CPH, and CH, and paved the way to new valorization perspectives of these residues.This work has therefore contributed to enrich the knowledge of the portfolio of high value-added biomolecules present in CP, CPH, and CH, through the practice of green Chemistry by using environmentally friendly solvents for their extraction.

La transformation de café et cacao génère de grandes quantités de sous- de co-produits tels que la pulpe de café (CP), la cabosse de cacao (CPH) et la coque de cacao (CH), qui représentent une source intéressante de biomolécules. Cependant, très souvent, ces résidus sont mal gérés et provoquent d’importants problèmes sociaux, économiques et environnementaux. Dans ce contexte, l’objectif de ce travail de thèse a été d’offrir des nouvelles perspectives pour la valorisation de ces résidus afin de : (i) obtenir des extraits riches en biomolécules (e.g. composés phénoliques (PC)) à partir de CP, CPH, CH et café vert, en utilisant des solvants verts (Deep Eutectic solvents - DES), (ii) l’activité biologique in vivo (modèle rat ; extrait café vert)et évaluer l’activité antioxydante in vitro (extraits de CP, CPH et CH) en utilisant la méthode VesiCAT (Vesicle Congugated Autoxidizable Triene) et (iii) compléter l’actuel portefeuille de molécules possédant des propriétés physicochimiques et/ou biologiques intéressantes issues de CP, CPH et CH à travers la recherche d’isoprostanoïdes (phytoprostanes (PhytoPs) et des phytofuranes (PhytoFs) et de précurseurs de thiols, dont la présence dans ces résidus n’a jamais été décrite.Les DES consistent en un mélange d’un sel organique (ammonium ou phosphonium) qui fonctionne comme un accepteur de liaisons hydrogène (HBA) et d’un donneur de liaisons hydrogène (HBD). Le mélange obtenu possède un point de fusion inférieur à celui des constituants pris individuellement. Six DES à base de chlorure de cholinium ou bétaïne (HBA) et d’acide lactique, glycérol ou 1,4-butanediol (HBD) et l’EtOH 70 % (v/v) (témoin) ont été testés et l’extraction a été assistée d’une agitation et chauffage (HSE) ou d’ultrasons à l’aide d’une sonde (UPAE). La teneur en PC totaux des extraits a été déterminée par la méthode de Folin-Ciocalteu et par HPLC et l’identification des composés extraits a été faite par UPLC-MS. Les meilleurs taux d’extraction de PC ont été atteints en utilisant le DES CCLA (chlorure de cholinium:acide lactique:eau, 1:2:1,5 ratio molaire) et l’UPAE. Trois composés ont été extraits et identifiés dans les résidus : acide chlologénique (CP), caféine (CP, CH) et théobromine (CH). Aucun composé n’a été extrait ni identifié dans la CPH, avec les conditions d’analyse utilisées. Lors de cette extraction, du furfural s’est formé suite à la déshydratation acide des pentoses libérées de l’hydrolyse catalysée par le DES des hémicelluloses présentes dans CP, CPH et CH.L’effet d’une formulation DES riche en acides chlorogéniques, obtenue à partir des grains de café vert, a été étudié in vivo chez le rat. Bien que les résultats obtenus sont préliminaires et que des travaux complémentaires sont nécessaires, cette recherche est pionnière et contribuerait à comprendre les facteurs et les risques liés à l’application des DES in vivo.L’étude de la capacité antioxydante des extraits éthanoliques de CP, CPH et CH, a montré que la méthode VesiCAT pourrait constituer un bon outil utile pour évaluer les performances antioxydantes d’extraits naturels.L’identification et la quantification pour la première fois des PhytoPs, PhytoFs et précurseurs de thiols a permis d’élargir le portfolio actuel des biomolécules rapportées dans la CP, la CH et la CPH, et ouvre la voie à des nouvelles perspectives de valorisation de ces résidus.Nous avons donc contribué à enrichir la connaissance du portfolio de biomolécules à haute valeur ajoutée présentes dans les résidus issus de la transformation du café et du cacao, tout en utilisant la chimie verte à l’aide des solvants respectueux de l’environnement pour leur extraction.
