DNA barcoding and metabarcoding as complementary approaches to investigate the impact of predation by an invasive species on earthworm communities

Earthworm communities in Europe may be severely disrupted by theintroduction of the exotic predator Obama nungara, a terrestrial Platyhelminthe native fromSouth America.Earthworm community structure and diversity were explored during three years in variousanthropized sites in France invaded by the flatworm, using morpho-molecular approaches.This made it possible to follow the temporal dynamics of the communities, in relation withthe variation of abundance of O. nungara on the sites. Sites where the flatworm were mostabundant experienced biodiversity loss, with a decrease in abundance of rarest earthwormspecies.DNA barcoding of the samples allowed to constitute libraries of DNA barcodes (COI and 16S)of the 21 morphospecies collected in the different sites corresponding to 36 MOTUs.This database was then used to analyse the metabarcoding data of the digestive contents of86 O. nungara collected on the sites: reads obtained from sequencing were affiliated toMOTUs from the database, allowing taxonomic identification of prey DNA from digestivecontents.Thus, the diversity of earthworm species consumed by the flatworm could be related todiversity and abundance of earthworm species on the sites. 26 earthworm species weredetected in the digestive contents, thus O. nungara seems to present a large dietary diversity.The complementary use of DNA barcoding and metabarcoding approaches therefore makesit possible to explore questions such as the availability of earthworm prey for the flatwormdepending on the microhabitat occupied (galleries within soil, litter) and preferentialpredation choice.
