As far as it could, be ascertained this work represents the first real attempt to define the factors that influence the HI test with FPLV. Most workers prefer to use the SN test, a procedure which is time consuming, laborious and not easily applicable as a routine diagnostic testo The techniques of HA and HI are so simple that they easily find a place as sérodiagnostic tools in the investigation of many other diseases in most laboratories. The test was defined by pH, the number of HA units of virus, RBC concentration, incubation, temperature, incubation time, non-specific agglutinins and inhibitors. The optimum pH was 6.8. HI titres varied in inverse proportion to the number of HA units of virus« For every two-fold increase in the number of HA units HI titres were depressed three-fold. A strong positive correlation was found between HI titres and RBC concentration. For every one fold increase in RBC concentration HI titres were increased eight-fold. The minimum incubation time was 3 hours. 4°C was the only suitable operating temperature. The test was influenced to a significant degree by the presence of non-specific agglutinins and inhibitors. Their removal by heat, followed by kaolin and RBC absorption was necessary before specific HI could be discerned.
