Effets fonctionnels respiratoires induits par l'inhalation de glycopyrrolate chez le cheval poussif en crise

Английский. An acute respiratory attack with bronchospasm is associated with excessive stimulation of the parasympathic nervous system. It may be reduced by administration of anticholinergic agents. Atropine and ipratropium bromide have been tested. This study is to evaluate the efficacy of the synthetic antimuscarinic compound glycopyrrolate as a bronchodilator in horses. Respiratory crisis was induced in seven allergic horses by exposure to natural allergens. Prior to the study, it was established that the attack could be stopped using intravenous atropine sulphate. Pulmonary function tests (ventilation and arterial blood gas analysis) were performed on the horses before and then after aerosol administration of 1500 µg of glycopyrrolate (Robinul®). The attack was characterised by an increase in total pulmonary resis¬tance and maximum pleural pressure variation and by decreased compliance. This study demonstrates that glycopyrrolate exhibits affects that are just as rapid and effective as intravenous atropine or aerosol administration of 1200 µg dry powder Ipratropium. In addi¬tion the treatment was well tolerated and no side-effects were observed in contrast to those seen with intravenous administration of atro¬pine. Therefore this drug has an important role to play among the therapeutics available to practitioners for acute respiratory attacks in horses

Французский. L'apparition d'une crise aiguë de pousse s'accompagne de bronchospasme lié à une stimulation excessive du système nerveux parasympathique. Il peut dès lors être levé par l'administration d'agents anti-cholinergiques. L'atropine et l'ipratropium bromide ont déjà été testés. Le présent travail a pour but d'évaluer l'efficacité du glycopyrrolate, une molécule synthétique anti-muscarinique, comme agent bronchodilatateur chez le cheval. Sept chevaux poussifs ont été mis en crise par une exposition naturelle. La réversibilité de la crise par administration intraveineuse de sulfate d'atropine a été préalablement testée. Des tests de fonction pulmonaire (mécanique ventilatoire et analyse de gaz sanguins artériels) ont été réalisés sur les chevaux avant, puis après l'administration par aérosol de 1500 µg de glycopyrrolate (Robinul®). La crise se traduisait par une augmentation de la résistance pulmonaire totale et du maximum de variation de pression pleurale et par une diminution de la compliance. Il ressort de ce protocole que le glycopyrrolate a des effets aussi rapides et aussi puissants que l'atropine par voie intraveineuse ou que l'ipratropium bromide à la dose de 1200 µg par aérosol de poudre sèche. En outre, le traitement est parfaitement toléré, et aucun effet secondaire n'est observé contrairement à ce qui est noté lors de l'administration intraveineuse de sulfate d'atropine. Cette molécule a donc incontestablement une place parmi les traitements dont dispose le praticien équin pour traiter une crise de pousse chez le cheval
