Cytogenetic characterization of abaca (Musa textilis Nee) and its hybrids with Musa balbisiana Colla

This study aimed to determine whether progenies from the cross between M. textilis Nee (2n=20) and M. balbisiana (2n=22) produced at IPB [Institute of Plant Breeding, UPLB, College, Laguna, Philippines] are true hybrids. The chromosome number and the length parents (abaca Abuab and Inosa and banana Pacol) and their backcross hybrids were determined by constructing karyograms. Sample collection time and cold shock treatment duration were optimized. Root tips (1-4 cm) were collected from 10:00 am to 10:30 am when the number of dividing cells was high. Treatment with cold shock for 2 hours led to larger chromosomes and more well-spread cells to facilitate the construction of karyograms. Abuab and the abaca hybrids (BC sub 1, BC sub 2 and BC sub 3 have a diploid chromosome number of 2n=20. Inosa has either 2n=20 or 2n=22, which can vary from 2n=17 to 2n=23. Pacol, the cultivar of M. balbisiana Colla, has a chromosome number of 2n=22. The results of the chromosomal count of backcross progenies imply that BC sub 1, BC sub 2, and BC sub 3 are true hybrids of abaca and banana. This work provides a basis for karyotype analysis to classify Musa cultivars.
