Application of methylene blue color test for the detection of inherited susceptibility to hemolysis of Korean native cattle

Blood samples were obtained from Korean native cattle and dairy cattle of Holstein species in the slaughter house and methylene blue color tests were performed for the detection of the inherited susceptibity to hemolysis. Glucose-6-phosphate dehydrogenase activities expressed as the optical density obtained by methylene blue color test were the highest as 0.54 in male Korean cattle, 0.62 in female Korean cattle and 0.72 in dairy cattle of Holstein species. Percent hemolysis, packed cell volume and plasma protein contents were measured and compared with relation to the results of methylene blue color test and no correlation was observed in each

In order to investigate the morphological characteristics of dendritic cells in the mammary gland, the appearance on the clear cells(CLs) or ATPase-positive dendritic cells(APDCs) have been observed by the light microscope. CLs were observed in the mammary tissues of the experimental animals, such as mice, rats, guinea pigs, rabbits, cats, dogs, pigs, cows and Korean native goats, and these CLs were confirmed as the ATPase-positive cells of typical dendritic appearance(APDCs). The APDCs were distributed in between the secretory epithelial cells, between the secretory epithelial cells and the myoepithelial cells, the basal area of the secretory epithelial cells, the interalveolar and interlobular connective tissues, and in between the epithelial cells of secretory duct. The APDCs were observed more frequently during the middle period of lactation than the other periods, and were irregularly or uniformly distributed according to the location. The mean difference between the inbred and the noninbred counts in the mice was statistically significant(P0.0001), and the similar result was presented in the rats(P0.0000)

This experiment was carried out to provide the data of blood chemistry in use of application to clinical diagnosis in racehorses. Blood was collected from 30 racehorses (female 17, male 13). Calcium, inorganic phosphorus, blood glucose, blood urea nitrogen, cholesterol, bilirubin, fibrinogen and ketone body in serum were analyzed. The values of calcium and inorganic phosphorus were maintained a certain level without much variation regardless of sex and species. Ca/P ratio was in the range of 3.13-3.35. The value of blood glucose was 60mg/100ml. Age and sex of racehorses make no difference in the value. The value of blood urea nitrogen and total cholesterol showed a tendency to increase according to the age. Total cholesterol value was somewhat higher in male than in female. The value of total bilirubin showed a tendency to increase according to the age. There was no difference between sexes in the value. The value of fibrinogen showed no apparent difference with age and sex. Ketone body was negative in all of the cases. There was no variation on the species of the racehorses

The origin of Jindo dogs has been controversial. It has been informed by words of mouth that they are the offsprings of either the dogs of Sung dynasty (960-1279 A.D.) in China or the Mongolian dogs. Some breeds of dogs in Japan now have almost same appearance as jindo dogs. The authors reviewed the history of Japan and found that Korean hunting dogs were sent to Japan in early 5th century, which was before Sung dynasty of China. Dr. Jae-Keun Ryu of National Institute of Environmental Research has done some work on serums of dogs of both Korea and Japan; the results of his serological and molecular biological studies indicated that Korean dogs were sent to Japan 1,300-2,500 years ago. Jindo dogs appear to be the descendants of the dogs of th Stone Age. They have adapted themselves to the unfavorable environment of Jindo island so that they can maintain pure line-age and wild nature until today. It was concluded that they are Korean native dogs, not the offsprings of the dogs of Sung dynasty or the Mongolian dogs

A total of 593 mastitic pathogens was isolated from the clinical mastitic milk of dairy cattle in Kyonggi area from March 1984 to February 1988. The mastitic pathogens were further studied bacteriologically and examined for susceptibility patterns to 10 antibiotics. 173 pathogens were isolated in the first year of studies, 205 pathogens in the second year, 122 pathogens in the third year and 93 pathogens in the last year. The pathogens isolated from clinical mastitis were more in summer than other seasons. Staphylococcus spp (50%) and Streptococcus spp (18%) were the main pathogens in the first year of studies but coliforms (15%) and other bacteria (40%) were the main pathogens in the last year of studies. Coliform mastitis occurred only in summer and autumn. Antibiotic susceptibility patterns of the same genuses from clinical mastitis infections in different herds and in different years varied greatly. Therefore, treatment should be selected on the basis of susceptibility test results

The prevalence of yeasts in mammary glands of dairy cows and teat cups of milking machines was studied in Chinju area. The rate of subclinical yeast infection in 330 quarters was 3.6 %. Of 12 isolates from the milk, 4 Candida pseudotropicalis, 3 C. tropicalis, 2 C. krusei, 2 C. albicans and 1 Rhodotorula spp were identified. The 91.7 % of the isolates belonged to the genus Candida and C. pseudotropicalis was the predominant species. From 20.5 % of 200 teat cups tested, 51 strains of yeasts were isolated. These were 13 C. pseudotropicalis, 9 C. guilliermondii, 7 C. tropicalis, 5 C. krusei, 5 C. parapsilosis, 3 C. albicans, 2 Torulopsis glabrata, 2 Geotrichum candidum and 5 unidentified yeasts. C. pseudotropicalis was most frequently encountered

Some hematological and biochemical indices were assayed in Korean native cattle naturally harbouring Fasciola hepatica infection and compared with uninfected controls. Affected animals revealed reduction in total erythrocyte count and hemoglobin content, and increase in total leukocyte count. Infected cattle had significantly lower levels of serum total protein, serum albumin and magnesium. To establish a laboratory animal model for study on development of diagnostic methods for infectious bovine rhinotracheitis virus(IBRV), experimental infection of the virus to rabbits immunosuppressed with dexamethasone(DX) were carried out. When lymphocyte activity was measured by lymphocyte transformation to phytohaemagglutinin parallel with total and differential leucocyte counting, both groups treated with 2.0mg DX once and 1.0mg DX daily showed marked immunosuppression between 5 to 72 hrs. after administration. The degree of suppression of lymphocyte activities was more remarkable in the latter group. IBRV PQ7 strain at 10** (7.5) TCID50/0.2ml was inoculated into conjunctival sacs of rabbits immunosuppressed with DX and non-treated. During 3 weeks observation, the immunosuppressed groups revealed mild conjunctivitis, viremia and virus recovery by 33.3 to 100%, whereas the DX nontreated group showed viremia and virus recovery with no clinical conjunctivitis in one of three rabbits(33.3%). In conclusion, it was indicated that experimental infection of IBRV PQ7 strain to rabbit was limited in prerequisite to immunologic modification by administration of immunosuppressive drugs

In the course of studying porcine enterovirus infection in piglets, the vascular endothelial cells in the CNS of colostrum-deprived piglets with polioence-phalomyelitis were investigated by electron microscope. The experimental piglets were orally infected with the porcine enterovirus serogroup 3 isolated in Korea and necropsied at 7 days postinoculation. Crystalline arrays of viral particles were found in the vascular endothelial cells of the spinal cord and cerebellum. Aggregates of immature viral particles were occasionally observed in the vascular endothelial cells in the meninges. The rough ER was deprived of ribosomes, irregularly dilated and associated with viral crystals. There were abundant cytoplasmic filaments, dilatation of perivascular space, perivascular cuffing, and the partial disruptions of endothelial cell membrane and basal lamina

In order to know the effect of dexamethasone on the induction of the rabbit viral hepatitis, the pathological changes were observed in the native rabbits, 2 to 6 months old in age, that were injected by dexamethasone and liver emulsion of Angora rabbit naturally infected with viral hepatitis. The native rabbits injected by dexamethasone and liver emulsion were infected with viral hepatitis and died between 2 and 7 days after inoculation. Clinical signs and gross lesions were very similar to those of Angora rabbit which had naturally occurred. In microscopical findings, the hepatic lesions were characterized by peripheral necrosis of the lobules, and peripheral necrosis of the lobules with fatty changes of hepatic cells occurred in a few cases. Perivascular lymphocytic infiltration in the central nervous system was observed in some cases. The lesions of the other organs were very similar to those of Angora rabbit. On the other hand, the native rabbits that were injected by only liver emulsion of Angora rabbit naturally infected with viral hepatitis were not infected with the disease except in very few cases
