Efficacy of Bdellovibrio bacteriovorus 109J for the treatment of dairy calves with experimentally induced infectious bovine keratoconjunctivitis Полный текст

OBJECTIVE To determine the efficacy of Bdellovibrio bacteriovorus 109J for the treatment of calves with experimentally induced infectious bovine keratoconjunctivitis (IBK). ANIMALS 12 healthy dairy calves. PROCEDURES For each calf, a grid keratotomy was performed on both eyes immediately before inoculation with Moraxella bovis hemolytic strain Epp63–300 (n = 11 calves) or nonhemolytic strain 12040577 (1 calf). For each calf inoculated with M bovis Epp63–300, the eyes were randomly assigned to receive an artificial tear solution with (treatment group) or without (control group) lyophilized B bacteriovorus 109J. Six doses of the assigned treatment (0.2 mL/eye, topically, q 48 h) were administered to each eye. On nontreatment days, eyes were assessed and corneal swab specimens and tear samples were collected for bacterial culture. Calves were euthanized 12 days after M bovis inoculation. The eyes were harvested for gross and histologic evaluation and bacterial culture. RESULTS The calf inoculated with M bovis 12040577 did not develop corneal ulcers. Of the 22 eyes inoculated with M bovis Epp63–300, 18 developed corneal ulcers consistent with IBK within 48 hours after inoculation; 4 of those eyes developed secondary corneal ulcers that were not consistent with IBK. Corneal ulcer size and severity and the time required for ulcer healing did not differ between the treatment and control groups. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that B bacteriovorus 109J was not effective for the treatment of IBK; however, the experimental model used produced lesions that did not completely mimic naturally occurring IBK.

OBJECTIVE To characterize polymorphisms of the gene for cytochrome P450 isozyme 2D50 (CYP2D50) and the disposition of 2 CYP2D50 probe drugs, dextromethorphan and debrisoquine, in horses. ANIMALS 23 healthy horses (22 Thoroughbreds and 1 Standardbred). PROCEDURES Single-nucleotide polymorphisms (SNPs) in CYP2D50 were identified. Disposition of dextromethorphan (2 mg/kg) and debrisoquine (0.2 mg/kg) were determined after oral (dextromethorphan) or nasogastric (debrisoquine) administration to the horses. Metabolic ratios of plasma dextromethorphan and total dextrorphan (dextrorphan plus dextrorphan-O-β-glucuronide) and 4-hydroxydebrisoquine concentrations were calculated on the basis of the area under the plasma concentration-versus-time curve extrapolated to infinity for the parent drug divided by that for the corresponding metabolite. Pharmacokinetic data were used to categorize horses into the phenotypic drug-metabolism categories poor, extensive, and ultrarapid. Disposition patterns were compared among categories, and relationships between SNPs and metabolism categories were explored. RESULTS Gene sequencing identified 51 SNPs, including 27 nonsynonymous SNPs. Debrisoquine was minimally detected after oral administration. Disposition of dextromethorphan varied markedly among horses. Metabolic ratios for dextromethorphan ranged from 0.03 to 0.46 (mean, 0.12). On the basis of these data, 1 horse was characterized as a poor metabolizer, 18 were characterized as extensive metabolizers, and 3 were characterized as ultrarapid metabolizers. CONCLUSIONS AND CLINICAL RELEVANCE Findings suggested that CYP2D50 is polymorphic and that the disposition of the probe drug varies markedly in horses. The polymorphisms may be related to rates of drug metabolism. Additional research involving more horses of various breeds is needed to fully explore the functional implication of polymorphisms in CYP2D50.

OBJECTIVE To evaluate safety and toxicokinetic profiles associated with daily oral administration of grapiprant, a new analgesic that selectively blocks the prostaglandin E2 EP4 receptor, to cats. ANIMALS 24 healthy domestic shorthair cats (12 males and 12 females). PROCEDURES Cats were randomly assigned (3 of each sex/group) to receive a placebo capsule or grapiprant at 3, 9, or 15 mg/kg, administered PO once daily for 28 days, beginning on day 0. Food consumption and behavior were observed daily, body weight was measured weekly, and clinicopathologic tests were performed on blood and urine samples collected on days −7, 14, and 25. Blood samples for toxicokinetic analyses were collected after treatment on days 0 and 27. Cats were euthanized on day 28, and full necropsies and histologic evaluations were performed. RESULTS Grapiprant rapidly reached peak serum concentrations and maintained substantial concentrations throughout the 28-day period. By day 27, maximum serum concentrations ranged from 683 ng/mL to 4,950 ng/mL, which were attained by 1 to 4 hours after administration. Serum half-lives on day 27 ranged from approximately 2 to 14 hours (median, approx 5 to 6 hours). Grapiprant was well tolerated, and no adverse effects were detected at doses ≤ 15 mg/kg. No significant effects of grapiprant were identified on body weight, food consumption, clinicopathologic variables, or gross or histologic necropsy findings. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested the safety of daily oral administration of grapiprant to cats. Additional studies are needed to evaluate the efficacy of grapiprant for treatment of cats with osteoarthritis.

OBJECTIVE To evaluate repeatability and reproducibility of right ventricular Tei index (RTX) values derived from dual pulsed-wave Doppler, conventional pulsed-wave Doppler, and tissue Doppler echocardiography and to investigate relationships and repeatability among the 3 methods in healthy dogs. ANIMALS 6 healthy adult Beagles. PROCEDURE Echocardiography was performed on each dog on different days for 2 weeks (3 times/d) by 2 echocardiographers. Intraobserver within- and between-day and interobserver coefficients of variation (CV(s)) and intraclass correlation coefficients (ICCs) for RTX(s) derived from dual pulse-waved Doppler (RTX(DPD)), conventional pulsed-wave Doppler (RTX(PD)), and tissue Doppler (RTX(TD)) methods were determined. Degrees of agreement among RTX values derived from the 3 methods were assessed by modified Bland-Altman analysis. RESULTS Least squares mean (95% confidence interval) RTX(td) was 0.50 (0.46 to 0.54), which was significantly higher than that for RTX(DPD) (0.27 [0.23 to 0.31]) and RTX(PD) (0.25 [0.21 to 0.29]). Agreement between RTX(DPD) and RTX(PD) was good (bias [mean difference], 0.04 [95% confidence interval, −0.03 to 0.10]). The RTX(dpd) had high within-day (CV, 6.1; ICC, 0.77) and interobserver (CV, 3.5; ICC, 0.83) repeatability, but between-day repeatability was not high. The RTXtd had high within-day repeatability (CV, 6.0; ICC, 0.80), but between-day and interobserver repeatability were not high. Within-day, between-day, and interobserver repeatability of RTXPD were not high. CONCLUSIONS AND CLINICAL RELEVANCE RTX(dpd) measurement was a repeatable and reproducible method of cardiac evaluation in healthy dogs. The RTX(TD) values were significantly higher than the RTX(DPD) and RTX(PD) values; therefore, RTX values derived from different echocardiographic methods should be interpreted with caution.

OBJECTIVE To evaluate the correlation coefficient of the ratio between diameter of the caudal vena cava (CVC) and diameter of the aorta (Ao) in dogs as determined ultrasonographically with systolic pressure variation (SPV). ANIMALS 14 client-owned dogs (9 females and 5 males; mean ± SD age, 73 ± 40 months; mean body weight, 22 ± 7 kg) that underwent anesthesia for repair of skin wounds. PROCEDURES Anesthesia was induced. Controlled mechanical ventilation with a peak inspiratory pressure of 8 cm H2O was immediately started, and SPV was measured. During a brief period of suspension of ventilation, CVC-to-Ao ratio was measured on a transverse right-lateral intercostal ultrasonographic image obtained at the level of the porta hepatis. When the SPV was ≥ 4 mm Hg, at least 1 bolus (3 to 4 mL/kg) of Hartmann solution was administered IV during a 1-minute period. Bolus administration was stopped and the CVC-to-Ao ratio measured when SPV was < 4 mm Hg. Correlation coefficient analysis was performed. RESULTS 28 measurements were obtained. The correlation coefficient was 0.86 (95% confidence interval, 0.72 to 0.93). Mean ± SD SPV and CVC-to-Ao ratio before bolus administration were 7 ± 2 mm Hg and 0.52 ± 0.16, respectively. Mean ± SD SPV and CVC-to-Ao ratio after bolus administration were 2 ± 0.6 mm Hg and 0.91 ± 0.13, respectively. CONCLUSIONS AND CLINICAL RELEVANCE In this study, the CVC-to-Ao ratio was a feasible, noninvasive ultrasonographically determined value that correlated well with SPV.

OBJECTIVE To determine the pharmacokinetics of detomidine hydrochloride administered IV (as an injectable formulation) or by the oral-transmucosal (OTM) route (as a gel) and assess sedative effects of the OTM treatment in healthy dogs. ANIMALS 12 healthy adult dogs.PROCEDURES In phase 1, detomidine was administered by IV (0.5 mg/m2) or OTM (1 mg/m2) routes to 6 dogs. After a 24-hour washout period, each dog received the alternate treatment. Blood samples were collected for quantification via liquid chromatography with mass spectrometry and pharmacokinetic analysis. In phase 2, 6 dogs received dexmedetomidine IV (0.125 mg/m2) or detomidine gel by OTM administration (0.5 mg/m2), and sedation was measured by a blinded observer using 2 standardized sedation scales while dogs underwent jugular catheter placement. After a l-week washout period, each dog received the alternate treatment. RESULTS Median maximum concentration, time to maximum concentration, and bioavailability for detomidine gel following OTM administration were 7.03 ng/mL, 1.00 hour, and 34.52%, respectively; harmonic mean elimination half-life was 0.63 hours. All dogs were sedated and became laterally recumbent with phase 1 treatments. In phase 2, median global sedation score following OTM administration of detomidine gel was significantly lower (indicating a lesser degree of sedation) than that following IV dexmedetomidine treatment; however, total sedation score during jugular vein catheterization did not differ between treatments. The gel was subjectively easy to administer, and systemic absorption was sufficient for sedation. CONCLUSIONS AND CLINICAL RELEVANCE Detomidine gel administered by the OTM route provided sedation suitable for a short, minimally invasive procedure in healthy dogs.

The objective of this study was to evaluate the impacts of discharged waste effluent into water sources on heavy metals content in water, sediment and fish. A pilot study was carried out during the period from (March to May 2014). Samples were collected from water, sediment and fish (Oreochromis niloticus) from five fish farms and River Nile in Beni-Suef governorate to estimate Lead, Cadmium, Zinc and Copper. The results revealed a significant increase in Pb and Cd in water, sediment and fish samples collected from fish farms and these metals were within the permissible limits in River Nile samples. Zn and Cu in all samples of water and muscle of (farms and River Nile) were lower than permissible limits of WHO. The results of this study indicate that high levels of some heavy metals present in fish collected from the fish farms in Beni-Suef governorate are not accepted as food for human purposes.

The study was conducted to investigate the effect of low metabolizable energy diets with normal or narrow metabolizable energy to crude protein ratios (ME:CP) on performance, carcass characteristics, body composition and blood parameters in broilers fed from 1 to 42 days of age. The chicks were divided into 7 groups. The birds were fed starter & grower diets. Seven experimental diets were formulated in each phase; one control and 6 tested diets. The control diet was formulated according to the NRC of poultry (1994) and the other six diets fed three different levels of low energy diets (2900, 2700 & 2500 kcal/kg; one level for each 2 groups). The first three tested groups named ''normal calorie-protein ratio” groups in which the CP decreased in proportion to the decrease in ME, keeping the normal NRC ratio. In the second three tested groups, termed “narrow calorie-protein ratio” groups, the dietary protein was kept at the NRC levels leading to ratios narrower than that of the NRC. Results showed that chicks fed low ME diets with normal energy to protein ratio had lower body weight and feed utilization efficiency than the chicks fed the control diets. While, birds fed the low ME diets with normal protein NRC-levels and narrow ratios had nearly equal weight and feed conversion to the control. The body composition and carcass characteristics were not affected by the dietary treatments. Moreover, the blood parameters had no significant variations among the groups, except for total protein, ALT and AST which had an increased response to decreased dietary energy density. In conclusion, decreasing the dietary ME level without decreasing the crude protein level was more efficient economically and had no any adverse effect on the performance. However, decreasing of dietary ME with normal ME:CP ratio resulted in decreased performance and low economic efficiency.

Preventing the transmission of disease into and between poultry farms, hatcheries, and from wildlife, or their droppings, is essential to maintain healthy productive poultry flocks. This study was designed to assess the current situation of biosecurity measures in 3 types of poultry operations; 125 small-scale commercial broiler farms (sector III), hobby poultry premises (backyards; sector IV) and hatcheries selected from Giza, Fayoum and Beni-Suef province, Egypt. A structured questionnaire survey, interviews and observation were used to collect data on farm characterization, bio-security practices and disease preventive measures in sectors and hatcheries. The respondents were farm owners, farm managers, veterinarians, and workers. It has been found that the distance between small commercial broiler farms were 48.4% [2- 5 km] while for hobby poultry keepers 66.5% [≤100 m], and hatcheries 41.6% [1-3 km] and 30.6% [7-11 km]. In small-scale broiler farms practiced all in/ all out system (89 %) as compared to hatcheries and backyards (83.3 and 46.1 %, respectively). In the most of the small commercial broiler farms, the resting period between production cycles was higher on 4-7 days (32.5%). Methods most used for disposal of daily mortalities were on landfill (45.2%) among small commercial broiler farms. Meanwhile, hobby poultry premises and hatcheries included disposal in water canals (38.5 and 33.3%, respectively). On the other hand, Hobby poultry keepers reported that veterinarians rarely inspected their premises. Mortality rate/cycle was (11.9%) in almost broiler farms. About 64.3 % of the small commercial producers practiced all the recommended vaccination of their birds against highly pathogenic diseases. In conclusion, the majority of the small-scale broiler farms and all most backyards were far from the implementation of biosecurity measures. Biosecurity situation needs a combined effort from stakeholders, poultry keeper of hobby birds (backyard) to improve biosecurity level for those sectors.

Bovine tuberculosis, caused by Mycobacterium bovis, is a zoonotic disease causing approximately 6% of total human deaths. Its economic losses are not only a reduction of 10-20% in milk production and weight, but also infertility and condemnation of meat. Many serological tests are applied for detection of tuberculosis. ELISA test has the highest sensitivity and specificity than the other serological tests for the diagnosis of tuberculosis. Several forms of new technology were brought into the diagnostic approach to mycobacterial infection. The aim of this work was to detect bovine tuberculosis by application of different traditional tests and PCR. Tuberculin skin test was applied on 2650 cattle, only 63(2.4%) were positive. Forty eight (76.2%) of the slaughtered positive animals showed visible lesions (VL) while the other 15 (23.8%) had non-visible lesions (NVL). Bacteriological examination of 10 selected tuberculin positive samples revealed M. bovis from 6 processed samples (60%) while PCR and ELISA assays revealed positive in 8 cases (80%) and 7 cases (70%), respectively. It was concluded that PCR test is more sensitive and specific test to confirm the infection with tuberculosis.
