Poultry waste management practices in Bangladesh: Farmer's perceptions, and food and environmental hazards

Objective: The poultry industry plays a key role in developing socio-economic and health sectors in Bangladesh. Poultry waste is a potential environmental threat as untreated poultry waste is used in vegetable gardens. The study aimed to investigate the current situation of small-scale poultry farms and their waste management practices in selected areas of Bangladesh and detect Escherichia coli and Salmonella in vegetables from farms using untreated poultry waste as fertilizer. Materials and Methods: A structured questionnaire-based survey was conducted in 86 small-scale poultry farms from different upazilas of Mymensingh and Khulna districts. 104 samples, including vegetables, poultry litter, water, and soil, were collected from vegetable gardens, ponds, fields, and wet markets in Mymensingh district to detect microbial contamination. Bacteria were identified based on their growth and colony morphology on selective media and motility tests. The presence of E. coli and Salmonella was confirmed by polymerase chain reaction (PCR) using a commercial PCR kit. Results: The survey revealed that mostly middle-aged males were involved in poultry farming. Most of the farmers had primary education and engaged in farming for about 5 years without training. In the study area, 37% of farmers collected droppings daily in the morning and used them as organic fertilizer. About 58% of farmers did not know the hygienic handlings of droppings and faced health problems. In PCR, either E. coli or Salmonella or both were confirmed in vegeta¬bles, litter, soil, and pond water. Objective: This study aimed to isolate Aliarcobacter cryaerophilus in ready-to-cook poultry meat in Bangladesh. Materials and Methods: Thirty drumstick samples were collected from super shops in Dhaka city (n = 10), Mymensingh city (n = 10), and Patuakhali town (n = 10). After sample processing, they were cultured in Blood agar media with Campylobacter base using a microfilter (0.42 nm). Suspected colonies were subjected to DNA extraction and PCR assay targeting 16SrRNA genes. Then, sequencing was performed for confirmation. Results: Of 30 samples, 3 (10%) were positive for A. cryaerophilus. Phylogenetic analysis shows that our isolate has strong similarities with one of the isolates from China. Conclusion: The presence of this organism in ready-to-cook poultry meat is a significant concern for consumers as it bears zoonotic importance. [J Adv Vet Anim Res 2023; 10(1.000): 113-117]

Objective: Camel mastitis is indeed a serious problem that can have significant impacts on animal health and production as well as pose a potential public health hazard. This work aimed to identify the bacterial species responsible for camel mastitis and evaluate the associated immunological and clinicopathological alterations in infected camels. Materials and Methods: Raw milk and blood samples were collected from 40 apparently healthy she-camels, and 40 she-camels suffered from clinical mastitis (CMG). Milk samples were sub¬jected to bacteriological examination. Serum immunological, biochemical, and hematological parameters were estimated and statistically analyzed. Results: Similar bacterial species were obtained from the two groups with different isolation rates. Staphylococcus epidermidis and Escherichia coli were the dominant species in the apparently healthy group, while Pseudomonas aeruginosa and Bacillus cereus were the dominant species in CMG. A significant (p < 0.05) elevation of the pro-inflammatory cytokines, acute phase proteins (APPs), free radicals, total protein, Glob, kidney and liver function tests, and triglyceride concentrations were detected in CMG, and a significant (p < 0.05) decrease in the anti-inflammatory cytokine, antiox¬idants, Alb, glucose, and T/LDL/HDL-cholesterol concentrations was observed in CMG. Microcytic hypochromic anemia with hypoferremia, hypotransferrinemia, hyperferritinemia, and neutrophilic leukocytosis was depicted in CMG. The estimated pro-inflammatory cytokines, APPs, and total anti¬oxidant capacity (TAC) yielded high sensitivity and specificity, but the highest likelihood ratio was for TAC, fibrinogen (Fb), and ferritin, and the highest percentages of increase were for IL-1α and IL-1β. Conclusion: The study emphasizes the importance of hygienic preventive measures to control camel mastitis and the importance of supportive treatment to reverse the hemato-biochemical and iron profile changes that result from the immune response in mastitic she-camels. TAC, Fb, ferritin, IL-1α, and IL-1β are good biomarkers for camel mastitis. [J Adv Vet Anim Res 2023; 10(2.000): 284-291]

Objective: This research investigated secoisolariciresinol diglucoside (SDG) flax extract effects on apoptosis, hedgehog (Hh), autophagy, and the anti-oxidation process in experimentally induced obesity. Materials and Methods: Forty rats were separated into two sets regarding either receiving a nor¬mal balanced diet or a high-fat diet (HFD) and then distributed into four groups: GI: The control group had a regular diet for 12 weeks. GII: animals received a high-fat meal and saline by gastric gavage. GIII: HFD obese rats treated with SDG extract orally (10 mg/kg/b.w.) and 1.18 mg SDG/kg in the diet for 4 weeks GIV: Normal balanced diet rats received SDG extract orally (10 mg/kg/b.w.) and 1.18 mg SDG/kg of chow for 12 weeks in addition to their regular balanced diet. Results: The administration of SDG extract exhibited a significant drop in body weight, glucose, lipid profile, and leptin compared to the obese group. It also improved the antioxidant levels (lowering the levels of malondialdehyde while increasing the total antioxidant capacity) and anti-inflammatory status (decreasing interleukin-6 and tumor necrosis factor-alpha). SDG extract downregulates the expression of HH genes (protein patched homolog 1, Hh-interacting protein, glioma-associated oncogene homolog 1, and smoothened receptor) in conjunction with the mod¬ulation of autophagy genes and apoptotic proteins. Conclusion: SDG extract showed improved anti-inflammatory and antioxidant status and down¬regulated the expression of HH genes while modulating autophagy genes and apoptotic proteins among obese rats, suggesting that it may be used to avert and manage obesity and its correlated complications by modulating oxidation, inflammation, autophagy, and apoptosis. Advanced future research on the SDG autophagy pathway to address obesity and its complications is mandatory. [J Adv Vet Anim Res 2023; 10(2.000): 321-335]

Phytoestrogens are non-steroid polyphenolic materials present in 300 plants. Regarding their structural similarities to estradiol, phytoestrogens attach to estrogen receptors and display anti-or pro-estrogenic activities. This review explored phytoestrogens' potential advantages and autophagy properties in light of their future application for disease management, highlighting how phytoestrogens could modulate autophagy. Research has examined the prospective benefits of phytoestrogens for the anticipation and management of various conditions, including signs of menopause, tumors, skin deterioration, osteoporosis, heart disease, neurodegenerative conditions, disorders of the immune system, and metabolic syndrome, owing to their therapeutic effects. As phytoestrogens can activate or inhibit autophagy, which has antioxidant, apoptotic, anti-mutagenic, anticancer, transcriptional, and genomic impacts on cancer and aging illnesses, phytoestrogens could influence diseases through the modulation of autophagy. The collaborative research on animal models, utilization of genetic techniques, and administration of pharmacolog¬ically active substances has indicated the possible therapeutic benefits of autophagy modulation in various illnesses. Further research is required to illustrate the pathways by which phytoestro¬gens modulate autophagy and the possible therapeutic effects on these diseases. Objective: This study aimed to explore the incidence and antimicrobial resistance (AMR) of Escherichia coli, Staphylococcus aureus, and Bacillus cereus in raw milk and some Egyptian dairy products, namely Kariesh cheese and rice with milk. Material and Methods: 112 samples (70 raw milk, 30 Kariesh cheese, and 12 rice with milk) were randomly collected from different districts in Cairo and Giza, Egypt. The samples were examined for E. coli, S. aureus, and B. cereus presence. The susceptibility of the obtained isolates was tested against 11 antimicrobials using the disk diffusion method, and further, the presence of AMR genes was examined. Results: The incidences of E. coli, S. aureus, and B. cereus were 69.64%, 12.5%, and 16.7% in the examined samples, respectively. The antibiogram indicated that E. coli isolates (n = 60) were resistant to gentamycin (73.33%), ampicillin (AM, 53.3%), and cefotaxime (CTX, 16.66%). Multidrug-resistant (MDR) E. coli strains (n = 5) were tested for β-lactams resistance genes. blaTEM was detected in all isolates, and two of them additionally carried blaCTX-M. Staphylococcus aureus isolates (n = 10) were resistant to AM (100%), followed by tetracycline (TE), CTX, and gentamycin (60% each). All MDR S. aureus strains (n = 4) carried blaZ and tetK, and three of them additionally carried aac(6')-aph (2''). Bacillus cereus isolates (n = 30) showed resistance to AM (100%), amoxi¬cillin (20%), and TE (6.66%). bla and tetA genes were detected in all MDR B. cereus isolates (n = 6). Conclusion: Our findings denote the high incidence of potential health hazards in raw milk and some of its products and the existence of AMR bacteria, including MDR strains, which can cause human illnesses that are difficult to treat. Objective: This study aimed to examine the diversity of phenotypic characteristics of female Alabio ducks (Anas platyrhynchos Borneo) and their eggs in South Kalimantan, Indonesia. Materials and Methods: A total of 200 18-month-old ducks and 300 eggs were selected using a survey method with multistage random sampling. These samples were obtained from two dis¬tricts in the province of South Kalimantan [Banjar (BJ) districts and Tanah Laut (TL)]. The observed data were analyzed descriptively with variations using the independent t-test for each location. Principal component analysis (PCA) was deployed to assess the overall variance and define vari¬ables with greater discriminatory power between individuals. Results: The observations on the body's physical characteristics and the eggs of Alabio ducks showed significant differences between ducks from BJ and TL areas (p < 0.05) except for yolk diam¬eter, albumen height, albumen index, and Haugh unit (HU) (p > 0.05). Most egg quality traits from BJ were greater than those from TL, which include egg weight, egg length, egg width, yolk weight, albumen weight, shell weight, shell thickness, and egg shape index. The PCA revealed medium-to-high communalities in the phenotypic body characteristics of Alabio ducks and their eggs. Conclusion: Significant physical characteristics and egg quality differences were found between the two locations, except for the albumen index and HU, with substantial variability within each trait. These traits could explain the total variation in the phenotypic characteristics of female Alabio ducks. [J Adv Vet Anim Res 2023; 10(2.000): 249-256]

Objective: This study was conducted to establish a rat model of acute exacerbation of idiopathic pul¬monary fibrosis (AE-IPF) using the combination of bleomycin (BLM) and lipopolysaccharides (LPS). Materials and Method: Twenty-four male Sprague Dawley rats were allocated into two equal groups: the sham or the bleomycin and lipopolysaccharides-induced AE-IPF group (BLM-LPS). On Day 7, BLM intratracheally and LPS intraperitoneally were both used to administer AE-IPF. The BLM-LPS group and its respective sham group were terminated on Days 8, 14, or 21. Samples of bronchoalveolar lavage fluid (BALF) and lungs were taken and investigated for cell count and histopathology. Results: On Day 8, histological analysis revealed inflammatory cell infiltration with edema and hyaline membrane, and the BALF differential cell count revealed high neutrophil counts. By having a higher collagen density area and Ashcroft modified score than the sham group on Day 14, the BLM-LPS group displayed significantly lower oxygen saturation, alveolar air area, and a fibrotic appearance. However, there was a spontaneous resolution in inflammation and fibrotic appearance on Day 21 after the BLM administration. Conclusions: By combining BLM and LPS, it was possible to create a successful rat model of AE-IPF. The present model showed the peak exacerbation on Day 8 and the fibrotic peak on Day 14, which gradually improved. Objective: The study aimed to investigate the hepatoprotective effects of Gastrodia elata rhi¬zome (GR) on thioacetamide (TAA)-induced liver injury in dogs. We evaluated serum biochemical and hematological parameters, with emphasis on alanine transaminase (ALT), alanine phosphates (ALP), and nitric oxide (NO) levels, in dogs with TAA-induced liver injury. Materials and Methods: The animals were divided into a control group (Con), TAA group, Silymarin group (Sil, 50 mg/kg), Gastrodia rhizome low dose (GRL) (low) + TAA, GRH (high) + TAA, and GR high-dose group (GRH) control group. GRL and GRH were given daily at 50 and 100 mg/kg, respectively. TAA was given on days 1, 4, and 7 at a dose of 300 mg/kg. Results: GR significantly reduced liver injury in treated animals, as indicated by lowered levels of ALT (about 32% at day 21 in both GRL + TAA and GRH + TAA groups), ALP (about 17% and 21% at day 21 in both GRL + TAA, GRH + TAA groups, respectively), and NO (about 36% at day 21 in both GRL + TAA, GRH + TAA groups) compared to the TAA control group. Hematological parameters showed mild changes during the experiment. High-performance liquid chromatography analysis revealed gastrodin, a major component of the GR extract, constitutes 2.6% of the extract. Conclusion: The GR demonstrated significant hepatoprotective effects against TAA-induced liver injury in dogs. Objective: This study analyzes the mycobiome in wild and captive Sumatran orangutans. Materials and Methods: Nine orangutan feces samples from the wild and nine from captivity were divided into three repeats from 11- to 15-year-olds in good health. The Illumina platform for analysis of ITS bioinformatics was used according to the Qiime2 and CCMetagen approaches. Results: Wild Sumatran orangutans include 53% Ascomycota, 38% uncultured fungi, and 4% Basidiomycota. Orangutans in captivity are 57% Ascomycota, 26% uncultured fungi, and 2% Basidiomycota. Based on genus level, uncultured Neurospora (31%), Penicillium (10%), Aspergillus (3%), Fusarium (3%), Candida (2%), Cutaneotrichosporon (2%), and Limonomyces (2%) are found in wild orangutans. The most prevalent genus among captivity orangutans is Aspergillus (32%), followed by fungal sp. (11%), Lasiodiplodia (18%), Devriesia (2%), and Sordariomycetes (2%). According to the Chao1 diversity index and Shannon and Simpson, there was no significant difference between wild and captive Sumatran orangutans. Conclusion: Neurospora is unique to wild Sumatran orangutans, although Aspergillus predom¬inates in captive orangutans. We hypothesize that the gut mycobiome of wild orangutans will resemble that of orangutans in captivity. The excellent range of food sources in the forest does not result in the prevalence of fungi in the typical gut microbiome. [J Adv Vet Anim Res 2023; 10(2.000): 228-236]
