Сканирующая электронная микроскопия как способ оценки иммуногенности и реактогенности противовирусных вакцин

In the conditions of the Republic of Belarus by means of application of a scanning electron microscopy there was realized a morphological assessment of the efficiency of mono- and associated antiviral vaccines. It is shown, that analyzed biological preparations invoke in poultry a number of structural disorders among which it is possible to mention both immunomorphological changes, and pathomorphological processes. The highest reactogenicity was showed by of monovaccines against infectious bronchitis (produced by Research Institute of Animal Protection, Russia); infectious laryngotracheitis (Institute of Experimental Veterinary of S.N.Vyshelesskij, Belarus); and also an associated vaccine against infectious bronchitis, infectious bursal disease and Newcastle disease (Research Institute of Animal Protection, Russia). These vaccines caused essential structural disorders in tissues at a place of administration, liver and kidneys. Administration of the vaccine against infectious bursal disease and associated vaccines against infectious bronchitis, infectious bursal disease, infectious laryngotracheitis and Newcastle disease (Institute of Experimental Veterinary of S.N.Vyshelesskij) strengthened the processes of lymphatization in fabricic bursa. All studied biological products caused activization of morphological reorganization of immune apparatus of a spleen and did not render essential influence on thymus structure. | С использованием сканирующей электронной микроскопии проведена морфологическая оценка эффективности моно- и ассоциированных противовирусных вакцин. Показано, что указанные биопрепараты вызывают в организме птиц ряд структурных нарушений, среди которых можно выделить как иммуноморфологические изменения, так и патоморфологические процессы. Наибольшую реактогенность проявляли моновакцины против инфекционного бронхита кур (ФГУ ВНИИЗЖ, Россия), инфекционного ларинготрахеита (ИЭВ им. С.Н. Вышелесского), а также ассоциированная вакцина против ИБК, инфекционной бурсальной болезни и болезни Ньюкасла (ФГУ ВНИИЗЖ, Россия), которые вызывали существенные структурные нарушения в ткани на месте введения, печени и почках. Использование вакцины против ИББ и ассоциированной вакцины против ИБК, ИББ, ИЛТ и БН (ИЭВ им. С.Н. Вышелесского) усиливало процессы лимфатизации в фабрициевой бурсе. Все изучаемые биопрепараты вызывали активизацию морфологической перестройки иммунного аппарата селезенки и не оказывали существенного влияния на структуру тимуса.

The research was realized by an example on 30 chickens aged 1, 10, 20, 30 60-days old, and 10 hens: 4-months, one-year-old and 2-year-old in the conditions of the Republic of Belarus. All poultry stock was maintained in the conditions of industrial production at the Gorodokskaya Poultry Farm of Vitebsk region (Belarus). As a result of the study there was revealed a certain pattern of variation of a quantity of active forms of enzymes in organs chickens of different age. Application of histochemical methods made it possible to reveal different levels of metabolic processes in cellular elements of thyroid gland, which demonstrated the habit of formation and functioning of the organ at different stages of postnatal ontogenesis in hens. Coefficients of acid phosphatase activity in thyrocytes of thyroid gland of hens and indexes of alkaline phosphatase activity in secretion forming cells were analyzed. | Исследование провели на 30 цыплятах в возрасте 1-, 10-, 20-, 30- 60-дней, и 10 курах: 4-месячные молодки, годовалые и 2-летние особи. Все поголовье птицы содержалось в условиях промышленного производства на Городокской птицефабрике Витебской области. В результаты исследований установлена определенная закономерность изменения количества активных форм ферментов в органах птиц разного возраста. Использование гистохимических методов позволило выявить разные уровни метаболических процессов в клеточных элементах щитовидной железы, которые демонстрируют характер становления и функционирования органа на разных этапах постнатального онтогенеза кур. В табличной форме представлены коэффициенты активности кислой фосфатазы в тироцитах щитовидной железы кур и показатели активности щелочной фосфатазы в секретообразующих клетках.

In the conditions of the Republic of Belarus there was realized a morphological analysis of pectoral and leg muscles of broiler chickens of cross Cobb-500 in the process of administration of metabolic activator Catosal. Research results showed the positive influence of catosal on the myogenesis: diameter of muscle fiber, the amount of bigger diameter muscles fibers in the first order strand and homogeneity of muscle strands. Under the influence of Catosal preparation there was stated the increasing of the amount of muscle tissue in pectoral and leg parts of chicken carcass. | Приведены данные морфологических исследований грудных и ножных мышц цыплят-бройлеров кросса Кобб-500 при применении активатора метаболизма катозал. Установлено, что катозал активизирует миогенез за счет увеличения диаметра мышечных волокон, содержания в пучках первого порядка мышечных волокон большей толщины, а также большей однородности мышечных пусков. Под действием препарата происходит увеличение содержания мышечной ткани как в грудной части тушки, так и в окороках.

On the basis of realized in the conditions of the Republic of Belarus research it was established, that Cryptosporidial infection was found at adult and young pigs. The data on influence studying cryptosporidiosis invasion on pathomorphological changes in internal organs of piglets in the conditions of an associative course with infectious. Associative diseases give clearer clinical findings and proceed much harder, with heavier morphological changes. The majority of diseases in the conditions of an associative course proceeds harder in comparison with a mono-infection or invasion. In the conditions of mono-invasion the basic changes are localized in small intestines, causing acute and subacute catarrhal enteritis and a serous inflammation of regional mesenteric lymphonodus. These histological processes are presented as non-uniform locus of infection of a mucous membrane of intestines with atrophy of villus, hypertrophy of crypth and mononuclear polymorphic-nuclear infiltration of basal membranes. In the conditions of association of Cryptosporidial infection with infectious diseases it was noted that the pathomorphological changes progress and are showed not only by catarrhal, but also by hemorrhagic and local necrotic enteritis. Epithelium of mucous membrane is exposed to desquamation, intestines walls become thinner because of meteorism. Thick intestines are also involved into inflammatory process. Associations of Cryptosporidial infection with infectious diseases lead to development of severe pathomorphological changes in intestines. Alteration has prevailing character that is expressed in progressive development of atrophic, dystrophic processes and decline of cells and tissues that conducts to increase of permeability of vessels and proliferation of cells of reticuloendothelial system. | На основании проведенных исследований установлено, что криптоспоридиозная инвазия выявляется у поросят. Представлены данные по изучению влияния инвазии криптоспоридиоза на патоморфологические изменения во внутренних органах поросят при ассоциативном течении с инфекционными болезнями. Ассоциативные заболевания дают более яркую клиническую картину и протекают гораздо тяжелее, с более выраженными морфологическими изменениями в организме. Большинство болезней при ассоциативном течении протекает более тяжело в сравнении с моноинфекцией или инвазией. При моноинвазии основные изменения локализуются в тонком кишечнике, вызывая острый и подострый катаральный энтериты и серозное воспаление регионарных брыжеечных лимфоузлов. Гистологические данные процессы представлены, как неоднородные участки инфицирования слизистой оболочки кишечника с атрофией ворсинок, гипертрофией крипт и мононуклеарной полиморфнооядерной инфильтрацией базальной мембраны. При ассоциации криптоспоридиоза с инфекционными заболеваниями патоморфологические изменения прогрессируют и проявляются не только катаральным, но геморрагическим и очаговым некротическим энтеритами. Эпителий слизистой оболочки подвергается десквамации, стенки кишечника истончаются вследствие метеоризма. В воспалительный процесс также вовлекается толстый кишечник. Ассоциации криптоспоридиоза с инфекционными заболеваниями приводят к развитию в кишечнике тяжелых патоморфологических изменений. Альтерация имеет преобладающих характер, что выражается в прогрессивном развитии атрофических, дистрофических процессов и некроза клеток и тканей, что ведет к повышению проницаемости сосудов и пролиферации клеток ретикулоэндотелиальной системы.

In the conditions of the Republic of Belarus there was studied sterility, harmlessness and immunoactivity of an obtained production vaccine against swine pasteurellosis by an example of laboratory animals – white mice and rabbits. Research results showed that the obtained vaccine was sterile and harmless for the laboratory animals, and had a high immunogenic activity. The analyzed vaccine prevented disease incidence of white mice after its double application in dose of 0,1 ml per animal and after a single immunization of mice in dose of 0,2 ml per animal. The inactivated vaccine caused the formation of immunity of rabbits after double immunization in dose of 1 ml per animal with 14 days interval and provided 100% vitality of animals.

In the conditions of the Republic of Belarus there was studied the formation of nonspecific protection factors of the first year foals. By a principle of conditional analogues there was generated a group of clinically healthy foals at which monthly from bulbar veins there were taken tests of blood for hematological and biochemical analysis. From the obtained data it was visible, that within the first year of foal life there were taking place the processes of formation of nonspecific protection factors of an organism which were accompanied by a gradual increasing of quantity of erythrocytes, haemoglobin, leukocytes, certain change in the diagramme, and also increasing of bactericidal and lysozyme activity of blood serum. However, at seven-month age there was marked a considerable decrease in number of erythrocytes, leukocytes, concentration of haemoglobin, bactericidal and lysozyme activity of blood serum is marked, in leukogram share reduction of segmented neutrophils, and at the same time, the content of eosinocytes and lymphocytes increased. It testified the decreasing of protective forces of an organism during this period. Consequently, the seventh month in the postnatal period of foal life proved to be the most critical.

Despite of presence of a wide choice of antihelminthics, in the Republic of Belarus it is still a real problem of development of preparations that provide long-term treatment and preventive maintenance efficiency against helminthosis. At the same time, helminthes and anthelminthics render the immunosuppression action. The solution of this problem can be a combined application of antihelminthics and adjuvants. There were proposed preparations Albepolis and Averpolis. Their application made it possible not only to release an organism of animals from parasites, but also to correct the state of immune system. The obtained research data confirm the available data about the immunosuppression action of parasites on an organism of animals. There was also is noted the immunosuppression action of Albendazol and Aversektin C in therapeutic doses. Albendazol possess a wide spectrum of action against trematodes, nematodes, cestodes. It interrupts the metabolism, oppresses fumarate reductase activity and adenosine triphosphate synthesis of a parasite. It proved to be low-toxic for homoiothermal animals. Albendazol rendered positove influence on growth, development and increase of cattle productivity.

The spectrum of acaricides changes continuously and improves. At the same time, big attention is given to research of new, more effective preparations meeting modern requirements. Research on studying the efficiency of Akaribil preparation against swine sarcoptosis (Sarcoptidae) was realized in the conditions of swine breeding farm in the Republic of Belarus. The preparation possesses high insect-acaricidal activity. In the conditions of the preparation storage and repeated opening of a ware in the course of tests there were stated no changes in drug smell or color. The scheme of the preparation application is applied in industrial production technology. It possesses anti-inflammatory properties, antiseptic stimulating healing of skin damages, does not irritate integuments, renders weak irritating effect on mucous membranes of eyes. In recommended doses Akaribil does not cause any adverse effects and complicating disease. There were stated no contraindications to preparation application. High therapeutic efficiency against arachnoentomosis was proved. Application the preparation made it possible not only to reduce disease incidence and animal death, but also to facilitate the severity of scabietic diseases. As a result of realized research it was established that efficiency of Akaribil against swine sarcoptosis was 100%.

In the conditions of the Republic of Belarus there was studied the preventive efficiency of various forms of a complex additive Felutsen against polymicroelementoses of calf-bearing dry cows and its influence on some of biochemical blood indexes. Research results proved high prophylactic efficacy of various forms of the additive Felutsen against polyhypomicroelementoses of calf-bearing dry cows. Restoration of a microelement homeostasis of cows made it possible to normalize the functional activity of liver, to prevent development renal insufficiency and dystrophic changes in bone tissue. The most expressed metabolic changes in blood of calf-bearing dry cows were stated after application of the carbohydrate-mineral additive Felutsen-briquette universal. There was given the analysis of structure of studied forms of complex additive Felutsen; indexes of trace element metabolism in cows; indexes of protein metabolism and level of nonprotein nitrogen; indexes of blood characterizing the functional condition of liver and carbohydrate metabolism of cows; indexes of trace element metabolism in cows after the termination of application of the additive.

In the conditions of the Republic of Belarus there were analyzed high productive traits of meaty calves and cross breeds bred in accordance with a system cow-calf, as well as the possibilities of using veal meat obtained in ecologically safe areas for infant food production. At the age of 6,5-7 months the weight of fresh carcass of bull-calves of sharolez breed in comparison with aberdin-angus х black-motley herdmates was on 41,6 kg (Р less than 0,001) higher, carcass yield and slaughter yield – on 13,8% (Р less than 0,001) and 13,6% (Р less than 0,001), respectively. The fat content in sharolez meat was 4,9% against 8,9%; protein - 21,9% against 18,8%. The black-motley suckling bull-calves surpassed their herdmates which were grown up by technology of dairy cattle breeding at the age of 6-6,5 months according to indexes of weight of fresh carcass - on 16,3 kg or 21,7% (Р less than 0,01); according to indexes in a slaughter yield - on 16,6% (Р less than 0,001); and according to indexes meat flesh in a carcass - on 1,5%. According to quality indexes, amino acid structure, acceptable concentration of toxic elements content, pesticides, antibiotics, radionuclides and other harmful substances the veal meat of calves which were bred on a system a cow-calve in ecologically pure zones proved to be important meat raw materials to produce infant food products.
