Effect of orally administered melatonin on intraocular pressure of ophthalmologically normal dogs Полный текст

OBJECTIVE To determine the effect of orally administered melatonin on the intraocular pressure (IOP) of ophthalmologically normal dogs. ANIMALS 20 ophthalmologically normal dogs (40 eyes). PROCEDURES In a randomized crossover study, each dog received a 7-day regimen of melatonin (0.1 to 0.2 mg/kg, PO, q 12 h) and a placebo (150 mg of lactose powder in a capsule, PO, q 12 h), with a 7-day washout period between treatment regimens. Rebound tonometry was used to measure the IOP in both eyes of each dog 5 times at 2-hour intervals on days 0 (before administration of the first dose), 2, 4, and 7 (after administration of the last dose) of each treatment period. Repeated-measures ANOVA was used to evaluate the effects of treatment, day, and IOP measurement time within day on IOP. RESULTS Intraocular pressure was not significantly associated with treatment but was associated with day and the interaction between day and IOP measurement time within day. The mean ± SD IOP was 14.26 ± 2.95 and 14.34 ± 2.69 mm Hg for the melatonin and placebo regimens, respectively. Within each treatment period, the mean IOP tended to decrease from day 0 to 7 as well as within each day, which was attributed to the dogs becoming acclimated to the study protocol and natural diurnal variations in IOP. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that oral administration of melatonin (0.1 to 0.2 mg/kg, q 12 h for 7 d) did not significantly affect the IOP of ophthalmologically normal dogs.

OBJECTIVE To compare use of a 3-level self-locking suture (3LSLS) technique with use of a previously described modified 3-loop pulley (M3LP) technique to repair rupture of the proximal aspect of patellar tendons in limbs from canine cadavers. SAMPLE Paired hind limbs of 6 adult mixed-breed dogs. PROCEDURES A limb from each pair was randomly assigned to be repaired by the 3LSLS technique or M3LP technique with size-2 ultrahigh-molecular-weight polyethylene and size-0 monofilament polypropylene suture, respectively. Limbs were prepared, and each patellar tendon was transected at a site chosen to simulate rupture at the proximal aspect. Tendons were repaired with the assigned techniques and mechanically tested with a biaxial servohydraulic test system; the clamp was distracted until the repair failed. Force at 1 -mm gap formation, 3-mm gap formation, and repair failure and gap size at failure were measured and compared between methods. Mode of failure was recorded. RESULTS There was no significant difference between methods for the force required to produce a 1 - or 3-mm gap in the repair. The 3LSLS technique required a significantly higher load for complete failure; gap formation immediately before failure was significantly greater for this method than for the M3LP technique. Four of 6 repairs with the M3LP technique and 0 of 6 repairs with the 3LSLS technique failed by suture breakage. CONCLUSIONS AND CLINICAL RELEVANCE The 3LSLS technique with size-2 ultrahigh-molecular-weight polyethylene suture was as effective at resistance to 1 - and 3-mm gap formation as the M3LP with size-0 monofilament polypropylene suture.

OBJECTIVE To compare the effects of 3 walkway cover types on temporospatial and ground reaction force measurements of dogs during gait analysis with a pressure-sensitive walkway (PSW). ANIMALS 35 client- and staff-owned dogs (25 nonlame and 10 lame). PROCEDURES In a crossover study design, all dogs were evaluated at a comfortable walk on a PSW to which 3 cover types (a 0.32-cm-thick corrugated vinyl mat or a 0.32- or 0.64-cm-thick polyvinyl chloride yoga mat) were applied in random order. Temporospatial and ground reaction force measurements were obtained and compared among cover types within the nonlame and lame dog groups. RESULTS Several variables, including maximum peak pressure, maximum force (absolute and normalized as a percentage of body weight), and vertical impulse (absolute and normalized) differed significantly in most comparisons among cover types for both nonlame and lame dogs. There was no significant difference in maximum force values between the 0.32-cm-thick corrugated vinyl and 0.64-cm-thick polyvinyl chloride cover types for both nonlame and lame dogs. CONCLUSIONS AND CLINICAL RELEVANCE To the authors’ knowledge, the cover type used during data collection with a PSW is rarely provided in published reports on this topic. The findings in this study suggested that to ensure that PSW data for dogs are collected in a standardized manner, the same cover type should be used during follow-up visits to evaluate clinical outcomes, for the duration of research studies, and at all locations for multi-institutional studies. The cover type should be specified in future PSW studies to allow direct comparisons of findings between studies.

OBJECTIVE To determine the impact of mechanical ventilation (MV) and perfusion conditions on the efficacy of pulse-delivered inhaled nitric oxide (PiNO) in anesthetized horses. ANIMALS 27 healthy adult horses. PROCEDURES Anesthetized horses were allocated into 4 groups: spontaneous breathing (SB) with low (< 70 mm Hg) mean arterial blood pressure (MAP; group SB-L; n = 7), SB with physiologically normal (≥ 70 mm Hg) MAP (group SB-N; 8), MV with low MAP (group MV-L; 6), and MV with physiologically normal MAP (group MV-N; 6). Dobutamine was used to maintain MAP > 70 mm Hg. Data were collected after a 60-minute equilibration period and at 15 and 30 minutes during PiNO administration. Variables included Pao2, arterial oxygen saturation and content, oxygen delivery, and physiologic dead space-to-tidal volume ratio. Data were analyzed with Shapiro-Wilk, Mann-Whitney U, and Friedman ANOVA tests. RESULTS Pao2, arterial oxygen saturation, arterial oxygen content, and oxygen delivery increased significantly with PiNO in the SB-L, SB-N, and MV-N groups; were significantly lower in group MV-L than in group MV-N; and were lower in MV-N than in both SB groups during PiNO. Physiologic dead space-to-tidal volume ratio was highest in the MV-L group. CONCLUSIONS AND CLINICAL RELEVANCE Pulmonary perfusion impacted PiNO efficacy during MV but not during SB. Use of PiNO failed to increase oxygenation in the MV-L group, likely because of profound ventilation-perfusion mismatching. During SB, PiNO improved oxygenation irrespective of the magnitude of blood flow, but hypoventilation and hypercarbia persisted. Use of PiNO was most effective in horses with adequate perfusion.

OBJECTIVE To determine whether the lactate threshold of dogs could be determined by a visual method and to assess the extent of agreement and bias among treadmill velocities for the lactate threshold as determined by visual (LTv) and polynomial (LTp) methods, glucose threshold as determined by visual (GTv) and polynomial (GTp) methods, and heart rate deflection point (HRdp) as a method for estimating the aerobic capacity of dogs. ANIMALS 18 healthy adult Beagles. PROCEDURES Each dog underwent a standardized incremental treadmill exercise test once. The test ended when the dog began to show signs of fatigue. Plasma lactate and glucose concentrations and heart rate (HR) were plotted against exercise intensity (treadmill velocity) for the duration of the test, and the LTv, GTv, and HRdp were determined visually. The LTp and GTp were determined by means of a second-order polynomial function. One-way ANOVA, Pearson correlation, Bland-Altman analyses, and ordinary least products regression were used to assess the extent of agreement and bias among the various threshold velocities. RESULTS Mean velocity did not differ significantly among the thresholds evaluated. There was a strong positive correlation between the LTv velocity and the velocity for GTv (r = 0.91), LTp (r = 0.96), GTp (r = 0.94), and HRdp (r = 0.95). CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that LTv could be determined for dogs undergoing intense exercise on a treadmill, and the treadmill velocity corresponding to the LTv was associated with the velocity for the other hallmarks of endurance. Thus, that method may be useful for prescription and evaluation of conditioning programs for dogs.

OBJECTIVE To evaluate and report measurements of the radiographic cardiac silhouette of healthy juvenile and adult ospreys (Pandion haliaetus). ANIMALS 54 ospreys (22 adults, 19 juveniles, and 13 birds of undetermined age) without clinical signs of cardiac disease and with adequate ventrodorsal radiographic images for cardiac silhouette assessment. PROCEDURES Radiographs of ospreys were assessed to determine cardiac width at the widest point as well as sternal width and thoracic width at the same level. Two-way mixed-effects models were used to evaluate interrater reliability for mean rating. Multivariable linear regression analysis was used to create predictive models of cardiac width and to establish a theoretical reference range for healthy ospreys. RESULTS Cardiac width of healthy ospreys was approximately 90% to 92% of sternal width and 67% to 69% of thoracic width. Both sternal width and thoracic width were significant predictors of cardiac width in independent predictive models as well as in a combined model after controlling for age. Thirty-four of 41 (83%) measured cardiac widths were within the theoretical reference range. CONCLUSIONS AND CLINICAL RELEVANCE Ospreys are sentinels used in monitoring environmental health. Environmental factors may have an impact on the cardiac health of ospreys, but reference values for healthy ospreys have not been established for use in assessing cardiomegaly in this species. The radiographic ratios and predictive model obtained in this study may be useful for objective evaluation of cardiomegaly in ospreys.

The objective of this study was to evaluate the effect of late-gestation vaccination of beef heifers with 2 doses of a killed-virus (KV) vaccine containing bovine herpesvirus 1 (BoHV-1), bovine viral diarrhea virus 1 (BVDV-1), and bovine viral diarrhea virus 2 (BVDV-2) on the serum concentrations of antibody against BoHV-1, BVDV-1, and BVDV-2 in heifers and their calves and on the IgG concentration in the calves. Of the 47 pregnant beef heifers selected, 26 received 2 doses of the vaccine at 6.5 to 8 mo of gestation (at pregnancy check), and 21 received 2 doses of saline. The mean log2 serum titers of neutralizing antibody against BoHV-1, BVDV-1, and BVDV-2 before vaccination did not differ significantly between the treatment groups; however, at calving all 3 mean titers were significantly greater (P < 0.05) in the vaccinated heifers than in the control heifers. At 24 h after birth the mean serum IgG levels in the calves did not differ significantly between the 2 groups, at 30.18 and 32.28 g/L, respectively (P < 0.05); however, the mean log2 serum titers of antibody to all 3 viruses were greater in the calves nursing colostrum from the vaccinated heifers than in the calves nursing colostrum from the nonvaccinated heifers and significantly so for BoHV-1 and BVDV-1 (P < 0.001 and P = 0.009, respectively). Thus, late-gestation vaccination of beef heifers could result in a greater and more consistent deposition of specific antibodies in colostrum, reducing the variability of initial titers in calves and increasing the duration of maternal immunity.

This study was planned to evaluate the extracts of Aloe vera (AV) at three different pH as a growth promoter, immunostimulant and coccidiostat agent in broiler production. This experiment was undertaken for a period of 35 days in the Institute of Animal Sciences, Faculty of Animal Husbandry, University of Agriculture Faisalabad, Pakistan. A total of 150 dayold broiler birds (Cobb) were randomly assigned to 5 treatments with 3 replicates of 10 chicks each. Birds were offered commercial broiler starter and finisher diets supplemented with 100 mg/kg enramycin (ANT), 50 ml/liter AV of pH 3 (AV1), 50 ml/liter AV of pH 7 (AV2), 50 ml/liter AV of pH 12 (AV3) and only fresh clean water (CON).

Aeromonas hydrophila is a Gram negative, positive oxidase, anaerobic, and opportunistic bacteria that, under certain conditions, become a pathogen (in humans and fish). This bacterium causes toxin and host infection in which different antibiotic resistance in isolated strains has been reported in different regions of the world. The aim of this study was to determine the prevalence of this bacterium and its susceptibility to common antibiotics in Tabriz city.50 samples from 5 Reared Oncorhynchus mykiss farms in Tabriz city (For each farm,10 numbers) were randomly assigned to suspected fish to the disease. By using biochemical tests, 14 samples (28%) from 2 Fish farms (40%) were identified as A.hydrophila. Antibiogram for these specimens showed that the bacterium had the highest resistance tovancomycin (100 %) and clindamycin (92.8%) antibiotics, and has the most sensitivity to the antibiotics ofsultrim, tetracycline and oxytetracycline with 71.4%.Considering the different antibiotic resistance pattern in this study and other similar studies,the necessity of examining the pattern of resistance in each region seems necessary.

In this study, microbiological quality of tantuni that have been consumed in the province of Van was examined. Materials and Methods: For this purpose; 100 tantuni samples, whose 79 of them were red meat tantuni (raw and cooked) and 21 of them chicken tantuni (raw and cooked) were used as material.According to analysis findings with regard to aerobic mesophilic organisms, coliform group microorganisms, E. coli, micrococcus/staphylococcus, S. aureus, C. perfringens and yeast-mould at the samples of raw and cooked red meat tantuni were found to be 5.67 and 3.98, 2.88 and 0.21, 0.87 and 2.00, 2.99 and 2.27, 1.33 and 0.25, 0.05 and 1.00, 4.43 and 0.63 log cfu/g respectively. In the same order, at the samples of raw and cooked chicken tantuni were found to be; 4.35 and 3.77, 2.84 and 1.00, 1.15 and 2.00, 0.95 and 1.22, 2.00 and 2.00, 1.00 and 1.00, 4.05 and 0.11 log cfu/g respectively. Salmonella spp. could not be isolated in the tantuni samples that had been investigated. In the raw red meat tantuni samples 5.26% (1/19) had S. aureus, in the cooked red meat tantuni samples 1.66% (1/60) had S. aureus, and 3.33% (2/60) yeast-mould which were not compatible with the limit values that were stated at the Turkish Food Codex were found. However, values obtained from this study show that during preparation and production of the goods and in the other stages; hygienic rules have not been carried out.In conclusion to secure the product safety, it is essential to be cautious for the temperature and time during preparation, reservation temperature and GMP/GHP based applications in the preparation of tantuni.
