The point prevalence of gastro-intestinal parasites in calves, sheep and goats in Magadi division, south-western Keny

Helminths cause great economic loss in livestock in Africa, and can be categorized as either direct or indirect losses. Arid and semi-arid lands (ASAL) in Kenya comprise 71 % of total land area and harbour the largest population of cattle, sheep and goats. However, little information on the distribution and impact of gastro-intestinal (GIT) parasitism in these animals is available. This survey was conducted to establish the prevalence of GIT parasites infecting calves, sheep and goats and their relative importance in Magadi division, which is semi-arid. Faecal samples were obtained directly from the rectum of 109 calves, 133 goats and 20 sheep and submitted to the laboratory for faecal worm egg counts, and coccidial oocysts examination using a modified McMaster method. The significance of differences in mean egg count per gram (epg) between animal species and herds (farms) were assessed using analysis of variance. The overall prevalence of nematodes in the calves, sheep and goats was 69.2 %, 80 % and 82 %, respectively. About 10 % of sheep and goats had epgs higher than 1 000, the remainder having light to moderate infections. The overall prevalence of coccidial oocysts in calves, sheep and goats was 30 %, 44 % and 45 %, respectively. Poor productivity in ASAL areas, where nutrition is often poor, is likely to be pronounced in the presence of parasite infections. These findings indicate that viable internal parasite control should be implemented in the study area in order to increase the productivity of the livestock there.

Striped mice, Rhabdomys pumilio, were trapped over a period of 17 months in the Thomas Baines Nature Reserve, and placed in cages, over water, until all the ticks they harboured had detached. The mice were then returned to the reserve. Four ixodid tick species were recovered from the mice of which the larvae and nymphs of Rhipicephalus follis and Rhipicephalus simus were the most numerous. Most larvae of R. follis detached from mice trapped from March to July, and most nymphs in March and from June to September. Most larvae of R. simus detached from mice trapped from December to March, and most nymphs from January to March and during May and June. Seven ixodid tick species were collected from striped mice, house rats, Rattus rattus, vlei rats, Otomys spp. and Praomys sp. captured in the vicinity of human dwellings or animal holding facilities in the Grahamstown district. One third of the manufacturer's prescribed dose of diminazene has long been used to block treat the South African unfrozen Babesia bigemina and Babesia bovis (redwater) vaccine reactions, with no known adverse effects. It is known that the inhibitory effect of antibabesial drugs is more pronounced in animals inoculated with the frozen vaccine than those with the unfrozen vaccine. Reports of vaccine failures in some animals in which diminazene was used for block treatment of the reactions following inoculation with frozen South African redwater vaccine led us to reinvestigate the required waiting period before treatment and the reduced dose necessary for successful treatment and development of immunity. Results from febrile reactions in cattle following vaccination indicated day 7 as the optimal day for administering block treatment. Treatment of B. bigemina vaccine reactions in cattle on day 7 at a level of 0.35 mg/kg (1/10 fraction of the normal dose) diminazene killed all the parasites while B. bovis vaccine parasites survived treatment using diminazene at levels between 0.35 mg/kg and 1.16 mg/kg. However, various other factors, such as the degree of natural resistance of different cattle breeds and individual animals, the accuracy of diminazene content according to the manufacturer's label claim and the accuracy of the drug dose administered, all influence the successful immunization of animals. Consequently block treating of Babesia vaccines with diminazene on day 7 after vaccination is not recommended.

The aim of this study was to determine for reference purposes the values of serum albumin, a1-globulin, a2-globulin, b-globulin, g-globulin, and a-lipoprotein (high density lipoprotein), pre-b-lipoprotein (very low density lipoprotein) and b-lipoprotein (low density lipoprotein) fractions of normal ostriches (Struthio camelus) in Turkey. Five male and five female ostriches, 18 months old, were used. All the ostriches were fed on a diet that contained 15.14 % crude protein and 2 950 Kcal/kg of metabolizable energy. The serum protein and lipoprotein fractions were measured using agarose gel electrophoresis. The fractions were found to be 60.96 % albumin, 0.24% a1-globulin, 15.91 % a2-globulin, 13.34 % b-globulin, 9.55 % g-globulin, 53.77 % HDL, 0.60 % VLDL and 48.09 % LDL.

Inactivation of SARS coronavirus by means of povidone-iodine, physical conditions, and chemical reagents

The efficacy of several povidone-iodine (PVP-I) products, a number of other chemical agents, and various physical conditions were evaluated for their ability to inactivate the severe acute respiratory syndrome coronavirus (SARS-CoV). Treatment of SARS-CoV with PVP-I products for 2 min reduced the virus infectivity from 1.17 x 10**6 TCID sub(50)/ml to below the detectable level. The efficacy of 70% ethanol was equivalent to that of PVP-I products. Fixation of SARS-CoV-infected Vero E6 cells with a fixative including formalin, glutaraldehyde, methanol, and acetone for 5 min or longer eliminated all infectivity. Heating the virus at 56 deg C for 5 min dramatically reduced the infectivity of the virus from 2.6 x 10**7 to 40 TCID sub(50)/ml, whereas heating the virus for 60 min or longer eliminated all infectivity. Irradiation with ultraviolet light at 134MicroW/square cm for 15 min reduced the infectivity from 3.8 x 10**7 to 180 TCID sub(50)/ml; however, prolonged irradiation (60 min) failed to eliminate the remaining virus, leaving 18.8 TCID sub(50)/ml. We believe that these findings will be useful for the implementation of infection contiol measures against SARS, and for the establishment of effective guidelines for the prevention of SARS outbreaks.

In MRJ/MpJ mice, there is a genetic mutation of exonuclease 1 (Exol), in which the exon 9 is sometimes deleted. In the present study, to check the gen-eration of the spliced exons, exon 8-intron 8-exon 9 (pCX/Ex/EIE/B and pCX/ Ex/EIE/M) plasmids were temporally transfected in vitro into BALB 3T3 cells, and RT-PCR using appropriate primer pair was carried out 1 day after transfection. In these constructions, pCX/Ex/EIE/B was derived from genomic sequence of C57BL/6 mice, and pCX/Ex/EIE/M was from MRL/MpJ. A spliced band was detected in pCX/Ex/EIE/B , but was present little or very weakly in pCX/Ex/EIE/M . Next, the same spliced band was demonstrated in pCX/Ex/EIE/M(T) plasmid, in which the branchpoint sequence (BPS) of pCX/Ex/EIE/M including the exon 9 was changed into that of pCX/Ex/EIE/B. The splicing did not occur in the del1/B mutant, in which 1960 nucleotides of the intron 8 were deleted, whereas it was detected in the del2/B plasmid deleted 1036 nucleotides in its middle region. These results suggest that the nucleotide T to A mutation of the BPS in the intron 8 is at least a sufficient for generation of splice variants (tr-l and tr-2 Exol).

Although tumor necrosis factor (TNF) alpha is an important key factor in degeneration of equine superficial digital flexor tendon (SDFT), the dynamism of TNF receptors and associated factors on tendinocytes has not been elucidated. To reveal signaling events mediated by TNF-receptors (TNF-Rs) in tendinocytes, we focused on four signaling factors, TNF-R1, TNF-R2, TNF-Rassociated factor 2 (TRAF2) and nuclear factor-kappa B (NE-KappaB) , and investigated the distribution and production of these factors. Cultured tendinocytes were obtained from SDFTS of thoroughbred horses. The tendinocytes were treated with 10ng/ ml equine TNFAlpha medium for 6 hours and then the four factors on tendinocytes were visualized by using an immunohistochemical method, and the amounts of the four factors were determined by Western blot analysis. Although TNF-R1 and TNF-R2 co-localized on the same tendinocyte, in untreated control cells (normal condition), immunoreactivity against TNF-R1 was very weak but TNF-R2 showed a strong reaction. However, TNF-R1 showed the same high level of reaction as TNF-R2 in TNFAlpha-treated cells (inflamed condition). Intense TRAF2 and NF-KappaB were detected at inflamed condition, however both factors were also detected at normal condition. The distinct distributions of the four factors under different conditions (normal and inflamed condition) in uitro not only reflect the dynamism of the cytokines but may also provide important clues for a means to prevent from occurrence of tendonitis and progress of tendon degeneration.
