Some reproductive hormones in relation to ovarian activity in rats

The present study is an endeavor to explore the relationship between induced hormonal alterations and ovarian activity in female rats. Fifty mature cycling female Albino rats were left for one week for acclimatization and offered balanced diet and water ad libitum. Animals were daily examined by vaginal smears to determine regularity of the estrous cycle. Rats were equally divided into 5 groups; control, hyperglycemic, hypoglycemic, hypercorticosteroid and hypocorticosteroid. At the end of 3 cycles, individual sera were obtained to determine glucose, malondialdehyde (MDA), estradiol and progesterone levels. Moreover, tissue specimens of the ovaries and the Fallopian tube were taken for histopathological examination. Results showed that all hormonal treatments induced alterations in the cellular characteristics of the cycle. Gonadosomaic index (GSI) did not show any remarkable variation. Alloxan or insulin treatments affected significantly serum glucose level in rats as. On the other side, hypercorticism led to marked elevation of glucose while hypocorticism showed no significant effect. Serum MDA showed significant elevation only in hyperglycemic and hypocorticoid groups. Hypoglycemia led to a significant decrease in serum estradiol while other treatments had no effect. Upon progesterone, hypoglycemia resulted in an increased level of the hormone while other treatments led to reduced levels of the hormone. It was evident that abnormalities of the adrenal corticosteroids and / or pancreatic insulin levels are concomitant with irregular estrous cycle as well as ovarian and Fallopian tube alterations which led to deviated gonadal folliculogenesis and steroidogenesis.

Avian paramyxovirus type-1 (APMV-1) viruses of the lentogenic pathotypes are often isolated from wild aquatic birds and may mutate to high pathogenicity when they cross into poultry and cause debilitating Newcastle disease. This study characterised AMPV-1 isolated from fresh faecal droppings from wild aquatic birds roosting sites in Uganda. Fresh faecal samples from wild aquatic birds at several waterbodies in Uganda were collected and inoculated into 9–10-day-old embryonated chicken eggs. After isolation, the viruses were confirmed as APMV-1 by APMV-1-specific polymerase chain reaction (PCR). The cleavage site of the fusion protein gene for 24 representative isolates was sequenced and phylogenetically analysed and compared with representative isolates of the different APMV-1 genotypes in the GenBank database. In total, 711 samples were collected from different regions in the country from which 72 isolates were recovered, giving a prevalence of 10.1%. Sequence analysis of 24 isolates revealed that the isolates were all lentogenic, with the typical 111GGRQGR’L117 avirulent motif. Twenty-two isolates had similar amino acid sequences at the cleavage site, which were different from the LaSota vaccine strain by a silent nucleotide substitution T357C. Two isolates, NDV/waterfowl/Uganda/MU150/2011 and NDV/waterfowl/Uganda/MU186/2011, were different from the rest of the isolates in a single amino acid, with aspartate and alanine at positions 124 and 129, respectively. The results of this study revealed that Ugandan aquatic birds indeed harbour APMV-1 that clustered with class II genotype II strains and had limited genetic diversity.

Medicinal herbal feed that used as feed additives are widely applied in livestock production and may be applicable to aquaculture production systems. The present study explores the modulatory effects of incorporation of herbal combination of black cumin seeds (Nigella sativa), peppermint (Mentha piperita) and fennel (Foeniculum vulgare), on African Sharptooth Catfish, Clarias gariepinus. For this purpose, a total of 72 catfish were randomly distributed into 6 groups of 12 fish each and fed on the experimental diets for 45 successive days. Fish of first group were fed on basic diet only while, those of the second one were fed on basic diet supplemented with 0.5% from each medicinal plant while, other 4 groups were subjected to challenge experiment by injection with Vibrio vulnificus. The third group was kept non-injected, fourth group was injected by saline, fifth and sixth groups were injected by Vibrio vulnificus after feeding with basic diet and basic diet with herbs, respectively. Results proved that, total antioxidant capacity, serum total proteins, albumins and globulins were observed to be significantly higher in the treated group as compared to the control. Lymphocytes percent increased significantly in herbal fed group comparing to the control group while, neutrophils percent decreased significantly. Although serum urea level was not affected by herbal supplement, serum creatinine level was decreased significantly. Body weight gain of C. gariepinus increased significantly after herbal administration. Catfish challenged with a Vibrio vulnificus isolate and received the herbal diet showed less mortality than the control group. Fish fed on the herbal diet exhibited normal histological structure of liver, kidney and spleen. In conclusion, based on the current results together with the low cost and the potential antioxidant and immune effects of mixed herbal medicinal plants used in the study, it is recommended to be used in fish feed to diminish the mortalities caused by some aquatic pathogens.

This work was planned to investigate the bacteria isolated from broiler chickens head suffered from naked eye pathological lesions. Out of 200 examined head lesions, the result revealed that the major pathogens associated with swollen head syndrome (SHS) were Escherichia coli, Streptococcus dysgalactiae and Pseudomonas aeruginosa. Antimicrobial sensitivity pattern against 11 different antimicrobials proved that isolates were resistant to most of the tested antimicrobial agents. PCR was applied on 4 MDR E. coli, 4 S. dysgalactiae and 2 P. aeruginosa for detection of some resistance and virulence genes. The results of E. coli isolates revealed that blaTEM gene was the most prevalent in all isolates (100%) followed by tetA (A), aada1, aada2 and aacC genes. Meanwhile tetA (B) gene was found in 3 (75%), while aadB gene was not detected in any isolates. All S. dysgalactiae proved to harbour 16srRNAgene also all S. dysgalactiae were 100% positive for tuf gene followed by speF gene which found in 2 isolate (50%). The results of PCR of P. aeruginosa isolates revealed that toxA gene was the most prevalent gene found in all isolates (100%) followed by lasI. Then, phzM gene was found in one isolate (50%).

The experiment was designed to investigate the immunomodulating effect of lector 50 on general health and immune response of broiler chicks to Mycoplasma gallisepticum vaccination in commercial broiler chicks. the obtained results reveled significantly higher effects on body weight ,bursal, and thymic index on lector treated group of chickens, while no effects on spleen index. Also significant improvement in total and differential leukocytic count as well as significantly higher antibody titer was detected by ELISA in lector 50 treated groups.

The current study was performed to evaluate the effect of acute and chronic hepatotoxicity induced by paracetamol and thioacetamide respectively on serum hormonal levels and biochemical parameters. Female Wistar albino rats were divided into 3 equal groups (C), (P) and (T). Group (C) were kept as control, group (P) were received paracetamol orally (500 mg/kg b.wt) daily for 15 days and those of group (T) were injected thioacetamide (200 mg/kg b.wt) intraperitonialy twice/ week for 90 days. In P group, results revealed significant elevation in liver enzyme activities (ALT, AST and ALP), T4, insulin (7th day), estrogen (7th and 15th days), triglycerides (7th day) and cholesterol levels throughout the experiment while serum proteins and T4 (15th day) showed significant decreased values. Whereas, at 90th days of chronic intoxicated group (T) resulted in significant elevation in liver enzyme activities (ALT, AST and ALP), bilirubin, estrogen, T4, triglycerides (60th and 90th days) and T3 (120th day). While the levels of T4 and cortisol (60th day), serum total protein, albumin, globulin (90th day) and insulin (120th day) showed significant decreased values when compared to control group. In conclusion, both paracetamol and thioacetamide cause different degrees of damage in liver of rats leading to clear changes in their hormonal and biochemical profiles.

Locally isolated EHV-1 (2002) was successfully adapted to different available cell lines (BHK, VERO, MDBK and MDCK). Characteristic cytopathic effect (CPE) was observed from the first passage and the virus growth became regular with constant incubation periods at 2nd day in BHK and VERO cells 4th day in both MDBK and MDCK cells post inoculation with maximum titers of 7, 8.5, 7.75, 6.2 log10 TCID50/ml in BHK6, VERO10, MDBK5 and MDCK5 cell lines; respectively. Growth kinetics of EHV-1 in VERO cells, showed initial increase in cell associated viruses (CAV) and cell free viruses (CFV) were at 8 and 48 hours post inoculation then reaching their peak at 72 and 96 hours post inoculation in CAV and CFV; respectively. Detection of viral protein in infected cells was done using IFT. It revealed that high fluorescence was shown at 4 till 72 hours post inoculation in CAV and 72 hours in CFV. In conclusion, EHV-1 is more cell associated and IFT is considered as a rapid and sensitive test for detection of viral protein

Diabetes is characterized by hyperphagia, and polydypsia. However, the mechanisms by which diabetes produces these effects are not clear. This study was conducted to examine changes in serum insulin and leptin levels in induced-type 1 diabetes mellitus in relation to concomitant changes in body weight, glycemic state and lipid profiles in rats. Moreover, we aimed to clarify that the treatment with lipoic acid (LA) is capable of reversing these effects or not. Ninety-six male rats were divided into 3 groups, control group (32 rats) was considered as normal non-diabetic, 64 rats were subcutaneously injected with alloxan (120 mg/kg.b.wt) for induction of diabetes. Then the diabetic rats were divided into two equal subgroups, the first is diabetic group that was not treated with LA, and the other is LA-treated diabetic group that was treated with LA at a dose 100 mg/kg b.wt / day for four weeks. Body weight, serum lipid profile, glucose, insulin, homeostasis model assessment– insulin resistance (HOMA-IR) and leptin were measured. The data showed significant increase in serum triacylglycerol, total cholesterol and glucose levels as well as HOMA-IR while significant decrease in the mean body weight gain, serum insulin and leptin levels in diabetic group in comparison with control group. The treatment with lipoic acid led to significant decrease in serum fasting and postprandial glucose, triacylglycerol and total cholesterol levels as well as slight decreased HOMA-IR with significant increased levels of serum insulin and leptin in comparison with diabetic group. It could be concluded that alloxan-induced diabetes led to hyperglycaemia, insulin resistance, hyperlipideamia and hypoleptinamia. Moreover, treatment with lipoic acid ameliorates these changes and improves insulin sensitivity.

Fifty five milk samples were collected from dairy Friesian cows suffering from clinical mastitis in a farm at Quena Governorate. The samples were subjected to microbiological examination .The main isolated pathogenic bacteria were identified biochemically as Escherichia coli (36.3%), Staphylococcus aureus (23.6%) Streptococcus agalactiae (18.2%), Klebsiella pneumoniae (12.7%) and bacterial free samples (9.1%) . Sensitivity tests for the bacterial isolates revealed that the tested strains were sensitive to ciprofloxacin, nortril, gentamicin and lincospectin

Salmonellosis is a major problem for the poultry industry, and this problem represents a critical food safety hazard. Resistance to antimicrobial agents within nontyphoidal Salmonellae is a serious problem. The present study aimed to analyze genetically some β-lactamase resistance genes and some virulence associated genes in Salmonella isolates from broiler chicken. Five hundred samples were collected from diseased broiler chickens of different ages (3-6 weeks) from different farms in Assiut Governorate in Egypt during the period from January 2015 to December 2015. Bacteriological examination showed that 26 Salmonella isolates were recovered with a prevalence rate of 5.2% Serotyping of Salmonella isolates showed that S. Enteritidis S. Typhimurium, and S. Kentuky were identified at rates of 50%, 30.8% and 19.2%, respectively. Results of antibiogram showed that 18 Salmonella isolates (92.3%) were multidrug resistant. All isolates were screened for the presence of 2 β-lactamase resistance genes (blaCTX and blaCMY) as well as 3 virulence genes (stn, invA and hilA) using multiplex PCR. The overall prevalences were 53.9% for blaCTX and 34.6% for blaCMY. Meanwhile, stn, invA and hilA genes were found in 96.2%, 100% and 84.7% of isolates, respectively.
