Monitoring of Brucella sp., Coxiella burnetii and aflatoxin M1 in goat milk from Johor Полный текст

Forty-nine fresh goat’s milk samples produced by local farmers and sold in market for public consumption as well as raw goat milk in Johor, Malaysia were analysed for total plate count(TPC) , E. coli, Coliform, Brucella melitensis, Brucella abortus,Coxiella burnetii as well as aflatoxin M1 (AFM1) content, as measures for food safety. The mean counts per ml for TPC were 4.90 x 105, 6.50 x 105, 1.60 x 105 and 1.48 x 106 for pasteurised, unpasteurised and unknown (status of pasteurisation) milk sold in the market as well as the raw milk from milkcollection center (MCC), respectively. Among pasteurised samples, only one had TPC count higher than the permitted level whereas the rest were all within the permitted level. The mean counts per ml for E. coli were <1.00 x 102 for pasteurised and unknown milkwhereas 1.67 x 101 for unpasteurised and 1.18 x 102 for raw milk. The mean counts per ml for coliform were 9.53 x 103, 9.76 x103, 1.20 x 102 and 1.16 x 104 for pasteurised, unpasteurised, unknown milk and raw milk, respectively. Overall, no significantdifferences on the bacterial counts in both pasteurised and unpasteurised milk. All milk samples were negative of B. melitensis and B. abortus, but one unknown sample fromthe market and two raw samples from MCC were positive of C. burnetii through the ELISA test. The unknown sample from the market showed the presence of C. burnetii when further analysed microscopically. Meanwhile, no sample exceeded the permitted level of AFM1 in milk.

A preliminary study on the effect of probiotics in fermented palm kernel meal (FPKM) and total mixed ration (TMR) to improve milk production in dairy cattle was carried out. TMR feeding enhances feed intake, improves the ecology of the rumen leading to stimulated microbial activity to digest more feed, and then finally increases productivity of the cows. Probiotics ofBacillus spp. was given through concentrate after a fermentation process for three days. The trial was carried out at the Dairy Unit, Veterinary Institute, Kluang, Johore. Twenty Mafriwal cows with low milk production were selected and divided into twoequivalent groups. Each group consisted of 10 cows. Group A were fed with TMR. Group B received TMR and 100 grams perhead/day FPKM (FPKM, probiotics Bacillus spp. and mollases). The cows were given TMR daily which consisted of 20 kg per headof fresh Napier grass, 5 kg per head palm kernel meal, 1 kg per head soya bean meal, 1 kg per head corn, 0.5 kg per head molassesand 0.5 kg per head of enalac. Feeding trial period was carried out for four weeks. Feed was given twice a day, morning and lateafternoon, meanwhile water was given adlibitum. Animals were given the same diet for seven days for an adaptation period.Milk was sampled twice a day for four weeks and were recorded accordingly. The average of daily and weekly milk productionwere calculated. The animals from Group B showed an improvement of milk production before trial to the fourth week from 5.2litres/head/day to 6.99 litres/head/day i.e. 14.80% to 34.44% as compared to Group A.

Ever since infectious bursal disease (IBD) was recognised in Nigeria over forty years ago, it continues to pose a threat to poultry production with limited information on the likely role of other avian species especially those raised in close proximity with chickens. For this study, blood samples were obtained from184 unvaccinated apparently healthy birds comprised of Japanese quails (63) andindigenous chickens (60) on smallholdings as well as pigeons (61) in a live-bird market in Osun and Oyo states, southwest Nigeria.Sera from these birds were analysed for IBD virus antibodies using a commercial ELISA kit. Overall, 69 (37.5%) sera were positive for IBDV with 52.8% (65/184) and 6.6% (4/184)from birds on smallholdings and live-bird market, respectively. These findings indicate that these birds were sub-clinically infected and could serve as reservoirs shedding the virus into the environment and perhaps, corroborate the suggestion that the inability to effectively control or eradicate the disease from poultry flocks in Nigeria may be due to limited information on the contributions of other avian species other than chicken in the spread of IBD virus.

As of 2016, a total of 38 research projects have been undertaken, with the involvement of 38 principal researchers and 266 co-researchers from various Malaysian universities, research institutes and government agencies, in collaboration with industry partners, to help solve some of the issues faced by EBN industry. In the early stage, research areas were focused onfundamental works which are important to provide the scientific basis for some of the claims made by producers, assisting in issues raised by ranchers and industry, and to gather new knowledge in swiftlet and EBN. The researchers worked mainly based on the problems faced by the industry. Besides publications in journals, proceedings and books, more than 30 Masters, PhDs and technical experts have been trained under these projects. In addition, several patents have been filed, social innovations and ideas have been shared with the community and industry. A few products are ready to go to the next level for commercialisation. Industry partners are involved in many of these stepsand outputs. Some of the issues faced by the industry have been partially solved and are in the process of refinement. The engagement with the industry will be further strengthened by getting more industry partners to be involved in the relevantprojects specific to problems faced by the industry, for the benefits of EBN industry and nation’s wealth creation.

Chromobacterium violaceum is a facultative anaerobic, gram-negative rodshaped bacteria normally found in soil and stagnant water of tropical and subtropical countries. Infections of Chromobacterium violaceum are rare among mammals, but the first human infection were reported in Malaysia in 1927. This clinical case reports two separate cases of Chromobacterium violaceum infection in two sub-adult male Black-handed Gibbon (Hylobates agilis). Both animals were presented with the history of diarrhea, pyrexia and inappetance. From the physical examination, the findings were high body temperature, dull, inactive, soft fecal stained at the rectum and small open wound at lower limb of one of the case. The treatment given was antipyretic and multivitamin. Unfortunately, due to poor prognosis both patients died within 48 hours after presentation and treatment. Autopsy examinations wereperformed to find out the cause of death. Post mortem examination findings revealed hepatomegaly with multiple size whitish-yellow spots on the liver surface, congestion and enlargement of spleen and lymph node, congestion of lung and loss of demarcation between renal cortex and medulla of the kidney. The cellular changes via histopathological findings of kidney, spleen, lung and liver were suggestive of septicaemia. The gross and histopathologicalfindings were supported by the isolation of Chromobacterium violaceum via bacterial isolation and identification from lung, liver, spleen and kidney. Thus, the cause of death of the two sub-adult male Black-handed gibbon in this case are due to septicaemiadue to Chromobacterium violaceum infection.

Napier grass (Pennisetum purpureum) has been the most promisingand high yielding fodder giving dry matter yields. This experiment was conducted to determine the effect of cutting intervals on the yield and nutrient composition of six cultivars of Napier grass (Pennisetum purpureum) which is 3rd Generation Napier, India Napier, Kobe Napier, Red Napier, Taiwan Napier and Zanzibar Napier. The grasses were cut close to the ground levelto get a uniform stand on day 70 after planting and the cutting intervals were at 35 and 42 days and carried out for 3 times.After each harvest, the rates of maintenance fertiliser used were 150 kg of nitrogen, 60 kg of phosphorus and 100 kg of potassium per hectare per year. Harvested plant material was weighed, pre-dried in a forced-air drying oven at 60 °C overnight before grinding. Ground samples were used to determine dry matter (DM), crude protein (CP), crude fibre (CF) and metabolised energy. The data were analysed using Statistical Analysis System (SAS™) followed by Tukey’s post-hoc test. AP value of less than 0.05 (p<0.05) are considered statistically significant. Results showed that increasing the cutting interval (i.e. advancing age of maturity) increased dry matter and crude fibre significantly.However, in terms of nutrient content, crude protein and metabolised energy percentage was markedly decreased as thecutting interval increased. From the results presented, it is clear that cutting a stand of Napier grass at 35 days will achieve greater yield and nutrient content.

In Malaysia, Guinea grass (Megathrysus maximus) and Napier grass(Pennisetum purpureum) are considered as good palatable fodder for goats and have been used widely. The objective of this study is to evaluate the potential grasses for goats to have optimum growth performance and optimum feeding cost. Fourteen male Jamnapari goats with an average age of five months were used in this experiment. All animals were individually weighed anddivided equally (seven goats) into two treatment groups. Group 1 was fed with cut and carry Guinea grass while group 2 animals were fed cut and carry Napier grass, and both groups were given the same quality and quantity of PKC and molasses. There was no significant difference observed in the average final weight (AFW), average weight gain (AWG) and average daily gain (ADG) among the two dietary treatment group. However, dry matter intake (DMI) and feed conversion ratio (FCR) showed a significant difference. The group fed with Napier grass showed lower ADMI and FCR. From the viewpoint of economic evaluation, feeding Guinea grass and Napier grass as an animal feed is economincally viable for local farmers.

The major causes of the decline of the Dugong population along the urban coast of Mersing, Johor includes gill netting, subsistence hunting, habitat loss from extreme weather events that are likely to be exacerbated by climate change, humansettlement, breach of the bots and sea water pollution. In March 2017, a male Dugong dugon, estimated age of 10 to 20 years, wasincidentally found dead near Pulau Tinggi, Mersing, Johor by a fisherman. The entire body was found to have old and new scarsat the anterior part of the abdomen. The right eye was protruded out and bleeding which indicated that the eye was piercedby a sharp object. Necropsy revealed the upper small intestine and the stomach compartments were semi-impacted with a massive helminth burden (more than 1,000 nematodes). Paradujardinia halicoris worms were identified based on morphological characteristics. Zoonotic fungus named Madurella mycetomi were isolated from heart and lung after incubation for 14 days.Histologically, the lung was confirmed to have the presence of big mast cells which formed capsules, indicating presence offungal spores causing systemic mycoses where the macrophages invade and engulf the spores. The shrinkage of myocardium myocyte and myocardium necrosis with mild vasculitis indicates heart failure. Groucott’s stain confirmed Madurella mycetomi infection that induced systemic pulmonary and myocardium mycosis in Dugong dugon.

OBJECTIVE To evaluate pharmacokinetic and pharmacodynamic characteristics of 3 doses of tapentadol hydrochloride orally administered in dogs. ANIMALS 6 healthy adult mixed-breed dogs. PROCEDURES In a prospective, randomized crossover study, dogs were assigned to receive each of 3 doses of tapentadol (10, 20, and 30 mg/kg, PO); there was a 1-week washout period between subsequent administrations. Plasma concentrations and physiologic variables were measured for 24 hours. Samples were analyzed by use of high-performance liquid chromatography–tandem mass spectrometry. RESULTS Tapentadol was rapidly absorbed after oral administration. Mean maximum plasma concentrations after 10, 20, and 30 mg/kg were 10.2, 19.7, and 31 ng/mL, respectively. Geometric mean plasma half-life of the terminal phase after tapentadol administration at 10, 20, and 30 mg/kg was 3.5 hours (range, 2.7 to 4.5 hours), 3.7 hours (range, 3.1 to 4.0 hours), and 3.7 hours (range, 2.8 to 6.5 hours), respectively. Tapentadol and its 3 quantified metabolites (tapentadol sulfate, tapentadol-O-glucuronide, and desmethyltapentadol) were detected in all dogs and constituted 0.16%, 2.8%, 97%, and 0.04% of the total area under the concentration-time curve (AUC), respectively. Plasma AUCs for tapentadol, tapentadol sulfate, and tapentadol-O-glucuronide increased in a dose-dependent manner. Desmethyltapentadol AUC did not increase in a linear manner at the 30-mg/kg dose. Sedation scores and heart and respiratory rates were not significantly affected by dose or time after administration. CONCLUSIONS AND CLINICAL RELEVANCE Oral administration of tapentadol was tolerated well, and the drug was rapidly absorbed. Adverse events were not apparent in any dogs at any doses in this study.

Squamous cell carcinomas are malign tumors composed of squamous epithelium of skin and mucous membranes and showing squamous differentiation. A male, 11-year-old Chinchilla race cat was brought to Ankara University, Veterinary Faculty, Veterinary Hospital, Department of Internal Diseases with complaints of anorexia, weakness, dysphagia, fatigue and difficulty in swallowing solid food. Metronidazole (15 mg/kg, iv), amoxicillin-clavulanic acid (25 mg/kg, sc) and fluid treatment were administered to the patient, respectively. As a predisposing factor in the development of oral squamous cell carcinoma in cats, exposure to cigarette smoke, the use of flea collar, and especially nutrition with tuna fish- containing ingredients are mentioned.
