Brucellosis is an endemic disease in Zimbabwe caused by the genus Brucella. Brucella seroprevalence was recently reported to be high in the wildlife-livestock interface in the Chiredzi district and the neighbouring Gonarezhou National Park (GNP) in Zimbabwe, and higher amongst communal cattle with an abortion history and access to grazing in GNP than amongst communal cattle with no abortion history or access to grazing in GNP. The aim of this study was to investigate Brucella species in brucellosis seropositive cattle in the Chiredzi district with access to GNP using isolation and identification. Isolation of Brucella species from whole blood (n = 18) and milk samples (n = 10) from seropositive animals with an abortion history was based on the rose Bengal test (RBT) and enzyme-linked immunoassays (enzyme- linked immunosorbent assay [ELISA]; indirect ELISA and complement ELISA), using microbiology and polymerase chain reaction (PCR) methods. Brucella abortus was cultured and identified from blood and milk collected from seropositive cows in both communal areas. The Brucella-specific 16-23S intergenic spacer (ITS) PCR and multiplex AMOS-PCR assays verified the identification of the cultures. Our results confirmed that B. abortus is present in cattle on communal farms in the Chiredzi district in Zimbabwe and might cause cattle abortions. The need for implementing control measures and raising public awareness on zoonotic transmission of brucellosis are recommended. (Résumé d'auteur)

estimates of glomerular filtration rate (GFR) in dogs with various degrees of renal function. Animals—50 Great Anglo-Francais Tricolor Hounds with various degrees of renal function. Procedures—Boluses of iohexol (40 mg/kg) and creatinine (647 mg/kg) were injected IV. Blood samples were collected before administration and 5 and 10 minutes and 1, 2, 4, 6, and 8 hours after administration. Plasma creatinine and iohexol concentrations were assayed via an enzymatic method and high-performance liquid chromatography, respectively. A noncompartmental approach was used for pharmacokinetic analysis. Pharmacokinetic variables were compared via a Bland-Altman plot and an ANOVA. Results—Compared with results for creatinine, iohexol had a significantly higher mean ± SD plasma clearance (3.4 ± 0.8 mL/min/kg vs 3.0 ± 0.7 mL/min/kg) and a significantly lower mean volume of distribution at steady state (250 ± 37 mL/kg vs 539 ± 73 mL/kg), mean residence time (80 ± 31 minutes vs 195 ± 73 minutes), and mean elimination half-life (74 ± 20 minutes vs 173 ± 53 minutes). Despite discrepancies between clearances, especially for high values, the difference was < 0.6 mL/min/kg for 34 (68%) dogs. Three dogs with a low GFR (< 2 mL/min/kg) were classified similarly by both methods. Conclusions and Clinical Relevance—Plasma iohexol and creatinine clearances can be used interchangeably for screening patients suspected of having chronic kidney disease (ie, low GFR), but large differences may exist for dogs with a GFR within or above the reference range. Objective: To assess long-term effects and risk factors for the efficacy of hyperimmunization protocols against infectious bovine rhinotracheitis (IBR) during a longitudinal field study of dairy and dairy-beef mixed farms. Animals: Approximately 7,700 cows from 72 farms. Procedures: Farms were assigned to 3 treatment groups (hyperimmunization groups [HIGs] 1 and 2, which were hyperimmunized with glycoprotein E [gE]–deleted marker vaccines, and a nonintervention group [NIG]). Cattle in HIG 1 were initially vaccinated with an attenuated vaccine, whereas cattle in HIG 2 were initially vaccinated with an inactivated-virus vaccine. Cattle in both HIGs received booster inoculations with inactivated-virus vaccines at 6-month intervals. The risk for gE seroconversion was compared among experimental groups via a shared frailty model with a piecewise constant baseline risk to correct for seasonal and secular effects. Results: Risk for gE seroconversion significantly decreased over time for the HIGs, compared with the NIG. Seasonal changes in the risk of gE seroconversion were detected, with a higher risk during winter periods, compared with grazing periods. No significant difference was detected between HIGs 1 and 2. The only significant risk factor was the number of buildings for cattle on a farm; the higher the number of buildings, the lower the risk for gE seroconversion. Prevalence of IBR decreased over time in both HIGs but remained constant or increased in the NIG. Conclusions and Clinical Relevance: Hyperimmunization via repeated administration of attenuated and inactivated-virus gE-deleted marker vaccines as well as inactivated-virus vaccines may provide a method for control of IBR.

peer reviewed | Objective—To culture equine myoblasts from muscle microbiopsy specimens, examine myoblast production of reactive oxygen species (ROS) in conditions of anoxia followed by reoxygenation, and assess the effects of horseradish peroxidase (HRP) and myeloperoxidase (MPO) on ROS production. Animals—5 healthy horses (5 to 15 years old). Procedures—Equine skeletal myoblast cultures were derived from 1 or 2 microbiopsy specimens obtained from a triceps brachii muscle of each horse. Cultured myoblasts were exposed to conditions of anoxia followed by reoxygenation or to conditions of normoxia (control cells). Cell production of ROS in the presence or absence of HRP or MPO was assessed by use of a gas chromatography method, after which cells were treated with a 3,3′-diaminobenzidine chromogen solution to detect peroxidase binding. Results—Equine skeletal myoblasts were successfully cultured from microbiopsy specimens. In response to anoxia and reoxygenation, ROS production of myoblasts increased by 71%, compared with that of control cells. When experiments were performed in the presence of HRP or MPO, ROS production in myoblasts exposed to anoxia and reoxygenation was increased by 228% and 183%, respectively, compared with findings for control cells. Chromogen reaction revealed a close adherence of peroxidases to cells, even after several washes. Conclusions and Clinical Relevance—Results indicated that equine skeletal myoblast cultures can be generated from muscle microbiopsy specimens. Anoxia-reoxygenation– treated myoblasts produced ROS, and production was enhanced in the presence of peroxidases. This experimental model could be used to study the damaging effect of exercise on muscles in athletic horses.

International audience | Susceptibility of two French strains of stable flies Stomoxys calcitrans (L.) to six insecticides was assayed, using an exposure technique (1-hour contact) with treated filter papers. Three replicates per insecticide, per concentration (10 concentrations per insecticide), per fly strain (ENVT, Cabanac) and fly category (blood-engorged - non-blood-engorged) were performed using a total of 14,400 adult flies in this trial. The LD50 and LD90 are higher for the blood-engorged flies than the non-blood-engorged flies for both strains of S. calcitrans. The LD90 (mg/m(2)) of the engorged flies for both strains were respectively: cypermethrin (637.9, 54.9), deltamethrin (264.3, 28.1), fenvalerate (2392.5, 125.1), lambda-cyhalothrin (118.2, 41.3), permethrin (353.7, 88.1), and phoxim (194, 226.8). Phoxim, which has not been used in the ENVT for several years, showed the same susceptibility for both strains. The LD90 values obtained for the Cabanac strain (organic farm) were 1 to 4 times lower than the recommended doses of all five pyrethroids. For the ENVT strain (blood-engorged flies), the LD90 was 7.1 and 22.6 times over the recommended doses of both deltamethrin and fenvalerate respectively, which are commonly used insecticides on this site.

No presente trabalho foram analisadas amostras de músculo e gônadas de vieira crus e congelados, oriunda de maricultura de Angra dos Reis, Rio de Janeiro, Brasil. Esta pesquisa teve como finalidade a verificação da eficiência da irradiação, utilizando-se doses de 2kGy e 5kGy. Na Contagem de Bactérias Heterotróficas Aeróbias Mesófilas (CBHAM) observou-se diferença estatisticamente significativa do grupo controle em relação ao grupo irradiado a 5kGy; enquanto que para a Contagem de Bactérias Heterotróficas Aeróbias Psicrotróficas (CBHAP) não se observou nenhuma diferença estatisticamente significativa entre os grupos avaliados. O Número Mais Provável (NMP) de Enterococcus spp. não teve sua presença evidenciada em nenhuma das amostras analisadas. Conclui-se que a irradiação, nesta pesquisa, foi eficiente somente para bactérias mesófilas na dose de 5kGy. | The present article investigated the effectiveness of irradiation process to conservate frozen raw scallops’ muscle and gonads. Samples of this animal were collected from cultivated areas at Angra dos Reis, Rio de Janeiro state, Brazil. The samples were submitted to treatment of 2 and 5 kGy doses. Mesophilic Aerobic Heterotrophic Bacteria Count showed a statistically significant difference from the control group related to the 5kGy one, while for the Heterotrophic Bacteria Aerobic Psychrotrophic Count were not observed any statistically significant difference among the analysed groups. The Most Probable Number of Enterococcus spp. was not evident in any of the samples. It was concluded that irradiation was effective only for mesophilic bacteria using the 5kGy dose in this research.

O objetivo do presente estudo foi avaliar a ocorrência de anticorpos para o vírus da atrite-encefalite caprina (CAEV), Toxoplasma gondii e Neospora caninum e de possíveis associações entre a presença de anticorpos e a ocorrência de problemas reprodutivos em caprinos. Para tanto, foram colhidas amostras sangüíneas de 923 caprinos de ambos os sexos, acima de três meses de idade e oriundos de 17 propriedades leiteiras, de diferentes municípios do estado de São Paulo, Brasil. Os diagnósticos para T. gondii, N. caninum e CAEV foram baseados, respectivamente, na reação de imunofluorescência indireta (RIFI), teste de aglutinação para Neospora (NAT) e a imunodifusão em gel de ágar (IDGA). Um inquérito epidemiológico foi aplicado nas propriedades para obtenção de informações sobre dados reprodutivos (abortamentos, natimortalidade e nascimentos de filhotes fracos e prematuros) e de manejo zoossanitário. As ocorrências de anticorpos foram de 37,81% para CAEV, de 23,62% para T. gondii e de 17,23% para N. caninum. Não houve associação significativa entre a presença de anticorpos anti-CAEV e co-infecção com T. gondii ou N. caninum, sugerindo que o CAEV não predispõe os caprinos à infecção por estes agentes. Entretanto, quando havia, nas fazendas, animais com co-infecção pelo CAEV e T. gondii (p<0,01) ou CAEV e N. caninum (p<0,001) as ocorrências de falhas reprodutivas foram significativamente maiores, sugerindo que a imunossupressão causada pelo CAEV pode predispor os caprinos ao desenvolvimento de sintomas clínicos da toxoplasmose e neosporose, potencializando os riscos da ocorrência de problemas reprodutivos causados por estas enfermidades. | This study aimed at assessing the occurrence of antibodies against the caprine arthritis-encephalitis virus (CAEV), Toxoplasma gondii and Neospora caninum, as well as the associations between the presence of antibodies and the occurrence of reproductive failures in goats. Serum samples were collected from 923 goats of both sexes, over 3 months of age, from 17 dairy farms located in different municipalities of São Paulo State, Brazil. Infections by T. gondii, N. caninum and CAEV were evaluated by indirect methods of diagnosis based on indirect fluorescence antibody test (IFAT), Neospora agglutination test (NAT), and agar gel immunodiffusion (AGID), respectively. A survey was conducted on the farms to obtain information about reproduction dates (abortions, stillbirths and births of weak and premature kids) and zoosanitary management. Antibodies against CAEV, T. gondii and N. caninum was found in 37.81%, 23.62% and 17.23% respectively. There was no significant association between the presence of anti-CAEV antibodies and CAEV/T. gondii or CAEV/N. caninum co-infection, suggesting that CAEV does not predispose goats to infection by these agents. However, when CAEV/T. gondii (p<0.01) or CAEV/N. caninum (p<0.001) co-infection was present, the occurrence of reproductive failures was significantly higher what could indicate that CAEV-induced immunosuppression may predispose goats to develop the clinical symptoms of toxoplasmosis and neosporosis increasing the risks of the reproductive failures.
