Trends of polychlorinated dioxins, polychlorinated furans, and dioxin-like polychlorinated biphenyls in Chinook and Coho salmonid eggs from a Great Lakes tributary

Eggs from mature Chinook (Oncorhynchus tshawytscha) and Coho (Oncorhynchus kisutch) salmon were collected between 2004 and 2014 from the Salmon River fish hatchery in Altmar, New York. The egg samples were analyzed for seventeen polychlorinated dibenzodioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs), as well as four dioxin-like polychlorinated biphenyls (DL-PCBs) using USEPA methods 1613 and 1668. Salmonid eggs were chosen as a tissue of interest since salmon feed at all trophic levels of the food web as they grow, and spawn in a narrow range of ages providing consistent, representative, and temporal samples of contaminant exposure. First-order decay models indicate decreasing trends for all select contaminants in both species, expressed by a toxic equivalence (TEQ) half-life (t₁/₂) of 11 years in Chinook and Coho eggs. No significant statistical difference in contaminant elimination rates were noted between species. TEQ elimination rates for Coho and Chinook eggs were not significantly different (p > 0.05) when compared with published Lake Ontario whole-fish lake trout elimination rates. Our research demonstrates that salmonid eggs are an effective means to assess PCDD, PCDF, and DL-PCB exposures and long-term trends in the Great Lakes.

Ingestion of silver nanoparticles (AgNPs) is inevitable linked to their widespread use in food, medicines and other consumer products. However, their effects on human microbiota at non-lethal concentrations remain poorly understood.In this study, the interactions among 1 μg mL−1 AgNPs, the intestinal microbiota, and the probiotic Bacillus subtilis (BS) were tested using in-vitro batch fermentation models inoculated with human fecal matter.Results from metagenomic investigations revealed that the core bacterial community was not affected by the exposure of AgNPs and BS at the later stage of fermentation, while the proportions of rare species changed drastically with the treatments. Furthermore, shifts in the Firmicutes/Bacteriodetes (F/B) ratios were observed after 24 h with an increase in the relative abundance of Firmicutes species and a decrease in Bacteroidetes in all fermentation cultures. The co-exposure to AgNPs and BS led to the lowest F/B ratio.Fluorescent in-situ hybridization analyses indicated that non-lethal concentration of AgNPs negatively affected the relative percentage of Faecalibacterium prausnitzii and Clostridium coccoides/Eubacterium rectales taxa in the fermentation cultures after 24 h. However, exposure to single and combined treatments of AgNPs and BS did not change the overall diversity of the fecal microflora.Functional differences in cell motility, translation, transport, and xenobiotics degradation occurred in AgNPs-treated fermentation cultures but not in AgNPs+BS-treated samples.Compared to the control samples, treated fecal cultures showed no significant statistical differences in terms of short-chain fatty acids profiles, cytotoxic and genotoxic effects on Caco-2 cell monolayers.Overall, AgNPs did not affect the composition and diversity of the core fecal microflora and its metabolic and toxic profiles. This work indicated a chemopreventive role of probiotic on fecal microflora against AgNPs, which were shown by the decrease of F/B ratio and the unaltered state of some key metabolic pathways.

Thirteen secondary organic aerosol (SOA) tracers of isoprene, monoterpenes and sesquiterpenes were measured for PM₂.₅ aerosols collected at the summit of Mt. Wuyi (1139 m, a.s.l.), to investigate their seasonality and formation mechanism. Concentrations of the isoprene and monoterpene SOA tracers were much higher in summer than those in other seasons. In contrast, β-caryophyllinic acid was found to be the lowest in summer. Concentrations of those BSOA tracers showed a positive correlation with temperature (R² = 0.52–0.70), and a negative correlation with relative humidity (R² = 0.43–0.78). Moreover, thermodynamic model (i.e., ISORROPIA-II) calculation results showed that acidity conditions are favorable for BSOA formation. Robust linear correlations between the BSOA tracers and anthropogenic pollutants such as SO₂ (R² = 0.53–0.7) and NO₂ (R² = 0.37–0.54) were observed for all the samples, suggesting that SO₂ and NOx can enhance BSOA production in the remote mountain area of southeast China, which is related to an acid-catalyzed heterogeneous chemistry. Moreover, we also found a significant correlation between the concentrations of the BSOA tracers and levoglucosan especially for β-caryophyllinic acid, indicating that biomass burning plumes from the distant lowland regions could influence the production of BSOA in the mountain free troposphere. Our results clearly demonstrated that anthropogenic emissions in China could enhance BSOA formation in the distant mountain regions.

Microcystins (MCs) have been shown to be carcinogenic by animal and cellular experiments and found to be associated with the development of human hepatocellular carcinoma (HCC) through epidemiological studies. However, the molecular mechanism of microcystin-LR (MC-LR) induced HCC is still unclear. This study is determined to clarify the role and mechanism of LHX6 in MC-LR-induced hepatocarcinogenesis. Using the previously established MC-LR-induced malignant transformation model in L02 cells, we screened out LHX6, homeobox gene that was significantly changed. We found that LHX6 was significantly down-regulated in MC-LR treated L02 cells and the liver tissue of rats treated for 35 weeks with 10 μg/kg body weight of MC-LR. Expression of LHX6 in human tumor tissue was significantly down-regulated in high MC-LR-exposure group. LHX6 was hypermethylated in MC-LR treated L02 cells and up-regulated after treatment with 10 μM of 5-aza-2′-deoxycytidine. Furthermore, overexpression of LHX6 inhibited proliferation, invasion and migration of malignantly transformed L02 cells in vitro and in vivo, while knockdown of LHX6 resulted in an opposite phenotype. In addition, we found that up-regulation of P53 and Bax resulted in apoptosis, and that down-regulation of CTNNB1 and MMP7 led to migration of MC-LR treated L02 cells. Blockade of P53 and CTNNB1 by its inhibitor significantly diminished the effect of LHX6. These genes were working together during the process of MC-LR-induced hepatocarcinogenesis. Our study demonstrated for the first time that LHX6 gene expression is regulated by DNA methylation and can inhibit the proliferation, invasion and migration through Wnt/β-catenin and P53 signaling pathways during the MC-LR-induced hepatocarcinogenesis. This result may suggest that LHX6 gene can be used as a potential target gene and a biomarker for liver cancer treatment.

Cadmium (Cd) contamination of the soil is one of the most serious environmental problems of agricultural production. Phytoremediation has attracted increasing attention because it can safely remove the soil contaminates via plant uptake, accumulations and plant harvesting. However, the high Cd toxicity to plant tissues and treatment of the large amount of hazardous plant residues from phytoremediation have limited its commercial implementation. Here we show that the leaves of the tall fescue (Festuca arundinacea) can excrete Cd out to avoid Cd toxicity in plant tissues. Cd specific fluorescence spectroscopy with laser confocal scanning microscope, screening electron microscope with energy dispersive spectroscopy and guttation fluids analysis confirmed that leaf hydathodes were the pathway of Cd excretion in tall fescue. Element analysis showed that Cd was preferentially excreted out when compared to the ion nutrients. The amount of leaf Cd excretion was linearly increased in response to the Cd stress period. The phytoremediation efficiency was evaluated to remove 14.4% of soil Cd annually by the leaf Cd excretion in our experimental system. These findings indicate that a novel strategy of Cd phytoexcretion based on washing-off and collection of leaf surface Cd is feasible to avoid Cd toxic in plant tissues and the high treatment cost of hazardous plant residues.

Particulate matter (PM) deposited on canopy surfaces could be washed off and carried in throughfall to the ground. This would help plants recapture airborne PM on their canopy surfaces and then develop a PM purification capacity. Sixteen commonly greening plant species in north China (including 13 arbor species and 3 shrub species) were selected to investigate the washing process of plant-deposited PM during precipitation events. We measured the PM wash-off mass in throughfall under canopies of 16 plant species and in atmospheric precipitation during 14 precipitation events through field positioning experiments in 2015, compared the seasonal changes and species differences in PM wash-off mass, and discussed the predominant factors resulting in the variation. The results showed that plant-deposited PM was largely washed off by precipitation. The average wash-off mass of total suspended particulate (TSP) in throughfall was 1.3 times higher than that in precipitation, at 18.3 ± 0.7 kg hm−2 and 7.9 ± 0.9 kg hm−2, respectively. There were significant seasonal differences in TSP wash-off mass. The value was higher in summer at 22.3 ± 1.0 kg hm−2, followed by that of winter (10.8 ± 0.6 kg hm−2) and spring (8.9 ± 1.0 kg hm−2). TSP wash-off mass in throughfall greatly varied among plant species (F = 9.542, n = 627, p < 0.001). Of the 16 selected species, Platanus acerifolia (38.0 ± 5.8 kg hm−2) showed the largest difference from that of Liriodendron chinese (8.9 ± 0.6 kg hm−2) (n = 80, p < 0.001). PM wash-off mass of different particle sizes in throughfall increased with the increase of event-based precipitation. This study enhanced the quantitative understanding of plant-deposited PM washed-off by natural precipitation among plant species and seasons. The results could provide significant guidelines for the selection and allocation of plant species to improve the PM retention capacity of urban greening plants.

The occurrence of environmental antibiotic resistance genes (ARGs) are often attributed to selective pressure from antibiotics from point source pollution. However, the potential effects of natural production of antibiotics, environmental factors, and anthropogenic pressure on the development and spread of ARGs have not been fully investigated. This study evaluated the occurrence and distribution of erythromycin resistance methylase (erm) genes in urban soils. The ermA, ermB, ermC, ermD, ermF, ermG, ermT, and ermY genes were detected with detection frequencies ranging from 20% to 80% and abundances ranging between 5.95 × 101 and 6.94 × 106 copies g−1 dw soil. Both polyketide synthase (PKS) type I and type II biosynthesis genes—which are responsible for biosynthesis of polyketides, such as erythromycin—were detected in all soil samples with a range between 5.77 × 102 and 9.39 × 106 copies g−1 dw soil. The abundances of PKS genes were significantly correlated with 16S rRNA genes (r = 0.487 to 0.741, p < 0.001) and absolute abundances of ermB, ermC, ermD, ermG, and ermY (r = 0.302–0.490, p < 0.05), suggesting that the wide occurrence of ARGs in soils could be potentially driven by naturally produced antibiotics. Erythromycin was strongly correlated with ermB, ermC, ermF and ermY genes (r = 0.462 to 0.667, p < 0.05), but no significant correlation was observed between macrolides and PKS genes, suggesting other environmental factors may have contributed to detected macrolides. The fact that erm gene presented higher extent of variability than PKS genes in different land use types suggests that anthropogenic activity might also influence the occurrence of erm genes in urban soils.

Moss is usually as an initial colonizer in alpine glacier retreated regions. We hypothesized that moss can significantly facilitate the toxic metals accumulation in alpine ecosystems based on its strong ability of absorption and the role in soil development. Hence, we investigated the trace element pool sizes and enrichment factors, especially for mercury (Hg) by using the Hg isotopic compositions to determine the source contributions in a moss-dominated ecosystem over glacial erratic in Eastern Tibetan Plateau. Results show that Hg, lead (Pb) and cadmium (Cd) are highly enriched in organic soils. Specifically, Cd concentration is 5–20 times higher than the safety limit of the acid soil (pH ≤ 5.5) in China. Atmospheric depositions dominantly contribute to the Pb and Cd sources in organic soils, and followed by the moraine particles influences. The lowering pH in organic soils increasing with glacial retreated time results in the desorption of Cd in organic soils. Atmospheric Hg⁰ uptake by moss predominantly contributes to the Hg sources in organic soils. The average Pb accumulation rate over last 125-year is about 5.6 ± 1.0 mg m⁻² yr⁻¹, and for Cd is 0.4 ± 0.1 mg m⁻² yr⁻¹, and for Hg⁰ is 27.6 ± 3.2 μg m⁻² yr⁻¹. These elevated accumulation rates are caused by the high moss biomass and elevated atmospheric Hg, Pb and Cd pollution levels in China and neighbouring regions. Our study indicates that the moss not only as the bioindicator, but also plays an important role in the hazardous metal biogeochemical cycling in alpine regions.

In the family of alkylphenolic compounds, 4-tertiary-Octylphenol (4-t-OP) is extensively used in many products. In animal and in vitro studies, 4-t-OP exposure has been linked to cardiovascular disease (CVD) risk factors; however, there are no previous human epidemiological studies. In this study, 886 subjects were recruited from a cohort of Taiwanese adolescents and young adults to study the relationship between serum levels of 4-t-OP, CVD risk factors, and common carotid artery intima-media thickness (CIMT). The geometric mean (SD) 4-t-OP concentration was 32.52 (1.71) ng/mL. We found that serum levels of 4-t-OP were negatively associated with markers of glucose homeostasis (insulin, homeostasis model assessment of insulin resistance (HOMA-IR) and homeostasis model assessment of β-cell function (HOMA-β)), z score of body mass index (BMI z score) and CIMT but were positively associated with lipid profiles (high density lipoprotein cholesterol (HDL-C), Apolipoprotein A1). A one-unit elevation in natural log-transformed 4-t-OP (ng/mL) was negatively correlated with CIMT (mm) (β = −0.029, SE = 0.003, P < 0.001) in multiple linear regression analyses. The relationship between 4-t-OP and CIMT remained the same in all subgroups or if bisphenol A (BPA) was considered a covariate. In this study, we observed that higher levels of 4-t-OP levels were negatively correlated with markers of glucose homeostasis, BMI z score, and CIMT; positively correlated with lipid profiles (HDL-C and apolipoprotein A) in this cohort. Future research on exposure to 4-t-OP and CVD risk factors is warranted.

Denitrification is considered as the dominant nitrogen (N) removing pathway, however, anaerobic oxidation of ammonium (anammox) also plays a significant part in N loss in agricultural ecosystems. Large N inputs into agricultural soils may stimulate the growth of anammox bacteria, resulting in high activity and diversity of anammox bacteria and subsequent more N loss. In some specific niches, like oxic-anoxic interface, three processes, nitrification, anammox and denitrification couple with each other, and significant anammox reaction could be observed. Soil parameters like pH, dissolved oxygen, salinity, oxidation-reduction potential (ORP), and substrate concentrations impact the anammox process. Here we summarize the current knowledge on anammox activity and contribution to N loss, abundance and diversity of anammox bacteria, factors affecting anammox, and the relationship between anammox and other N loss pathways in agricultural soils. We propose that more investigations are required for (1) the role of anammox to N loss with different agricultural management strategies; (2) microscale research on the coupling of nitrification-anammox-denitrification, that might be a very complex process but ideal model for further studies responsible for N cycling in terrestrial ecosystems; and (3) new methods to estimate differential contributions of anammox, codenitrification and denitrification in total N loss in agricultural ecosystems. New research will provide much needed information to quantify the contribution of anammox in N loss from soils at landscape, ecosystem and global scales.
