Maternal urinary levels of glyphosate during pregnancy and anogenital distance in newborns in a US multicenter pregnancy cohort

Human exposure to glyphosate has become ubiquitous because of its increasing agricultural use. Recent studies suggest endocrine disrupting effects of glyphosate. Specifically, in our work in rodents, low-dose early-life exposure to Roundup® (glyphosate-based herbicide) lengthened anogenital distance (AGD) in male and female offspring. AGD is a marker of the prenatal hormone milieu in rodents and humans. The relationship between glyphosate exposure and AGD has not been studied in humans. We conducted a pilot study in 94 mother-infant pairs (45 female and 49 male) from The Infant Development and the Environment Study (TIDES). For each infant, two AGD measurements were collected after birth; the anopenile (AGD-AP) and anoscrotal (AGD-AS) distances for males, and anoclitoral (AGD-AC) and anofourchette distances (AGD-AF) for females. We measured levels of glyphosate and its degradation product aminomethylphosphonic acid (AMPA) in 2nd trimester maternal urine samples using ultra-high-performance liquid chromatography-tandem mass spectrometry. We assessed the relationship between exposure and AGD using sex-stratified multivariable linear regression models. Glyphosate and AMPA were detected in 95% and 93% of the samples (median 0.22 ng/mL and 0.14 ng/mL, respectively). Their concentrations were moderately correlated (r = 0.55, p = 5.7 × 10⁻⁹). In female infants, high maternal urinary glyphosate (above the median) was associated with longer AGD-AC (β = 1.48, 95%CI (−0.01, 3.0), p = 0.05), but this was not significant after covariate adjustment. Increased AMPA was associated with longer AGD-AF (β = 1.96, 95%CI (0.44, 3.5), p = 0.01) after adjusting for infant size and age at AGD exam. No associations were detected in male offspring. These preliminary findings partially reproduce our previous results in rodents and suggest that glyphosate is a sex-specific endocrine disruptor with androgenic effects in humans. Given the increasing glyphosate exposures in the US population, larger studies should evaluate potential developmental effects on endocrine and reproductive systems.

Cyanobacterial blooms have considerable effects on lacustrine microbial communities. The current study explored the temporal pattern of sedimentary archaea and bacteria during cyanobacterial bloom in a eutrophic lake. With the sampling period divided into bloom phase, interval phase and end phase according to the variation of physicochemical parameters, the structures and functions of both kingdoms presented a significant difference among phases. Bloom phases could be characterized with the lowest diversity and up-regulated functions in biodegradation of cyanobacterial metabolites driven by bacteria. Archaeal community showed an increased metabolic function during interval phases, including active methanogenesis sensitive to carbon input. The highest diversity and an enrichment of hub genera in microbial network were both observed in end phase, allowing for closer cooperation among groups involved in cyanobacteria-derived organic matter transformation. Although the archaeal community was less variable or diverse than bacteria, methanogenic functions dramatically fluctuated with cyanobacterial dynamics. And microbial groups related to methane cycling played an important role in microbial network. The results provided new insights into temporal dynamics of lacustrine microbial communities and microbial co-occurrence, and highlighted the significant ecological role of methane cycling-related microbes in lake sediments under the influence of cyanobacterial blooms.

Despite a growing focus on anthropogenic toxic metal pollution in urban environments, few studies have addressed the problem of quantification when more than two pollution sources are likely present, particularly within complex urban settings in the United States (U.S.). In this study, we utilize the MixSIAR package in R for source apportionment based on Pb isotopic signatures in lichen and road sediment in two urban-industrial centers in SW Ohio (OH). We show that ranges of pollutant contributions are more useful than only visualizing mean or raw values of source apportionment, because this avoids overinterpretation of data when certain sources have a large range of uncertainty. We point out both the dominance of industrial pollution as well as the legacy of leaded gasoline pollution in typical mid-sized U.S. cities, which is evident in both road sediment and lichens. Leaded gasoline contribution to Pb in Middletown, OH lichens mostly vary between ~10 and 25%, while in Hamilton, OH the contribution to lichens and road sediment tends to be relatively negligible except for two road sediment samples and one lichen sample, where median contributions are ~20–30%. Industrial combustion pollution source contributions vary between ~25 and 75% in Hamilton, and ~50–100% in Middletown, OH. Furthermore, comparing pollution sources in lichens to modern particulate matter can provide a record of how pollutant sources change over time, such as our traffic lichen (Sample Li-9) plotting closer to leaded gasoline on a bivariate mixing diagram than modern traffic particulate matter, or our coke plant lichen containing slightly less Pb contribution from industrial combustion sources relative to modern coke plant particulate matter. Lastly, when applicable, multi-source mixing models should be complimented in future studies with additional isotopic source tracers such as Cu, Zn, Nd, and Os to further elucidate unique sources of metal pollutants in addition to Pb.

Karst ecosystems make an important contribution to the global carbon cycle, in which carbon-fixing microorganisms play a vital role. However, the healthy functioning of karst ecosystems is threatened because pollutants easily diffuse and spread through them due to their strong hydraulic connectivity. The microbiome of a karst river contaminated with antibiotics was studied. Through co-occurrence network analysis, six ecological clusters (MOD 1–MOD 6) with different distribution characteristics were determined, of which four were significantly correlated with antibiotics. The carbon fixation pathways in different ecological clusters were varied, and the dominant hydroxypropionate-hydroxybutyrate cycle and reductive acetyl-CoA pathway were negatively and positively correlated with antibiotics, respectively. Long-term antibiotic contamination altered the selection of carbonic anhydrase (CA) encoding genes in some of the CA-producing mineralization microorganisms. The selection of different carbon fixation pathways is a possible strategy for the microbial community to compensate for the adaptation costs associated with the pressure of antibiotics contamination and emergence of antibiotics resistance. Bayesian network analysis revealed that some carbon sequestration functions (such as β-CA and reductive acetyl-CoA pathway) surpassed certain antibiotic resistance genes in the regulation of environmental factors and microbial networks. An ecological cluster (MOD5) that possibly homologous to antibiotic contamination was the final node of the microbial community in karst river, which indicated that ecological clusters were not only selected by antibiotics, but were also regulated by multiple environmental factors in the karst river system. The carbon sequestration pathway was more directly reflected in the abundance of ecological groups than in the influence of CA. This study provides new insights into the feedback effect of karst system on typical pollutants generated from human activities.

Extreme temperatures from marine heatwaves (MHWs) and pollution are dominant stressors in tropical marine ecosystems. However, we know little about the role of transgenerational effects of metals and MHWs in shaping the offspring's vulnerability to these stressors. We addressed this fundamental knowledge gap by exposing the planktonic copepod Pseudodiaptomus incisus to copper (Cu: control, 15 and 60 μg L⁻¹) under 2 temperatures (30 and a simulated marine heatwave at 34 °C) in the first generation (F1) and 16 treatments in F2: offspring from each of 4 F1 conditions (control or 15 μg Cu L⁻¹ × 30 or 34 °C) was reared in 4 F2 conditions (control or 15 μg Cu L⁻¹ × 30 or 34 °C). We assessed changes in copepod performance, particularly survival, adult size, grazing, and reproduction. In F1, Cu or marine heatwave (MHW) exposures reduced all fitness traits of F1; the effects were particularly strong when both stressors were present. Transgenerational effects of Cu or MHW also strongly reduced F2 performance. Direct Cu and MHW effects on the offspring were further strengthened by transgenerational effects, resulting in more substantial reductions in F2 performance when both generations were exposed to these stressors. As copepods are major food resources for corals, shrimps, or fish larvae and juveniles, strong transgenerational and direct effects of Cu and MHW can have a cascading effect on entire coastal food webs. These results highlight the importance of considering the interaction of transgenerational and direct effects of multiple stressors, particularly relevant for short-lived organisms in tropical marine ecosystems.

Evidence of the effects of long-term household air pollution (HAP) on human mental health is limited. This study aimed to explore the longitudinal relationship between long-term household air pollution exposure from solid fuel use and depression based on nationally representative follow-up dataset. A total of 7005 middle- and old-age adults from the latest four waves (2011, 2013, 2015, and 2018) of China Health and Retirement Longitudinal Study (CHARLS) were involved. The Center for Epidemiologic Studies Depression scale (CES-D 10) was used to measure depressive symptoms and individuals who got more than 12 points were considered to have depression symptoms. We conducted Cox proportional hazards regression models to examine the association between household air pollution and depression in overall population, and subgroup stratified by socio-demographic factors, lifestyle behaviors, chronic diseases, and residential environments. We found long-term household air pollution exposure from solid fuel use was significantly associated with higher depression risk among Chinese older adults (HR 1.27, 95% CI 1.14–1.42 in heating; 1.26, 1.13–1.40 in cooking). Longer duration of household air pollution exposure (1.47, 1.28–1.68 in heating; 1.36, 1.19–1.56 in cooking) and household air pollution from crop residue/wood burning (1.66, 1.41–1.94 in heating; 1.37, 1.23–1.53 in cooking) was correlated with higher depression risk. For subgroups analysis, the effect of household air pollution from solid fuel on depression varied. Compared with those who using clean fuel, older adults living in small size houses or houses with small number rooms had increasing depression risks if they used solid fuel for heating or cooking. Our findings indicate long-term household air pollution exposure from solid fuel use is associated with higher depression risk. Reducing household air pollution by restricting solid fuel use can be effective ways to prevent depression for Chinese older adults and decrease related public health burden.

Beaches are key attractions for tourism and recreation, and considerable effort is made to keep beaches clean, yet many beaches still have substantial litter loads. Lasting solutions to reduce the amounts of marine litter require an understanding of litter sources. We collected bottles and other single-use containers at 32 sites around the South African coast to infer their sources based on their age and country of manufacture. Bottle densities varied greatly among beaches (8–450 bottles·km⁻¹), depending on proximity to local urban centres and beach cleaning frequency. Most bottles were plastic, despite well-developed recycling initiatives for PET and HDPE bottles in South Africa. Street litter was dominated by bottles made in South Africa (99%), but foreign-manufactured bottles comprised up to 74% of bottles at some beaches, with an increase from urban (4%) through semi-urban (24%) to remote beaches (45%). Most foreign bottles were PET drink bottles from China and other Asian countries, followed by South America and Europe, with little regional variation in the contribution from these sources. This fact, coupled with their recent manufacture dates (mainly <2 years old), indicates that most foreign PET drink bottles are dumped illegally from ships. By comparison, foreign HDPE bottles were more common along the southeast coast of South Africa than along the west coast, consistent with many of these bottles arriving by long-distance drift across the Indian Ocean from southeast Asia. The most common country of origin for these bottles was Indonesia, and most newly-arrived HDPE bottles were 4–6 years old. To tackle beach litter in South Africa we need to greatly reduce plastic leakage from land-based sources, both locally and in southeast Asia, as well as improve measures to prevent the illegal dumping of plastics and other persistent wastes from ships.

Understanding the movement of polycyclic aromatic hydrocarbons (PAHs) from emission sources to sediments is important for achieving long-term pollution control of PAHs in sediments. In this study, by exploring the correlation of individual PAHs concentrations (CPAHₛ) in Taihu Lake sediments reported in the past twenty years with their annual emissions (EPAHₛ) in the lake region, it was observed that mean concentrations of PAHs with low logKₒw (i.e., logKₒw≤4.00) in Taihu Lake sediments were correlated best with their emissions without lagging between the sediment sampling time and the PAHs emitting time. However, for PAHs with middle logKₒw (i.e., 4.00<logKₒw≤4.57) or high logKₒw (i.e., logKₒw>4.57), their mean concentrations in sediments were correlated best with the emissions of PAHs emitted 1 or 2 years before the sediment sampling time. The longer lagging time of PAHs with middle or high logKₒw from emission sources to lake sediments could be attributed to their retardation in soils and river sediments around the lake. Moreover, the retardation in soils and river sediments is dependent on PAHs logKₒw and degradation half-life, indicating the dependence of PAHs concentration in sediments on their environmental behaviors, including sorption and degradation. Kₒw dependent movement and the time lagging observed in Taihu Lake for PAHs from emission sources to sediments could be valuable for developing measures to control PAHs, especially for congeners with high logKₒw.

The crop-livestock system is responsible for a large proportion of global reactive nitrogen (Nr) losses, especially from China. There are diverse livestock systems with contrasting differences in feed, livestock and manure management. However, it is not yet well understood which factors greatly impact on the nitrogen (N) budgets and losses of each system. In this study, we systematically evaluated the N budgets of the crop-livestock production system from 1980 to 2050 in China by identifying the differences of 20 distinct livestock systems. During 1980 to 2010, the total N flow through the crop-livestock system increased from 21.4 to 49.7 Tg, with large variations in different input/output pathways, due to the strong livestock transitions of production towards to a monogastric and landless industrial system. Different systems contributed differently to the total N budgets in 2010. For example, the landless industrial system contributed 67% of livestock product N output, but accounted for 80% of total mineral N fertilizer use and feed N imports by the whole crop-livestock system. The mixed system had the highest rate of N use efficiency at system level due to high dependence on recycled N. N losses were diversely distributed by different systems, with the mixed ruminant system responsible for the majority of NH₃–N emission in livestock production, and the grazing ruminant system dominant in NO₃–N losses in feed production. The total N entering the crop-livestock system is estimated to be 53.9 Tg with total N losses of 41.3 Tg in 2050 under a business-as-usual scenario. However, this amount could be significantly decreased through combined measures that indicate a considerable potential for future improvements. Overall, our results provide new insights into N use and the management of livestock production.

Antibiotic resistance in aquatic ecosystems presents an environmental health issue worldwide. Urban recipient water quality is susceptible to effluent discharges with antibiotic resistance contaminants and needs to be protected, particularly for those as sources of drinking water production. Knowledge on aquatic resistome profiles in downstream of wastewater treatment plants allows a better understanding of the extent to which antibiotic resistance contaminants emerge and spread in recipient waters, but such information remains very limited in Sweden. The key objective of this study was to determine the resistome profiles of numerous antibiotic resistance genes (ARGs), mobile genetic elements (MGEs) and other genes in urban recipient water systems connected to Sweden's major drinking water reservoir. This was achieved through analysis of surface water samples for 296 genes using high-throughput quantitative PCR arrays. A total of 167 genes were detected in at least one of the samples, including 150 ARGs conferring resistance to 11 classes of antibiotics, 7 integrase MGEs and 9 other genes. There was a spatial difference in the resistome profiles with the greatest average relative abundance of resistance genes observed in the water body of Västerås followed by Uppsala, Stockholm and Eskilstuna, as similar to the general pattern of the antibiotic sales for these regions. ARGs against β-lactams and sulfonamides showed the highest average relative abundance in the studied water bodies, while vancomycin resistance genes were only found in the Uppsala water environment. Generally, the recipient water bodies were detected with higher numbers of genes and greater relative abundances as compared to the upstream sites. Anthropogenic pollution, i.e., wastewater discharge, in the recipient water was also reflected by the finding of intI, sul1 and crAssphage. Overall, this study provided the first quantitative assessment of aquatic environmental resistomes in Sweden, highlighting the widespread of antibiotic resistance contaminants in urban recipient waters.
