Biotransformation in the zebrafish embryo –temporal gene transcription changes of cytochrome P450 enzymes and internal exposure dynamics of the AhR binding xenobiotic benz[a]anthracene Полный текст

Not much is known about the biotransformation capability of zebrafish (Danio rerio) embryos. For understanding possible toxicity differences to adult fish, it might be crucial to understand the biotransformation of chemicals in zebrafish embryos i.e. as part of toxicokinetics.The biotransformation capabilities were analysed for two different stages of zebrafish embryos in conjunction with the internal concentrations of a xenobiotic. Zebrafish embryos of the late cleavage/early blastula period (2–26 hpf) and the early pharyngula period (26–50 hpf) were exposed for 24 h to the AhR binding compound benz[a]anthracene (BaA). Time dependent changes in cyp transcription (cyp1a, cyp1b1, cyp1c1 and cyp1c2) as well as concentration & time-dependent courses of BaA in the fish embryo and the exposure medium were analysed. Additionally, the CYP mediated formation of biotransformation products was investigated.We found correlations between transcriptional responses and the internal concentration for both exposure types. These correlations were depending on the start of the exposure i.e. the age of the exposed embryo. While no significant induction of the examined gene transcripts was observed in the first 12 h of exposure beginning in the blastula period a correlation was apparent when exposure started later i.e. in the pharyngula period. A significant induction of cyp1a was detected already after 1.5 h of BaA exposure. Gene transcripts for cyp1b1, cyp1c1 and cyp1c2 showed expressions distinctly different from cyp1a and were, in general, less inducible by BaA in both exposure windows. The toxicokinetic analysis showed that the biotransformation capability was fivefold higher in the older fish embryos. Biotransformation products of phase I reactions were found between 32 hpf and 50 hpf and were tentatively identified as benz[a]anthracene-phenol and benz[a]anthracene-dihydrodiol-epoxide.In conclusion, not only duration but also onset of exposure in relation to the developmental stage of zebrafish embryos is important in the analysis and interpretation of effects due to different biotransformation capabilities.

Silver nanoparticles (AgNPs) are highly toxic to aquatic organisms, however, there is no consensus whether the toxicity is caused solely by released Ag-ions or also by reactive oxygen species (ROS). Here, the effects of protein-coated AgNPs (14.6 nm, Collargol) were studied on viability, oxidative stress and gene expression levels in wild type strains (CU427 and CU428) of unicellular ciliate Tetrahymena thermophila. Viability-based 24 h EC50 values of AgNPs were relatively high and significantly different for the two strains: ∼100 mg/L and ∼75 mg/L for CU427 and CU428, respectively. Similarly, the expression profiles of oxidative stress (OS) related genes in the two strains were different. However, even though some OS related genes were overexpressed in AgNP-exposed ciliates, intracellular ROS level was not elevated, possibly due to efficient cellular antioxidant defence mechanisms. Compared to OS related genes, metallothionein genes were upregulated at a considerably higher level (14 versus 5000-fold) suggesting that Ag-ion mediated toxicity mechanism of AgNPs prevailed over OS related pathway. Also, comparison between Ag-ions released from AgNPs at EC50 concentration and the respective EC50 values of AgNO3 indicated that Ag-ions played a major role in the toxicity of AgNPs in T. thermophila. The study highlights the importance of combining physiological assays with gene expression analysis in elucidating the mechanisms of action of NPs to reveal subtle cellular responses that may not be detectable in bioassays. In addition, our data filled the gaps on the toxicity of AgNPs for environmentally relevant and abundant organisms. The parallel study of two wild type strains allowed us to draw conclusions on strain to strain variability in susceptibility to AgNPs.

The 2010 Deepwater Horizon (DWH) oil spill released millions of barrels of oil and dispersant into the Gulf of Mexico. The timing of the spill coincided with the spawning season of Crassostrea virginica. Consequently, gametes released in the water were likely exposed to oil and dispersant. This study aimed to (i) evaluate the cellular effects of acute exposure of spermatozoa and oocytes to surface slick oil, dispersed mechanically (HEWAF) and chemically (CEWAF), using flow-cytometric (FCM) analyses, and (ii) determine whether the observed cellular effects relate to impairments of fertilization and embryogenesis of gametes exposed to the same concentrations of CEWAF and HEWAF. Following a 30-min exposure, the number of spermatozoa and their viability were reduced due to a physical action of oil droplets (HEWAF) and a toxic action of CEWAF respectively. Additionally, reactive oxygen species (ROS) production in exposed oocytes tended to increase with increasing oil concentrations suggesting that exposure to dispersed oil resulted in an oxidative stress. The decrease in fertilization success (1-h), larval survival (24-h) and increase in abnormalities (6-h and 24-h) may be partly related to altered cellular characteristics. FCM assays are a good predictor of sublethal effects especially on fertilization success. These data suggest that oil/dispersant are cytotoxic to gametes, which may affect negatively the reproduction success and early development of oysters.

The concentration and spatial distribution of microplastics in the Bay of Brest (Brittany, France) was investigated in two surveys. Surface water and sediment were sampled at nine locations in areas characterized by contrasting anthropic pressures, riverine influences or water mixing. Microplastics were categorized by their polymer type and size class. Microplastic contamination in surface water and sediment was dominated by polyethylene fragments (PE, 53–67%) followed by polypropylene (PP, 16–30%) and polystyrene (PS, 16–17%) microparticles. The presence of buoyant microplastics (PE, PP and PS) in sediment suggests the existence of physical and/or biological processes leading to vertical transfer of lightweight microplastics in the bay. In sediment (upper 5 cm), the percentage of particles identified by Raman micro-spectroscopy was lower (41%) than in surface water (79%) and may explain the apparent low concentration observed in this matrix (0.97 ± 2.08 MP kg−1 dry sediment). Mean microplastic concentration was 0.24 ± 0.35 MP m−3 in surface water. We suggest that the observed spatial MP distribution is related to proximity to urbanized areas and to hydrodynamics in the bay. A particle dispersal model was used to study the influence of hydrodynamics on surface microplastic distribution. The outputs of the model showed the presence of a transitional convergence zone in the centre of the bay during flood tide, where floating debris coming from the northern and southern parts of the bay tends to accumulate before being expelled from the bay. Further modelling work and observations integrating (i) the complex vertical motion of microplastics, and (ii) their point sources is required to better understand the fate of microplastics in such a complex coastal ecosystem.

Urban soil contamination with trace metals is a major obstacle to the development of urban agriculture as crops grown in urban gardens are prone to accumulate trace metals up to toxic levels for human consumption.Phytoextraction is considered as a potentially cost-effective alternative to conventional methods such as excavation. Field trials of phytoextraction with Noccaea caerulescens were conducted on urban soils contaminated with Cd, Cu, Pb, and Zn (respectively around 2, 150–200, 400–500, and 400–700 μg g−1 of dry soil). Metallicolous (Ganges population) and non-metallicolous (NMET) populations were compared for biomass production and trace metal uptake. Moreover, we tested the effect of compost and fertilizer addition. Maximal biomass of 5 t ha−1 was obtained with NMET populations on some plots. Compared to Ganges— the high Cd-accumulating ecotype from South of France often used in phytoextraction trials— NMET populations have an advantage for biomass production and for Zn accumulation, with an average Zn uptake of 2.5 times higher. The addition of compost seems detrimental due to metal immobilization in the soil with little or no effect on plant growth. In addition to differences between populations, variations of growth and metal accumulation were mostly explained by soil Cd and Zn concentrations and texture.Our field trials confirm the potential of using N. caerulescens for both Cd and Zn remediation of moderately contaminated soils—with uptake values of up to 200 g Cd ha−1 and 47 kg Zn ha−1—and show the interest of selecting the adequate population according to the targeted metal.

The micronucleus frequency in root tip cells was positively correlated and the number of mitotic cells negatively, to the extractability of Zn in soils and to the concentration of Zn in secondary roots.Exposure to pure biochar caused a lower production of roots than most soil substrates, but led to the lowest number of observed micronuclei. In conclusion, biochar amendments can reduce the genotoxicity associated with the presence of metallic contaminants in soils, thereby potentially improving plant growth.

Total mercury (T-Hg) and cadmium (Cd) were measured in twenty species of fish to study their bioaccumulation patterns and trophodynamics in the Augusta Bay food web. Adult and juvenile fish were caught in 2012 in Priolo Bay, south of the Augusta harbour (Central Mediterranean Sea), which is known for the high trace element and polycyclic aromatic hydrocarbon contamination level. T-Hg concentration was found to significantly increase along δ15N and from pelagic to benthic sedentary fish, revealing a marked influence of trophic position and habitat use (sensu Harmelin 1987) on T-Hg accumulation within ichthyofauna. Cd showed the opposite pattern, in line with the higher trace element (TE) excretion rates of high trophic level fish and the lower level of Cd environmental contamination.Trophic pathways were first characterised in the Priolo Bay food web using carbon and nitrogen stable isotopes (δ13C, δ15N) and a single main trophic pathway characterised the Priolo Bay food web. Biomagnification was then assessed, including basal sources (surface sediment, macroalgae), zooplankton, benthic invertebrates and fish. T-Hg and Cd were found to biomagnify and biodilute respectively based on the significant linear regressions between log[T-Hg] and log[Cd] vs. δ15N of sources and consumers and the trophic magnification factors (TMFs) of 1.22 and 0.83 respectively. Interestingly, different Cd behaviour was found considering only the benthic pathway which leads to the predatory gastropod Hexaplex trunculus. The positive slope and the higher TMF indicated active biomagnification in this benthic food web due to the high bioaccumulation efficiency of this benthic predator. Our findings provide new evidences about the role of Priolo sediments as a sources of pollutants for the food web, representing a threat to fish and, by domino effect, to humans.

The production and use of pentachlorophenol (PCP) was recently prohibited/restricted by the Stockholm Convention on persistent organic pollutants (POPs), but environmental data are few and of varying quality. We here present the first extensive dataset of the continent-wide (Eurasia and Canada) occurrence of PCP and its methylation product pentachloroanisole (PCA) in the environment, specifically in pine needles. The highest concentrations of PCP were found close to expected point sources, while PCA chiefly shows a northern and/or coastal distribution not correlating with PCP distribution. Although long-range transport and environmental methylation of PCP or formation from other precursors cannot be excluded, the distribution patterns suggest that such processes may not be the only source of PCA to remote regions and unknown sources should be sought. We suggest that natural sources, e.g., chlorination of organic matter in Boreal forest soils enhanced by chloride deposition from marine sources, should be investigated as a possible partial explanation of the observed distributions. The results show that neither PCA nor total PCP (ΣPCP = PCP + PCA) should be used to approximate the concentrations of PCP; PCP and PCA must be determined and quantified separately to understand their occurrence and fate in the environment. The background work shows that the accumulation of airborne POPs in plants is a complex process. The variations in life cycles and physiological adaptations have to be taken into account when using plants to evaluate the concentrations of POPs in remote areas.

Cold weather was estimated to account for more than half of weather-related deaths in the U.S. during 2006–2010. Studies have shown that cold-related excessive mortality is especially relevant with decreasing latitude or in regions with mild winter. However, only limited studies have been conducted in the southern U.S. The purpose of our study is to examine impacts of cold weather on mortality in 12 major Texas Metropolitan Areas (MSAs) for the 22-year period, 1990–2011. Our study used a two-stage approach to examine the cold-mortality association. We first applied distributed lag non-linear models (DLNM) to 12 major MSAs to estimate cold effects for each area. A random effects meta-analysis was then used to estimate pooled effects. Age-stratified and cause-specific mortalities were modeled separately for each MSA. Most of the MSAs were associated with an increased risk in mortality ranging from 0.1% to 5.0% with a 1 °C decrease in temperature below the cold thresholds. Higher increased mortality risks were generally observed in MSAs with higher average daily mean temperatures and lower latitudes. Pooled effect estimate was 1.58% (95% Confidence Interval (CI) [0.81, 2.37]) increase in all-cause mortality risk with a 1 °C decrease in temperature. Cold wave effects in Texas were also examined, and several MSAs along the Texas Gulf Coast showed statistically significant cold wave-mortality associations. Effects of cold on all-cause mortality were highest among people over 75 years old (1.86%, 95% CI [1.09, 2.63]). Pooled estimates for cause-specific mortality were strongest in myocardial infarction (4.30%, 95% CI [1.18, 7.51]), followed by respiratory diseases (3.17%, 95% CI [0.26, 6.17]) and ischemic heart diseases (2.54%, 95% CI [1.08, 4.02]). In conclusion, cold weather generally increases mortality risk significantly in Texas, and the cold effects vary with MSAs, age groups, and cause-specific deaths.

Degradation of 2,4,6-trichloropenol (TCP) with peroxymonosulfate (PMS) catalyzed by iron porphyrin tetrasulfonate ([FePTS)] was investigated in an 8-to-1 (v/v) CH3OH-H2O mixture. Typical reaction medium contained a 4.00 mL methanol solution of TCP (0.100 mmol), a 0.50 mL aqueous solution of catalyst (5.0 × 10⁻⁴ mmol), and 0.100 mmol PMS (as 0.031 g of Oxone). The reaction was performed at ambient temperature. The conversion of TCP was 74% in 30 min and 80% in 6 h when the catalyst was [FePTS]. Amberlite IRA-900 supported [FePTS] catalyst was also prepared. In the recycling experiments the homogeneous [FePTS] lost its activity after the first cycle, while [FePTS]-Amberlite IRA 900 maintained its activity for the first 2 cycles. After the second cycle, the conversion of TCP dropped to <10% for Amberlite IRA-900 supported [FePTS] catalyst. The degradation of TCP with PMS was also attempted using cobalt, copper, nickel and palladium porphyrin tetrasulfonate catalysts, however, no catalytic activity was observed with these structures.
