Reproductive toxicity of azoxystrobin to adult zebrafish (Danio rerio) Полный текст

In the past few decades, extensive application of azoxystrobin has led to great concern regarding its adverse effects on aquatic organisms. The objective of the present study was to evaluate the reproductive toxicity of azoxystrobin to zebrafish. After adult zebrafish of both sexes were exposed to 2, 20 and 200 μg/L azoxystrobin for 21 days, egg production, the fertilization rate, the gonadosomatic index (GSI) and hepatosomatic index (HSI), 17β-estradiol (E2), testosterone (T) and vitellogenin (Vtg) concentrations, and histological alterations in the gonads and livers were measured. Meanwhile, expression alterations of genes encoding gonadotropins and gonadotropin receptors (fshb, lhb, fshr and lhr), steroid hormone receptors (era, er2b and ar), steroidogenic enzymes (cyp11a, cyp11b, cyp17, cyp19a, cyp19b, hsd3b and hsd17b) in the hypothalamic-pituitary-gonad (HPG) axis and vitellogenin (vtg1 and vtg2) in the livers were also investigated. The results showed that reduced egg production and fertilization rates were observed at 200 μg/L azoxystrobin. In female zebrafish, reduced E2 and Vtg concentrations, decreased GSI, increased T concentrations, and histological alterations in the ovaries and livers were observed at 200 μg/L azoxystrobin, along with significant down-regulation of lhb, cyp19b, lhr, cyp19a, vtg1 and vtg2, and up-regulation of cyp17, hsd3b and hsd17b. In male zebrafish, increased E2 and Vtg concentrations, reduced T concentration and GSI, and histological alterations in the testes and livers were observed after exposure to 20 and 200 μg/L azoxystrobin, along with significant up-regulations of cyp19b, cyp11a, cyp17, cyp19a, hsd3b and hsd17b, vtg1 and vtg2. Moreover, cyp11a, hsd3b, cyp19a, vtg1 and vtg2 in male zebrafish were significantly up-regulated after treatment with 2 μg/L azoxystrobin. The results of the present study indicate that azoxystrobin led to reproductive toxicity in zebrafish and male zebrafish were more sensitive to azoxystrobin than female zebrafish.

Transport of environmental pollutants through porous media is influenced by colloids. Co-transport of As(V) and soil colloids at different pH were systematically investigated by monitoring breakthrough curves (BTCs) in saturated sand columns. A solute transport model was applied to characterize transport and retention sites of As(V) in saturated sand in the presence of soil colloids. A colloid transport model and the DLVO theory were used to reveal the mechanism and hypothesis of soil colloid-promoted As(V) transport in the columns. Results showed that rapid transport of soil colloids, regulated by pH and ionic strength, promoted As(V) transport by blocking As(V) adsorption onto sand, although soil colloids had low adsorption for As(V). The promoted transport was more significant at higher concentrations of soil colloids (between 25 mg L−1 and 150 mg L−1) due to greater blocking effect on As(V) adsorption onto the sand surfaces. The blocking effect of colloids was explained by the decreases in both instantaneous (equilibrium) As adsorption and first-order kinetic As adsorption on the sand surface sites. The discovery of this blocking effect improves our understanding of colloid-promoted As transport in saturated porous media, which provides new insights into role of colloids, especially colloids with low As adsorption capacity, in As transport and mobilization in soil-groundwater systems.

The Kuril Islands region is considered promising for development of salmon aquaculture. There are 41 salmon fish hatcheries in the Sakhalin Island and the Kuril Islands, 34 of them are hatcheries of the chum. Therefore, concentrations of six elements (Zn, Cu, Cd, Pb, As, and Hg) were determined in chum salmon were caught in this region. The contents of toxic elements (Cd, Pb, As, and Hg) don't exceed their maximum permissible concentrations (MPC) according to the Russian sanitary standards, but concentration of Pb are closely to MPC. Increased concentrations of Pb in wild chum have the natural origin. The unusual conditions of the Western Pacific are formed under the influence such factors as volcanism and upwelling.

Manufactured nanoparticles (MNP) rapidly undergo aging processes once released from products. Silver sulfide (Ag2S) is the major transformation product formed during the wastewater treatment process for Ag-MNP. We examined toxicogenomic responses of pristine Ag-MNP, sulfidized Ag-MNP (sAg-MNP), and AgNO3 to a model soil organism, Caenorhabditis elegans. Transcriptomic profiling of nematodes which were exposed at the EC30 for reproduction for AgNO3, Ag-MNP, and sAg-MNP resulted in 571 differentially expressed genes. We independently verified expression of 4 genes (numr-1, rol-8, col-158, and grl-20) using qRT-PCR. Only 11% of differentially expressed genes were common among the three treatments. Gene ontology enrichment analysis also revealed that Ag-MNP and sAg-MNP had distinct toxicity mechanisms and did not share any of the biological processes. The processes most affected by Ag-MNP relate to metabolism, while those processes most affected by sAg-MNP relate to molting and the cuticle, and the most impacted processes for AgNO3 exposed nematodes was stress related. Additionally, as observed from qRT-PCR and mutant experiments, the responses to sAg-MNP were distinct from AgNO3 while some of the effects of pristine MNP were similar to AgNO3, suggesting that effects from Ag-MNP is partially due to dissolved silver ions.

Heavy metals (Cr, Co, Ni, As, Cd, and Pb) can be bound to PM adversely affecting human health. Quantifying the source impacts on heavy metals can provide source-specific estimates of the heavy metal health risk (HMHR) to guide effective development of strategies to reduce such risks from exposure to heavy metals in PM2.5 (particulate matter (PM) with aerodynamic diameter less than or equal to 2.5 μm). In this study, a method combining Multilinear Engine 2 (ME2) and a risk assessment model is developed to more effectively quantify source contributions to HMHR, including heavy metal non-cancer risk (non-HMCR) and cancer risk (HMCR). The combined model (called ME2-HMHR) has two steps: step1, source contributions to heavy metals are estimated by employing the ME2 model; step2, the source contributions in step 1 are introduced into the risk assessment model to calculate the source contributions to HMHR. The approach was applied to Huzou, China and five significant sources were identified. Soil dust is the largest source of non-HMCR. For HMCR, the source contributions of soil dust, coal combustion, cement dust, vehicle, and secondary sources are 1.0 × 10−4, 3.7 × 10−5, 2.7 × 10−6, 1.6 × 10−6 and 1.9 × 10−9, respectively. The soil dust is the largest contributor to HMCR, being driven by the high impact of soil dust on PM2.5 and the abundance of heavy metals in soil dust.

Severe trace metal pollution due to industrial effluents releases was found in Jiulong River Estuary, Southern China. In this study, water samples were collected during effluent release events to study the dynamic changes of environmental conditions and metal partitioning among dissolved, particulate and colloidal phases controlled by estuarine mixing. Intermittent effluent discharges during low tide caused decreasing pH and dissolved oxygen, and induced numerous suspended particulate materials and dissolved organic carbon to the estuary. Different behaviors of Cu, Zn, Ni, Cr and Pb in the dissolved fraction against the conservative index salinity indicated different sources, e.g., dissolved Ni from the intermittent effluent. Although total metal concentrations increased markedly following effluent discharges, Cu, Zn, Cr, Pb were predominated by the particulate fraction. Enhanced adsorption onto particulates in the mixing process resulted in elevated partitioning coefficient (Kd) values for Cu and Zn, and the particle concentration effect was not obvious under such anthropogenic impacts. Colloidal proportion of these metals (especially Cu and Zn) showed positive correlations with dissolved or colloidal organic carbon, suggesting the metal-organic complexation. However, the calculated colloidal partitioning coefficients were relatively constant, indicating the excess binding capacity. Overall, the intermittent effluent discharge altered the particulate/dissolved and colloidal/soluble phase partitioning process and may further influence the bioavailability and potential toxicity to aquatic organisms.

Fungicides in aquatic environments can impact non-target bacterial and fungal communities and the invertebrate detritivores responsible for the decomposition of allochthonous organic matter. Additionally, in some aquatic systems daily water temperature fluctuations may influence these processes and alter contaminant toxicity, but such temperature fluctuations are rarely examined in conjunction with contaminants. In this study, the shredding amphipod Hyalella azteca was exposed to the fungicide pyraclostrobin in three experiments. Endpoints included mortality, organism growth, and leaf processing. One experiment was conducted at a constant temperature (23 °C), a fluctuating temperature regime (18–25 °C) based on field-collected data from the S. Llano River, Texas, or an adjusted fluctuating temperature regime (20–26 °C) based on possible climate change predictions. Pyraclostrobin significantly reduced leaf shredding and increased H. azteca mortality at concentrations of 40 μg/L or greater at a constant 23 °C and decreased leaf shredding at concentrations of 15 μg/L or greater in the fluctuating temperatures. There was a significant interaction between temperature treatment and pyraclostrobin concentration on H. azteca mortality, body length, and dry mass under direct aqueous exposure conditions. In an indirect exposure scenario in which only leaf material was exposed to pyraclostrobin, H. azteca did not preferentially feed on or avoid treated leaf disks compared to controls. This study describes the influence of realistic temperature variation on fungicide toxicity to shredding invertebrates, which is important for understanding how future alterations in daily temperature regimes due to climate change may influence the assessment of ecological risk of contaminants in aquatic ecosystems.

Concentration and spatial distribution of six phthalic acid esters (PAEs) and eight phenols in sediments of urban rivers, namely the Xi River (XR) and Pu River (PR) in Shenyang city, Northeast China were investigated and the ecological risk of these target pollutants was assessed based on the risk quotient (RQ) approach. Target PAEs and phenols were detected in most of sediment samples collected from the XR and PR. The concentrations of total PAEs in sediments varied from 22.4 to 369 μg/g dw in the XR and 3.71–46.9 μg/g dw in the PR. The levels of phenols ranged from 2.72 to 106 μg/g dw in the XR and 0.811–25.0 μg/g dw in the PR, respectively. The dominant pollutants in both XR and PR were DEHP, phenol and 4-methylphnol. The sampling locations XR1-3 in the XR suffered severe contamination from PAEs and phenols. The sites PR1 and PR6 were heavily polluted by phenols and PAEs, respectively. Almost all target PAEs and phenolic compounds in sediment of the XR exhibited medium or high ecological risk to organisms and the ecological risk in the PR mainly originated from PEAs, phenol and 4-methylphenol. These results would provide guidance for individual pollutant control and indicate that it is imperative to take some effective measures to reduce the pollution of those contaminants.

This study presents a detailed kinetic investigation on the uptake, acropetal translocation and transformation of BDE-47, 6-OH-BDE-47 and 6-MeO-BDE-47 in maize (Zea mays L.) by hydroponic exposure. Root uptake followed the order: BDE-47 > 6-MeO-BDE-47 > 6-OH-BDE-47, while 6-OH-BDE-47 was the most prone to acropetal translocation. Debromination rates of BDE-47 were 1.31 and 1.46 times greater than the hydroxylation and methoxylation rates, respectively. Transformation from BDE-47 to lower brominated OH/MeO-PBDEs occurred mainly through debromination first followed by hydroxylation or methoxylation. There was no transformation from 6-OH-BDE-47 or 6-MeO-BDE-47 to PBDEs. Methylation rate of 6-OH-BDE-47 was twice as high as that of 6-MeO-BDE-47 hydroxylation, indicating methylation of 6-OH-BDE-47 was easier and more rapid than hydroxylation of 6-MeO-BDE-47. Debromination and isomerization were potential metabolic pathways for 6-OH-BDE-47 and 6-MeO-BDE-47 in maize. This study provides important information for better understanding the mechanism on plant uptake and transformation of PBDEs.

Haze caused by high particulate matter loadings is an important environmental issue. PM2.5 was collected in Nanjing, China, during a severe haze–fog event and clear periods. The particulate-bound elements were chemically fractionated using sequential extractions. The average PM2.5 concentration was 3.4 times higher during haze–fog (96–518 μg/m³) than non-haze fog periods (49–142 μg/m³). Nearly all elements showed significantly higher concentrations during haze–fog than non-haze fog periods. Zn, As, Pb, Cd, Mo and Cu were considered to have higher bioavailability and enrichment degree in the atmosphere. Highly bioavailable fractions of elements were associated with high temperatures. The integrated carcinogenic risk for two possible scenarios to individuals exposed to metals was higher than the accepted criterion of 10⁻⁶, whereas noncarcinogenic risk was lower than the safe level of 1. Residents of a city burdened with haze will incur health risks caused by exposure to airborne metals.
