Phytoplankton pigments and epifluorescence microscopy as tools for ecological status assessment in coastal and estuarine waters, within the Water Framework Directive Полный текст

Inverted microscopy is widespread employed for the analysis of phytoplankton composition within water quality monitoring networks. However, the analysis at the lowest taxonomical level is not always required for ecological status assessment. In addition, inverted microscopy can underestimate the small phytoplankton, and not always distinguish photoautotrophic from heterotrophic cells. In this study, as alternative tools, epifluorescence microscopy and High Performance Liquid Chromatography (HPLC) were employed to characterize phytoplankton communities within waters of different trophic condition. Epifluorescence microscopy confirmed its effectiveness to count the small phytoplankton. Furthermore, significant correlations between nutrients of anthropogenic origin and nanoplankton abundances were found. However, this technique resulted very time-consuming. HPLC together with the CHEMTAX program was more appropriate than inverted microscopy, in terms of cost-effectiveness. Also, the main variability patterns observed in the phytoplankton community structure by HPLC coincided with previous findings in the study area. Nevertheless, a rapid screening at the inverted microscope is recommended.

The development and broad use of passive acoustic monitoring techniques have the potential to help assessing the large-scale influence of artificial noise on marine organisms and ecosystems. Deep-sea observatories have the potential to play a key role in understanding these recent acoustic changes. LIDO (Listening to the Deep Ocean Environment) is an international project that is allowing the real-time long-term monitoring of marine ambient noise as well as marine mammal sounds at cabled and standalone observatories. Here, we present the overall development of the project and the use of passive acoustic monitoring (PAM) techniques to provide the scientific community with real-time data at large spatial and temporal scales. Special attention is given to the extraction and identification of high frequency cetacean echolocation signals given the relevance of detecting target species, e.g. beaked whales, in mitigation processes, e.g. during military exercises.

The large green-tide events that occurred in the Yellow Sea in 2008 (3489km²) and 2009 (4994km²) are shown to be novel events preceded only once by a much smaller event in 2007 (82km²). The blooms originated in the coastal area of Jiangsu province and spread north-east towards the Shandong Peninsula. The blooms grew at different rates and mesoscale variability in surface winds explained the differences in the spatial and temporal patterns of blooms in 2008 and 2009. The 2009 bloom was tracked to its origin immediately offshore of extensive intertidal flats between Yancheng and Nantong where recent rapid expansion of Porphyra aquaculture has occurred. We review published hypotheses which have been advanced to explain the occurrence of blooms and in light of our findings, we conclude that the accumulation and disposal of waste Ulva prolifera from Porphyra aquaculture rafts is the most likely cause of the blooms.

The Adriatic coast of Punta Marina (Ravenna) is protected by 3-km long low crested breakwater structures (LCSs). Through a 3-years long multidisciplinar study, we assessed the impact of such defensive structures on environmental and biological condition. LCSs create pools where conditions are very different from the surrounding nearshore system. Mechanical disturbance by currents and waves varied greatly in intensity and frequency between seaward and landward sides of the structures. Sedimentary budget was positive at the landward side, but it was due to a gain on the seafloor and not on the emerged beach. The budget at seaward was negative. LCSs determine differences in benthic assemblages, alter the seasonal pattern of communities, and modify seasonal fluctuations of animal assemblages. Landward sheltered areas can be seen as “lagoonal island” surrounded by a “sea of marine habitat”. Differences in ecological quality status, obtained through M-AMBI, are due to the sum of these factors.

The relationship between chemical exposure and disease outbreak in fish has not been fully defined due to the limitations of experimental systems (model fish and pathogens). Therefore, we constructed a system using the Japanese flounder, Paralichthys olivaceus, and viral haemorrhagic septicemia virus (VHSV), and evaluated it by heavy oil (HO) exposure. The fish were exposed to HO at 0.3, 0.03, 0.003, and 0g/L following VHSV infection at doses of 10².⁵ or 10³.⁵ tissue culture infectious dose (TCID)₅₀/fish. As a result, groups given the dual stressors showed more than 90% mortality. Although VHSV infection at 10².⁵ and 10³.⁵ TCID₅₀/fish without HO exposure also induced high mortality, at 68.8% and 81.3%, respectively, HO exposure induced faster and higher mortality in the virus carrier fish, indicating that chemical stressors raise the risk of disease outbreak in fish. The experimental system established in this study could be useful for chemical risk assessment.

Polybrominated diphenyl ethers (PBDEs) are now found ubiquitously in the aquatic environment and biota, and there is a growing concern that PBDEs may disrupt endocrine systems, leading to reproductive impairments of aquatic animals. In our study, zebrafish (Danio rerio) were exposed to the 5ng/L, 1μg/L and 50μg/L of DE-71 for the duration of the whole life cycle (120days, from eggs to adults). The expression of selected genes along the brain–pituitary–gonadal (BPG) axis and liver, and the levels of plasma sex hormones were examined. In male fish, up-regulation of GnRH in brain, FSHβ and LHβ in pituitary, FSH-receptor, LH-receptor, and CYP19a in testis was clearly evident, while down-regulation of CYP11a and 3β-HSD was found in testis. In female fish, a 2.4-fold up-regulation of 3β-HSD was found in ovary upon exposure to 50μg/L of DE-71. GnRH in brain, FSHβ and LHβ in pituitary were also up-regulated, while ERβ, TH and TPH in brain and GnRH-receptor in pituitary were significantly down-regulated. Hepatic ERα, AR and VTG in males were all down-regulated, while hepatic ERα and AR in female were up-regulated. Serum estradiol (E₂) was reduced in both male and female upon exposure to DE-71, while significant increases in serum testosterone (T) and 11-keto-testosterone (11-KT) were only found in male but not female fish. The ratio of T/E₂ as well as the ratio of 11-KT/E₂ in male fish increased in a dose-dependent manner upon exposure to DE-71. Our overall results showed that whole life exposure of DE-71 altered the expression of regulatory genes and receptors at all three levels of the BPG axis in zebrafish, and the responses are sex dependent. The observed disruption of GnRH and GtHs can be further related to the subsequent disruption in both levels and balance sex steroid hormones.

Quantifying the uncertainty associated with monitoring protocols is essential to prevent the misclassification of ecological status and to improve sampling design. We assessed the Posidonia oceanica multivariate index (POMI) bio-monitoring program for its robustness in classifying the ecological status of coastal waters within the Water Framework Directive. We used a 7-year data set covering 30 sites along 500km of the Catalonian coastline to examine which version of POMI (14 or 9 metrics) maximises precision in classifying the ecological status of meadows. Five factors (zones within a site, sites within a water body, depth, years and surveyors) that potentially generate classification uncertainty were examined in detail. Of these, depth was a major source of uncertainty, while all the remaining spatial and temporal factors displayed low variability. POMI 9 matched POMI 14 in all factors, and could effectively replace it in future monitoring programs.

In this study, baseline information about the environmental status of Ushuaia (UB) and Golondrina (GB) bays is presented. Surface and bottom seawater and freshwater discharged from land were evaluated. Multivariate analysis identified different water quality zones within the bays, two of them located next to the north and northwest coastlines of UB, where the majority of human activities are developed. Porosity, total organic matter, biochemical components, ammonium, and phytopigments were determined in sediment samples from each quality zone. Benthic fluxes of nutrients and dissolved oxygen were assessed in situ using opaque chambers. In northwest zone of UB, carbon equivalents of proteins and carbohydrates in surficial sediments were the same order as in hypertrophic ecosystems, whereas ammonium and phosphate released from sediment greatly exceeded the allochthonous sources. Management of municipal wastewater is required to remediate this chronic pollution.

Histopathological alterations can indicate time-integrated impacts on organisms stemming from alterations at lower biological organisation levels. Long-term (native mussels) and short-term (transplanted clams) changes in the tissues of molluscs exposed to the effluents from two land-based marine fish farms (LBMFFs) were determined. Histological alterations were related to the δ¹⁵N isotopic signal measured in mussels and macroalgae. Effluents from LBMFFs were found to cause severe and moderate gill filament exfoliation in clams and mussels, respectively. Some transplanted clams showed severe degrees of hemocytic phagocytosis in gonads and connective tissue. In an attempt to semi-quantitatively summarize the observed histopathological alterations, a weighted index of damage (WID) was calculated for each type of alteration, species and sampling site. The WID was clearly related to the δ¹⁵N descriptor of exposure. Further studies aimed at standardizing this relationship may establish critical thresholds of the descriptor for its implementation within environmental monitoring plans for LBMFFs.

The use of sonar by military vessels during military exercises may produce acoustic pollution of the marine environment. States have an obligation under international law to reduce and control this form of pollution. Regulation of the use of sonar is rendered more complex by the specific regime applicable to warships, i.e. sovereign immunity. Immunity however does not prevent in all cases action by States other than the flag State, notwithstanding the limitations imposed by Art. 236 of the United Nations Convention on the Law of the Sea. More importantly, immunity does not prevent the flag State from taking measures to reduce pollution caused by its own warships. Under some environmental treaties, flag States not only may, but have the obligation to adopt measures.
