Rapid quantification of infectious enterovirus from surface water in Bohai Bay, China using an integrated cell culture-qPCR assay Полный текст

To rapidly quantify infectious enteroviruses polluting the coastal seawaters, a newly developed integrated cell culture and reverse transcription quantitative PCR (ICC-RT-qPCR) assay was used to identify the contamination by enteroviruses in winter seawater samples of Bohai Bay, Tianjin, China. The gene copies of enteroviral 5′UTRs correlated to the initial inoculum numbers across the concentration range of 0.05–500PFUmL⁻¹ (correlation coefficient (R²) was 0.9667). ICC-qPCR revealed that five of seven samples (70.4%) were positive for infectious enteroviruses. The concentration of enteroviruses was estimated at 0.2–21PFUL⁻¹. The result demonstrated that the contamination of enteroviruses in this coastal area may constitute a potential public health risk. This study established a practical assay for widespread monitoring studies of aquatic environments for viral contamination and provided meaningful data for human waterborne viral risk assessment.

Histopathological alterations can indicate time-integrated impacts on organisms stemming from alterations at lower biological organisation levels. Long-term (native mussels) and short-term (transplanted clams) changes in the tissues of molluscs exposed to the effluents from two land-based marine fish farms (LBMFFs) were determined. Histological alterations were related to the δ¹⁵N isotopic signal measured in mussels and macroalgae. Effluents from LBMFFs were found to cause severe and moderate gill filament exfoliation in clams and mussels, respectively. Some transplanted clams showed severe degrees of hemocytic phagocytosis in gonads and connective tissue. In an attempt to semi-quantitatively summarize the observed histopathological alterations, a weighted index of damage (WID) was calculated for each type of alteration, species and sampling site. The WID was clearly related to the δ¹⁵N descriptor of exposure. Further studies aimed at standardizing this relationship may establish critical thresholds of the descriptor for its implementation within environmental monitoring plans for LBMFFs.

The accumulation of debris is an insidious problem throughout the world’s oceans. Here we document 234.24 items of macro-debris/km² in the shallow populated parts of Majuro lagoon (Republic of the Marshall Islands) which is the second highest standing stock of macro-debris recorded to date in any benthic marine habitat in the world. The majority of macro-debris was from household sources (78.7%) with the peak abundance recorded in areas of medium affluence. Marine debris causes suffocation, shading, tissue abrasion and mortality of corals and we show a significant negative correlation exists between the level of hard coral cover and coverage of marine debris. Given long decomposition times, even if the input of rubbish to Majuro lagoon is stopped immediately, the standing stock of debris will persist for centuries. Multiple new initiatives are needed to curtail the direct and indirect dumping of waste in Majuro lagoon.

Light-induced bird strikes are known to occur when vessels navigate during darkness in icy waters using powerful searchlight. In Southwest Greenland, which is important internationally for wintering seabirds, we collected reports of incidents of bird strikes over 2–3 winters (2006–2009) from navy vessels, cargo vessels and trawlers (total n=19). Forty-one incidents were reported: mainly close to land (<4km, 78%), but one as far offshore as 205km. Up to 88 birds were reported killed in a single incident. All occurred between 5p.m. and 6a.m. and significantly more birds were involved when visibility was poor (snow) rather than moderate or good. Among five seabird species reported, the common eider (Somateria mollissima) accounted for 95% of the bird casualties. Based on spatial analyses of data on vessel traffic intensity and common eider density we are able to predict areas with high risk of bird strikes in Southwest Greenland.

Southern resident killer whales in British Columbia and Washington are exposed to heavy vessel traffic. This study investigates their exposure to exhaust gases from whale-watching vessels by using a simple dispersion model incorporating data on whale and vessel behavior, atmospheric conditions, and output of airborne pollutants from the whale-watching fleet based on emissions data from regulatory agencies. Our findings suggest that current whale-watching guidelines are usually effective in limiting pollutant exposure to levels at or just below those at which measurable adverse health effects would be expected in killer whales. However, safe pollutant levels are exceeded under worst-case conditions and certain average-case conditions. To reduce killer whale exposure to exhaust we recommend: vessels position on the downwind side of whales, a maximum of 20 whale-watching vessels should be within 800m at any given time, viewing periods should be limited, and current whale-watch guidelines and laws should be enforced.

Coral community structure is often governed by a suite of processes that are becoming increasingly influenced by land-use changes and related terrestrial discharges. We studied sites along a watershed gradient to examine both the physical environment and the associated biological communities. Transplanted corals showed no differences in growth rates and mortality along the watershed gradient. However, coral cover, coral richness, and coral colony density increased with increasing distance from the mouth of the bay. There was a negative relationship between coral cover and mean suspended solids concentration. Negative relationships were also found between terrigenous sedimentation rates and the richness of adult and juvenile corals. These results have major implications not only for Pacific islands but for all countries with reef systems downstream of rivers. Land development very often leads to increases in river runoff and suspended solids concentrations that reduce coral cover and coral diversity on adjacent reefs.

Halogenated natural products (HNPs) are frequently detected in marine organisms. High HNP concentrations have previously been found in marine mammals from the Great Barrier Reef, Australia, including in the blubber of herbivorous dugongs (Dugong dugon). To identify the source of HNPs we initially focused on the analysis of Australian seagrass (Halophila ovalis) which serves as the principal food source for dugongs. GC/MS analysis of the seagrass indicated the presence of several organobromine compounds. One compound was identified as 2,3,4,5-tetrabromo-1-methylpyrrole (TBMP) by synthesis. Subsequent analysis of semipermeable membrane devices demonstrated that the photo-sensitive TBMP is widespread in the Great Barrier Reef (Queensland, Australia). The detection of larger TBMP concentrations in fish fillets from Chile and traces in mussels from New Zealand indicated that this potential HNP may be distributed throughout the Southern Pacific Ocean.

Effects of eutrophication on marine ecosystems have been widely studied, even if both the effects on deep subtidal rocky assemblages and response of different successional stages to nutrients impact are still not clear. In this context, the study aimed to evaluate the effects of nutrient enrichment on Mediterranean macroalgal assemblages associated with coralligenous habitat. A manipulative field experiment was carried out by supplying both mature and early successional stages of assemblages with nutrients. A total of 62 macroalgal species were identified. Multivariate and univariate analyses showed that the structure of both mature and early successional macroalgal assemblages of coralligenous significantly varied between areas treated with nutrients and not treated areas. Moreover, differences were stronger when macroalgal assemblages were in the early successional stage than in the mature one. Results highlighted the role played by nutrients in determining the structure of macroalgal coralligenous assemblages, furthermore suggesting possible synergetic effects with other kinds of disturbances.

We examined Cu contamination effects on macrobenthic communities and Cu concentration in invertebrates within Shelter Island Yacht Basin, San Diego Bay, California. Results indicate that at some sites, Cu in sediment has exceeded a threshold for “self defense” mechanisms and highlight the potential negative impacts on benthic faunal communities where Cu accumulates and persists in sediments. At sites with elevated Cu levels in sediment, macrobenthic communities were not only less diverse but also their total biomass and body size (individual biomass) were reduced compared to sites with lower Cu. Cu concentration in tissue varied between species and within the same species, reflecting differing abilities to “regulate” their body load. The spatial complexity of Cu effects in a small marina such as SIYB emphasizes that sediment-quality criteria based solely on laboratory experiments should be used with caution, as they do not necessarily reflect the condition at the community and ecosystem levels.

Mussels were maintained for 4weeks under different combinations of dissolved oxygen concentration (1.5, 3.0 and 6.0mgO₂l⁻¹) and salinity (15, 20, 25 and 30) in a 3×4 factorial design experiment. Clearance rate (CR), absorption efficiency (AE), respiration rate (RR) and scope for growth (SFG) decreased with decreasing salinity and dissolved oxygen concentration (DO), while excretion rate (ER) increased with decreasing salinity and increasing DO. The O:N ratio was <10 at salinities of 15 and 20, irrespective of DO levels. SFG was negative in most of the treatments, except for those under 6.0mgO₂l⁻¹ or at a salinity of 30 when DO was lower. The results may help explain the distribution pattern of Perna viridis in Hong Kong waters and provide guidelines for mussel culture site selection.
