Marine protected areas in Latin America and Caribbean threatened by polycyclic aromatic hydrocarbons

The present study is a literature-based analysis investigating occurrence and the possible consequences of polycyclic aromatic hydrocarbons (PAH) in marine protected areas (MPAs) of Latin America and Caribbean. The approach using overlapping of georeferenced MPA polygons with data compiled from peer-reviewed literature, published during the last 15 years, showed 341 records of PAH in 9 countries. PAH was reported to occur within the boundaries of 36 MPAs located in Argentina, Brazil, Colombia, Mexico, Nicaragua and Uruguay. According to quality guidelines, low to moderate impacts are expected in MPAs categorized in different management classes. Considering sediment samples, 13% of the records presented concentrations enough to cause occasional toxicity. Such level of risk was also seen in Ramsar sites and in Amazonian MPAs. In addition, based on concentrations reported in biota, occasional deleterious effects on organisms from Biosphere Reserves might occur. Diagnostic ratios pointed out petrogenic and pyrolytic processes as PAH predominant sources, and were mainly attributed to the proximity to ports, industries and urban areas. MPAs located in the vicinity of impact-generating areas may be under threat and require government attention and action, mainly through implementation of contamination monitoring programs.

Previous studies have shown that accumulation of perfluoroalkyl acids (PFAAs) in the tissues of aquatic species is highly variable. Movement and migration patterns in these species represent an important consideration when evaluating contaminant accumulation in exposed biota, and may have a large influence on the risk profiles for migratory seafood species. In this study, relationships between PFAA concentrations in muscle and liver tissue, and recent fish migration history (inferred from metals profiles in fish otoliths, otherwise known as otolith chemistry) were evaluated in Sea Mullet (Mugil cephalus). A greater number of PFAAs, and higher concentrations, were found in liver compared to muscle tissue. Perfluorooctane sulfonate (PFOS) was present in highest concentrations in both muscle and liver tissues, and there was strong correlation in concentrations between these two tissues. PFOS was found to decrease and increase alongside recent strontium and barium concentrations (respectively) in the otolith, suggesting higher concentrations of PFAAs in fish recently exposed to comparatively lower salinity environments. This study highlights how otolith chemistry can be employed to examine links between contaminant concentrations in fish, and their recent migration history. This approach shows promise for studying contaminant residues in mobile seafood species within the natural environment.

As ubiquitous, persistent organic pollutants, polycyclic aromatic hydrocarbons (PAHs) have adverse impacts on human health. Phenanthrene (Phe) is one of the most abundant PAHs in the environment. However, the long-term effects of exposure to environmental level of Phe on the kidneys and the potential mechanisms are unclear. T helper (Th) cells, a subtype of CD4⁺ T cells that play a central role in the renal immune microenvironment. In this study, male mice were chronically exposed to 5, 50, and 500 ng/kg bw Phe every other day for total 210 days. Those results indicated that environmental Phe exposure caused kidney hypertrophy, injury and fibrosis in the mice. Chronic, long-term environmental level of Phe exposure did not significantly alter the innate immune response but induced adaptive immune response changes (Th1/Th2 related cytokines release), causing a type 1 immune response in the 5 ng/kg bw Phe group and a type 2 immune response in the high dose groups (50 and 500 ng/kg bw). This study provides novel insights into the roles of adaptive immune response in long-term PAH exposure-induced chronic kidney injury and fibrosis, which is beneficial for further understanding the potential health hazards of PAHs and providing new avenues for immune intervention strategies to alleviate PAHs toxicity.

Decades of intensive discharge from industrial activities into coastal systems has resulted in the accumulation of a variety of persistent organic pollutants (POPs) in marine waters and sediments, having detrimental impacts on aquatic ecosystems and the resident biota. POPs are among the most hazardous chemicals originating from industrial activities due to their biotoxicity and resistance to environmental degradation. Bacterial communities are known to break down many of these aromatic compounds, and different members of naturally occurring bacterial consortia have been described to work in syntrophic association to thrive in heavily contaminated waters and sediments, making them potential candidates as bioindicators of environmental pollution. In this study environmental, sampling was combined with chemical analysis of pollutants and high-resolution sequencing of bacterial communities using Next Generation Sequencing molecular biology tools. The aim of the present study was to describe the bacterial communities from marine sediments containing high loads of POPs and to identify relevant members of the resident microbial communities that may act as bioindicators of contamination. Marine sediments were collected from a coastal bay area of the Baltic Sea historically influenced by intense industrial activity, including metal smelting, oil processing, and pulp and paper production. Different types of POPs were detected at high concentrations. Fiberbank sediments, resulting from historic paper industry activity, were found to harbour a clearly distinct bacterial community including a number of bacterial taxa capable of cellulolytic and dechlorination activities. Our findings indicate that specific members of the bacterial communities thrive under increasing levels of POPs in marine sediments, and that the abundances of certain taxa correlate with specific POPs (or groups), which could potentially be employed in monitoring, status assessment and environmental management purposes.

Biochar application to soils has become a focus of research during the last decade due to its high potential for C sequestration. Nevertheless, there is no exhaustive information on the long-term effects of biochar application in soils contaminated with trace elements. In this work, a 2-year field experiment was conducted comprising the application of different types of biochar to acidic and moderately acidic soils with high concentrations of As, Cu, Pb, Ba and Zn. In addition, representative samples of each biochar were buried in permeable bags that allowed the flow of water and microorganisms but not their physical interaction with soil aggregates. The biochars significantly adsorbed trace elements from polluted soils. However, given the high total concentration of these persistent trace elements in the soils, the application of biochars did not succeed in reducing the concentration of available metals (CaCl₂ extractable fraction). After 2 years of ageing under field conditions, some degradation of the biochars from olive pit, rice husk and wood were observed. This study provides novel information concerning the biochar alterations during ageing in polluted soils, as the decrease of aryl C signal observed by ¹³C nuclear magnetic resonance (NMR) spectroscopy and the presence of O-containing groups shown by Fourier Transform mid-Infrared Spectroscopy (FT-IR) in aged biochar which enhanced trace elements adsorption. Scanning electron microscopy (SEM) revealed slight changes on surface morphology of aged biochar particles.

Gala Lake is an internationally important lake due to its location on one of the world's most important bird migration routes. For this reason, water quality of the lake is of great concern. However, the lake surrounded by paddy fields may face eutrophication and toxic metal contamination due to excessive use of fertilizers and pesticides. In this study, impact of paddy fields on water quality of the Gala Lake was investigated. The concentrations of metal(loid)s and physico-chemical parameters in surface water samples taken from the lake were measured and compared with water quality guidelines. Also, human health risks and contamination status of metal(loid)s were assessed. The mean NO₂, SRP and BOD concentrations in the lake exceeded the permissible levels for both salmonid and cyprinid waters. The mean BOD value indicated contaminated water quality in the lake, while mean COD and SRP values indicated lightly contaminated water quality. The mean As, Cr and Pb values in the winter exceeded the drinking water limits set by WHO and EC, while the mean Cr and Zn values exceeded the limit values for the protection of freshwater aquatic organisms set by USEPA. Similarly, heavy metal pollution index and the degree of contamination values in the winter indicated that the lake water is moderately polluted. Health risk assessment results revealed that As and Cr in the lake water via ingestion exposure pathway may pose both non-carcinogenic and carcinogenic health risks to the residents. The results of this study indicated that paddy fields are a major source of nutrients, organic matter and toxic metal(loid)s to the Gala Lake. To improve the water quality of the lake, we suggest that excessive use of fertilizers and pesticides should be controlled to reduce metal(loid) and nutrient loads from the paddy fields.

Perennial contaminated groundwater seepage is threatening the downstream ecosystem of the Kazipally Pharmaceutical industrial area located in South India. The sources of seepage are unknown for the last three decades that challenging the regulatory authorities and industries. In general, water quality monitoring and geophysical techniques are applied to identify the sources. However, these techniques may lead to ambiguous results and fail to identify the seepage sources, especially when the area is urbanized/paved, and groundwater is already contaminated with other leakage sources that have similar chemical compounds. In the present study, a novel and multidisciplinary approach were adopted that includes satellite-based Land Surface Temperature (LST) observations, field-based Electrical Resistivity Tomography (ERT), continuous Soil Electrical Conductivity (SEC) and Volumetric Soil Moisture (VSM%) measurements along with groundwater levels monitoring to identify the sources and to control the seepage. The integrated results identified that the locations with the Standard Thermal Anomaly (STA) in the range of −0.5 to -1 °C, VSM% >50%, SEC > 1.5 mS/cm, bulk resistivity < 12 Ω m with shallow groundwater levels < 3 m below ground level (bgl) are potentially contaminated perennial seepage sources. Impermeable sheet piles have been installed across the groundwater flow direction to control the seepage up to 1.5 m bgl, where groundwater frequently intercepts land surface. The quantity of dry season groundwater seepage has been declined by 79.2% after these interventions, which in turn minimized the treatment cost of 1,96,283 USD/year and improved the downstream ecosystem.

The objective of this study was to evaluate the biological and nutritional characteristics of Spodoptera frugiperda (Lepidoptera: Noctuidae), an arthropod pest widely distributed in agricultural regions, after exposure to nano-CeO₂ via an artificial diet and to investigate the presence of cerium in the body of this insect through X-ray fluorescence mapping. Nano-CeO₂, micro-CeO₂, and Ce(NO₃)₃ were incorporated into the diet (0.1, 1, 10, and 100 mg of Ce L⁻¹). Cerium was detected in caterpillars fed with diets containing nano-CeO₂ (1, 10 and 100 mg of Ce L⁻¹), micro-CeO₂ and Ce(NO₃)₃, and in feces of caterpillars from the first generation fed diets with nano-CeO₂ at 100 mg of Ce L⁻¹ as well. The results indicate that nano-CeO₂ caused negative effects on S. frugiperda. After it was consumed by the caterpillars, the nano-CeO₂ reduced up to 4.8% of the pupal weight and 60% of egg viability. Unlike what occurred with micro-CeO₂ and Ce(NO₃)₃, nano-CeO₂ negatively affected nutritional parameters of this insect, as consumption rate two times higher, increase of up to 80.8% of relative metabolic rate, reduction of up to 42.3% efficiency of conversion of ingested and 47.2% of digested food, and increase of up to 1.7% of metabolic cost and 8.7% of apparent digestibility. Cerium caused 6.8–16.9% pupal weight reduction in second generation specimens, even without the caterpillars having contact with the cerium via artificial diet. The results show the importance of new ecotoxicological studies with nano-CeO₂ for S. frugiperda in semi-field and field conditions to confirm the toxicity.

The majority of experimental studies carried out to date, regarding the effects of pollutants on meiofauna have been conducted by means of closed systems, and rarely using open ones. The current work explored the impact of three Polycyclic Aromatic Hydrocarbons (PAHs), anthracene, pyrene and benzo[a]pyrene, applied alone or combined, on meiobenthic nematodes using both systems. The results revealed that single PAHs impacted the nematofauna similarly in closed or open systems with a higher toxicity observed for benzo[a]pyrene. However, the closed microcosms contaminated with PAHs became organically enriched, resulting in more non-selective deposit feeders and omnivores-carnivores. Taxonomic and functional effects related to combinations of PAHs were close to those of individual treatments in closed systems, however, for open ones, the outcomes were different. The caudal morphology influenced the response of taxa during their avoidance/endurance of hydrocarbons in open systems where the effects of PAHs mixtures appeared not only additive but also synergetic. Based on the results of the study, the use of open systems is preferred to closed ones as the research outcomes were more accurate and representing better conditions prevailing in nature.

Small extracellular vesicles (sEV) are small lipid bilayer particles released by cells. sEV have been shown to play critical roles in intercellular communication. Di (2-ethylhexyl) phthalate (DEHP), widely used as plasticizers, has been detected in the environment and human beings. DEHP was found to exist in the air particles and showed pulmonary toxicity. However, there’s little knowledge about the role of sEV in mediating the toxicity of DEHP-induced lung toxicity. We hypothesized that sEV mediated the toxicity of DEHP through their cargo. To validate this, lung epithelial cells (A549) were exposed to various concentrations (0, 0.2, 2 and 20 μM) of DEHP for 48 h. sEV extracted from DEHP-exposed A549 cells were cultured with unexposed A549 cells. Results showed that DEHP induced the epithelial-mesenchymal transition (EMT) and promoted the migration and invasion ability of A549 cells. The number of released sEV significantly increased in the culture media in DEHP-exposed groups compared to unexposed groups. The sEV can enter the unexposed A549 cells and enhance its EMT and the ability of migration and invasion. Treatment with GW4869 in DEHP-exposed A549 cells almost blocked the effects of DEHP-elicited sEV in normal A549 cells. Sequencing and functional analysis showed that the enrichment of significantly differentially expressed sEV miRNAs were related to tumor etiology. MiR-26a-5p was significantly enriched in DEHP-elicited sEV. Inhibition of miR-26a-5p in DEHP-exposed cells led to the downregulation of miR-26a-5p in sEV, and thus abolished the effects of DEHP-elicited sEV in normal A549 cells, whereas overexpression of miR-26a-5p restored the effects. The transcription factors twist is one of the downstream targets in the effects of sEV-miR-26a-5p on EMT process. In all, our results showed that DEHP exposure promoted the secretion of miR-26a-5p in sEV, which subsequently enhanced the EMT, migration and invasion ability in neighboring normal cells via the twist.
