Variability of PCB burden in 5 fish and sharks species of the French Mediterranean continental slope

Despite being generally located far from contamination sources, deep marine ecosystems are impacted by chemicals like PCB. The PCB contamination in five fish and shark species collected in the continental slope of the Gulf of Lions (NW Mediterranean Sea) was measured, with a special focus on intra- and interspecific variability and on the driving factors. Significant differences occurred between species. Higher values were measured in Scyliorhinus canicula, Galeus melastomus and Helicolenus dactylopterus and lower values in Phycis blennoides and Lepidorhombus boscii. These differences might be explained by specific abilities to accumulate and eliminate contaminant, mostly through cytochrome P450 pathway. Interindividual variation was also high and no correlation was observed between contamination and length, age or trophic level. Despite its major importance, actual bioaccumulation of PCB in deep fish is not as documented as in other marine ecosystems, calling for a better assessment of the factors driving individual bioaccumulation mechanisms and originating high variability in PCB contamination.

Pollution of the oceans by microplastics (<5 mm) represents a major environmental problem. To date, a limited number of studies have investigated the level of contamination of marine organisms collected in situ. For extraction and characterization of microplastics in biological samples, the crucial step is the identification of solvent(s) or chemical(s) that efficiently dissolve organic matter without degrading plastic polymers for their identification in a time and cost effective way. Most published papers, as well as OSPAR recommendations for the development of a common monitoring protocol for plastic particles in fish and shellfish at the European level, use protocols containing nitric acid to digest the biological tissues, despite reports of polyamide degradation with this chemical. In the present study, six existing approaches were tested and their effects were compared on up to 15 different plastic polymers, as well as their efficiency in digesting biological matrices. Plastic integrity was evaluated through microscopic inspection, weighing, pyrolysis coupled with gas chromatography and mass spectrometry, and Raman spectrometry before and after digestion. Tissues from mussels, crabs and fish were digested before being filtered on glass fibre filters. Digestion efficiency was evaluated through microscopical inspection of the filters and determination of the relative removal of organic matter content after digestion. Five out of the six tested protocols led to significant degradation of plastic particles and/or insufficient tissue digestion. The protocol using a KOH 10% solution and incubation at 60 °C during a 24 h period led to an efficient digestion of biological tissues with no significant degradation on all tested polymers, except for cellulose acetate. This protocol appeared to be the best compromise for extraction and later identification of microplastics in biological samples and should be implemented in further monitoring studies to ensure relevance and comparison of environmental and seafood product quality studies.

International audience | Antibiotic use in the early 1900 vastly improved human health but at the same time started an arms race of antibiotic resistance. The widespread use of antibiotics has resulted in ubiquitous trace concentrations of many antibiotics in most environments. Little is known about the impact of these antibiotics on microbial processes or “non-target” organisms. This mini-review summarizes our knowledge of the effect of synthetically produced antibiotics on microorganisms involved in biogeochemical cycling. We found only 31 articles that dealt with the effects of antibiotics on such processes in soil, sediment, or freshwater. We compare the processes, antibiotics, concentration range, source, environment, and experimental approach of these studies. Examining the effects of antibiotics on biogeochemical processes should involve environmentally relevant concentrations (instead of therapeutic), chronic exposure (versus acute), and monitoring of the administered antibiotics. Furthermore, the lack of standardized tests hinders generalizations regarding the effects of antibiotics on biogeochemical processes. We investigated the effects of antibiotics on biogeochemical N cycling, specifically nitrification, denitrification, and anammox. We found that environmentally relevant concentrations of fluoroquinolones and sulfonamides could partially inhibit denitrification. So far, the only documented effects of antibiotic inhibitions were at therapeutic doses on anammox activities. The most studied and inhibited was nitrification (25–100 %) mainly at therapeutic doses and rarely environmentally relevant. We recommend that firm conclusions regarding inhibition of antibiotics at environmentally relevant concentrations remain difficult due to the lack of studies testing low concentrations at chronic exposure. There is thus a need to test the effects of these environmental concentrations on biogeochemical processes to further establish the possible effects on ecosystem functioning

One of the main issue measuring gas emissions on such systems is related to their high heterogeneity of flow conditions (surface water distribution, feeding/resting periods etc). It is thus of importance to develop a specific methodology to be able first to determine representative gas emission fluxes and then to decide how filter‘s operation can affect these emissions. This study investigates greenhouse gas emissions from a full-scale VFCW for raw wastewater treatment. The plant designed for 800 population equivalents combines two stages of vertical subsurface flow filters planted with Phragmites. Australis. Gaseous emissions were continuously monitored during three weeks representing an overall feeding/resting period (1/3 weeks). Several automatic closed chambers connected to an infrared analyzer were placed at different strategic positions of the filters and allowed the measurement of tens of gaseous fluxes per day and per position. Dissolved N2O concentration was measured in the inlet and the outlet of each filtration stage using N2O micro-sensors. A combination of on-line sensors for NH4+ and NO3- and 24-h composite samples was used to characterize the process performances of each filtration stage. Finally, O2concentration was regularly measured in the air phase of the porous media in order to evaluate the aerobic conditions of the filters. Ammonium removal was on average of 94% during the monitoring period. The continuous measurement highlighted strong spatial and temporal variability of gaseous fluxes. This latter was observed at different time scales (day, week, feeding/resting cycle) and was linked to: (i) the intermittent feeding of the filters, (ii) oxygen content in the porous media and (iii) environmental conditions such as the ambient temperature. Dissolved N2O flux represented about 20% of the total flux (gas + liquid) which indicates the importance of accounting the dissolved flux in the N2O budget. The two filtration stages exhausted contrasted fluxes of methane and nitrous oxide in relation to carbon load and oxygen availability for carbon removal and nitrification. From a methodological point of view, this study indicates that: (i) continuous monitoring of greenhouse gas fluxes during at least an overall filtration cycle and (ii) appropriate spatial sampling strategy are decisive for a reliable determination of emission factors in VFCWs. The overall N2O emission factor estimated during the monitoring period was of 0.78% of the influent total nitrogen which is 28 times higher than the default IPCC factor (0.023% of the TN load). As N2O emissions were strongly correlated to the oxygen concentration within the porous media, it is suspected that nitrification was the main contributor to N2O production in the VFCW studied.

International audience | Insecticides have long been used as the main method in limiting agricultural pests, but their widespread use has resulted in environmental pollution, development of resistances, and biodiversity reduction. The effects of insecticides at low residual doses on both the targeted crop pest species and beneficial insects have become a major concern. In particular, these low doses can induce unexpected positive (hormetic) effects on pest insects, such as surges in population growth exceeding what would have been observed without pesticide application. Methomyl and chlorpyrifos are two insecticides commonly used to control the population levels of the cotton leafworm Spodoptera littoralis, a major pest moth. The aim of the present study was to examine the effects of sublethal doses of these two pesticides, known to present a residual activity and persistence in the environment, on the moth physiology. Using a metabolomic approach, we showed that sublethal doses of methomyl and chlorpyrifos have a systemic effect on the treated insects. We also demonstrated a behavioral disruption of S. littoralis larvae exposed to sublethal doses of methomyl, whereas no effects were observed for the same doses of chlorpyrifos. Interestingly, we highlighted that sublethal doses of both pesticides did not induce a change in acetylcholinesterase activity in head of exposed larvae

Passive sampling devices (PS) are widely used for pollutant monitoring in water, but estimation of measurement uncertainties by PS has seldom been undertaken. The aim of this work was to identify key parameters governing PS measurements of metals and their dispersion. We report the results of an in situ intercomparison exercise on diffusive gradient in thin films (DGT) in surface waters. Interlaboratory uncertainties of time-weighted average (TWA) concentrations were satisfactory (from 28% to 112%) given the number of participating laboratories (10) and ultra-trace metal concentrations involved. Data dispersion of TWA concentrations was mainly explained by uncertainties generated during DGT handling and analytical procedure steps. We highlight that DGT handling is critical for metals such as Cd, Cr and Zn, implying that DGT assembly/dismantling should be performed in very clean conditions. Using a unique dataset, we demonstrated that DGT markedly lowered the LOQ in comparison to spot sampling and stressed the need for accurate data calculation.

International audience | For herbivore insects, digesting can be somewhat challenging, as the defense mechanisms evolved by plants, including the release of phenolics like the non-protein amino acid l-3,4-dihydroxyphenylalanine (l-DOPA), can cause fitness costs. In addition, industrial and agricultural activities have elevated the amounts of iron that can be found in nature and more particularly FeSO4 that is used as fertilizer. Traces of iron can enhance the auto-oxidation of l-DOPA, in turn, generating reactive oxygen species (ROS) and consequently oxidative stress in insects. We examined the effects of the ion Fe2+ (as FeSO4) and l-DOPA on fifth instars of the desert locust Schistocerca gregaria. We measured the level of oxidative damage occurring to macromolecules (proteins and lipids) from midgut and thoracic tissues and assessed the activities of responsive antioxidant enzymes. Injected l-DOPA and redox-active metal iron generated ROS which caused oxidative damages to proteins and lipids to S. gregaria. The protein carbonyls and lipid peroxides present in tissue homogenates were elevated in treated insects. No synergism was observed when l-DOPA was co-injected with Fe2+. K (m) values of superoxide dismutase (SOD), glutathione reductase (GR), and glutathione peroxidase (GPx) were 4.3, 2.6, and 4.0 mM in thoracic muscles and 5.00, 2.43, and 1.66 mM in whole midgut for SOD, GR, and GPx, respectively, and 8.3 and 3.43 M for catalase (CAT) in the two tissues, respectively. These results suggest higher affinities of GPx and CAT to H2O2 in midgut than in muscles. The time-course changes in activities of antioxidant enzymes and amounts of protein carbonyls and lipid peroxides showed fluctuating patterns, suggesting complex interactions among macromolecules, l-DOPA and FeSO4, and their degradation products. Our results demonstrated the stressful effects of l-DOPA and FeSO4, proving that iron-containing fertilizers are pollutants that can strongly affect S. gregaria.

International audience | In estuarine ecosystems, metallic and organic contaminants are mainly associated with fine grain sediments which settle on mudflats. Over time, the layers of sediment accumulate and are then transformed by diagenetic processes mainly controlled by microbial activity, recording the history of the estuary's chemical contamination. In an environment of this specific type, we investigated the evolution of the chemical contamination and the structure of both total and active microbial communities, based on PhyloChip analysis of a 4.6-m core corresponding to a 40-year sedimentary record. While the archaeal abundance remained constant along the core, a decrease by one order of magnitude in the bacterial abundance was observed with depth. Both total and active microbial communities were dominated by Proteobacteria, Actinobacteria, and Firmicutes in all sediment samples. Among Proteobacteria, alpha-Proteobacteria dominated both total (from 37 to 60 %) and metabolically active (from 19.7 to 34.6 %) communities, including the Rhizobiales, Rhodobacter, Caulobacterales, and Sphingomonadales orders. Co-inertia analysis revealed a relationship between polycyclic aromatic hydrocarbons, zinc and some polychlorobiphenyls concentrations, and the structure of total and active microbial communities in the oldest and most contaminated sediments (from 1970 to 1975), suggesting that long-term exposure to chemicals shaped the structure of the microbial community.

International audience | Barrage fishponds may represent a significant surface water area in some French regions. Knowledge on their effect on water resources is therefore necessary for the development of appropriate water quality management plans at the regional scale. Although there is much information on the nutrient removal capacity of these water bodies, little attention has been paid to other agricultural contaminants such as pesticides. The present paper reports the results of a 1-year field monitoring of pesticide concentrations and water flows measured upstream and downstream from a fishpond in North East France to evaluate its capacity in reducing pesticide loads. Among the 42 active substances that had been applied on the fishpond's catchment, seven pesticides (five herbicides, two fungicides) were studied. The highest concentration in the inflow to the pond was 26.5 mu g/L (MCPA), while the highest concentration in pond outflow was 0.54 mu g/L (prosulfocarb). Removal rates of dissolved pesticides in the fishpond ranged from 0-8 % (prosulfocarb) to 100 % (clopyralid). Although not primarily designed for the treatment of diffuse sources of pesticides, the studied fishpond had the potential to do so.

While this configuration permits full nitrification, it also needs a large surface area (> 2 m²/pe). Therefore high ammonium removal efficiencies are of interest for compact systems such as single stage CW.The use of zeolite as filtrating medium could fix the problem thanks to itsexchange capacity and its high affinity for ammonium. Indeed, the ammonium is adsorbed during the feeding cycle then nitrified during the rest period.This study aims to assess the feasibility of ammonium removal by zeolite in a first stage VFF fed with raw sewage. Seven columns (40 cm-depth, 107 cm²) filled with zeolite under a gravel layer and one control without zeolitewere studied over four months. All columns, except a zeolite-control, were packed with inoculated media froma treatment plant. The columns have been operated in a sequential mode of feeding and rest periods of 3.5d and 7d, respectively, with synthetic solution of ammonium (100mgNH4-N/L). Two columns were fed with different concentrations of ammonium while two others were also fed sodium in order to assess the impact of the ammonium concentration and the presence of competitive species, respectively.
