Effects of the enzymes and osmoticum on the protoplasts isolation from major crops

Plant protoplasts were isolated from the mesophyll tissue and callus by the enzymatic method to obtain the basic informations for the protoplast culture, fusion, and the future genetic manipulation in plants. The optimal conditions for protoplast isolation varied according to the species. Large number of protoplast were readily isolated by the enzymes such as pectinase, macerozyme, cellulase, driselase, and hemicellulase. Among the cell wall degrading enzymes hemicellulase was appeared to be the most effective in isolating protoplast from the leaf sheath of rice. The osmoticum levels were also appeared to be an effective factor affecting the viability of protoplasts
