Conversion of starch to ethanol by immobilized glucoamylase gel-pellets and yeasts gel-pellets

An application of immobilized yeast and a modified technique of immobilized glucoamylase with calcium alginate gel were studied. Growing yeast cells were immobilized in calcium alginate gel pellets. Glucoamylase were immobilized in calcium alginate gel pellets with covalent bonds by the treatment with 20 percent enzyme solution, 0.12 M hexamethylenediamine and 1 percent glutaraldehyde. The optimum contact time of gel pellets with enzyme solution was 50 hr. Continuous production of ethanol from starch was carried out in reactor containing yeast gel pellets. By this system, the initial specific fermentation rate was about 21-22 g ethanol/hr.l-pellet, and a steady state was maintained for longer than 10 days at 2.5 and 2.75 of time factors
