NADH-dependent O2 consumption of purified plant /wheat/ plasma membranes

The O2 consumption of plasmalemma vesicles purified from the roots of spring wheat grown in the dark depends on the quantity of protein present in the reaction mixture. The NADH-dependent O2 consumption could not be inhibited either with rotence or with antimycin A, but was considerably reduced by KCN. Catalase also inhibited the NADH-dependent O2 consumption; the more, if added prior to NADH it completely eliminated the NADH stimulation. These results indicate that H2O2 could be a reaction product of NADH-dependent O2 consumption. Salicylhydroxamic acid /SHAM/ enhanced the O2 consumption of plasmalemma vehicles, but only in the presence of NADH. It is often used in in vivo experiments to measure the alternative oxidase capacity, whereas its use in experiments of bioenergetic nature is questionable because it substantially increases the NADH-dependent O2 consumption of the plasmalemma.
