Optimization of some parameters of gas chromatographic analysis of gasogenic galactosaccharides in bean

Influence of three procedures for extraction of raffinose, and stachyose on the degree of their isolation from bean seeds was studied using gas chromatography of trimethylsilylol (TMS) derativates of these sugars. Optimum condition of extraction was found to be heating of fragmented seeds with ethanol under reflux condenser. It was further found that trisaccharide melezitose may be used as an internal standard in gas chromatography of silylol derivates of raffinose and stachyose. Bean seeds were found to include about 5-7 times smaller content of raffinose, when compared with stachyose. Interspecies differences of stachyose content reach up to about 35 percent
