Asporogenic mutants of Bacilli: Isolation, characterization and their industrial application

In order to obtain enzyme preparations of the Bacillus sp. BPA-100 and BAT-300 (the sporogenic producers of alkaline proteinase and alpha-amylase, respectively) devoid of producer organisms, efforts were made to isolate spo mutants whose vegetative forms could be easily eliminated, spo mutants were isolated as chloroform-sensitive mutants. Two of the selected spo mutants BPA-101 (spo-11) and BAT-301 (spo-15) were blocked at stage O of the process of sporulation. The maximal relative yields of alkaline proteinase from strain BPA-101 and alpha-amylase from strain BAT-301 were approximately 30 and 50 % higher than those of their sporogenic parents, respectively. The viable cells BPA-101 and BAT-301 proved to be very sensitive to organic polyelectrolytes used for the coagulation and/or flocculation of substances from the fermentation broths. Thus, the final products were completely devoid of the asporogenic producer organisms.
