Karyological characterization of callus derived regenerated plantlets of Sesbania bispinosa (Jacq.) W.F. Wight

Competent calli were successfully induced from cotyledonary explants of diploid (2n=24) Indian dhaincha, Sesbania bispinosa (Jacq.) W. F. Wight and maintained on MS medium supplemented with 2,4-D (2 mg/1), BAP (0.5 mg/1) and coconut milk 10% (V/V). Plantlet regeneration and multiplication were also achieved from the competent calli with BAP (2 mg/a) and coconut milk 15% (V/V). Cytological study of the source plant as well as competent calli and regenerated plantlets were made both by chromosome counting and detail karyomorphological analysis. Original source plant revealed predominantly all diploid cells (2n=24). A high degree of diploid mitosis with least heterogeneous cells of aneuploids were also recorded in short- term culture of the competent calli. Comparative karyomorphological analysis did not reveal any difference between the source plant and callus derived regenerants. Within the limit of cytological techniques, present result highlights the possible strong selection in favour of plant regeneration from cytologically normal cells though heterogeneity of cells in competent calli were recorded. Regenerated plants also demonstrated a relative karyotypic stability.
