Serological control of sheep vaccinated with REV-1 Brucella vaccine by indirect Elisa

The indirect ELISA was applied to control the serological response of nine sheep three months old vaccinated with REV-1 brucella vaccine, crude S-LPS antigen derived from concentrated SAT antigen. Optical Density (OD)0.220 at 492 nm of 1:100 serum dilution was considered as threshold of positivity. One week postvaccination (PV) all sera except one were positive by Elisa as in RBT and SAT. In the second week PV the 9 sera were positive (mean titer 1:350). The antibody titer increased to maximum (mean titer 1:1000) at 4 weeks PV, it persisted at high level until 6 months PV and decreased gradually thereafter. No negativation was observed until 18 months PV, meanwhile 7/9, 5/9, and 2/9 sera were negative to SAT, RBT and CFT respectively. The above results show that: -Indirect Elisa seems to be as sensitive as the conventional assays at the beginning of immunisation of sheep with REV 1 vaccine. The ELISA detected antibodies persist longer than those detected by conventional assays. That makes difficult the use of Elisa for diagnosis in vaccinated flocks
