Inhibitory effect of hemicholinium-3 on the acetylcholine output and the peristaltic reflex in the isolated guinea-pig intestine

The effect of hemicholinium-3 on the output and synthesis of acetylcholine and peristaltic activity induced by distention was investigated in the isolated guinea-pig intestine. The increase of acetylcholine output and synthesis induced by circumferential or longitudinal distention was inhibited in the presence of hemicholinium-3. Hemicholinium-3 caused concentration-dependent inhibition of peristaltic activity. These inhibitions of acetylcholine metabolism and peristaltic activity were selectively antagonized by choline. It is concluded that the distention of intestinal wall elicits activation of the cholinergic nerve pathway in the intrinsic nerve plexues, leading to increase of the acetylcholine output from nerve fibers, and thus acetylcholine may be essential for the peristaltic reflex in some way
