Regeneration from lentil cotyledonary nodes and potential of this explant for transformation by Agrobacterium tumefaciens [Lens culinaris]

Shoots readily regenerated from lentil (Lens culinaris Medik. 'Laird') cotyledonary node explants in vitro on a medium containing benzylaminopurine (BAP). Shoots originated from the axils of the cotyledonary petioles in 100 of nodes, and the regeneration process was capable of continuing for at least 20 weeks. Approximately 50 of the shoots rooted on hormone-free, half strength B5 medium. Rooted shoots were capable of developing into mature fertile plants. A kanamycine dose of 50 mg/l significantly reduced shoot regeneration. The axils of cotyledonary petioles were not amenable to transformation via Agrobacterium tumefaciens, as measured by assays of the reporter gene B-glucuronidase (GUS)
