Effects of colchicine on aseptic culture of ginger (Zingiber officinale Rosc.)

When the shoots of ginger were treated in 0, 0.25, 0.50, 0.75 and 1.00 % sterile colchicine solution for 1, 2, 3 and 4 days, their survival rate and growth decreased when increasing concentration and duration. Colchicine caused morphological abnormalities on leaf and stem of VM2 and VM3 plants. Some plants with broader leaves and thicker stem and one plant with variegated leaves were observed. Root tip cell were cytologically studied and found that VM2 plants derived from VM1 plants treated with 0.25 % colchicine solution for 1 day and 0.50 % colchicine solution for 2 days were 50.00 and 33.33 % tetraploid respectively whereas VM3 plants derived from tetraploid VM2 plants were 60.00 % tetraploid and 40.00 % diploid.
