Plant regeneration from anthers of Oryza sativa L. cv. Jumli Marshi and Chhomrong - two cold tolerant rice varieties of Nepal

Panicles of cold tolerant rice cultivars - Jumli Marshi and Chhomrong - corresponding to uninucleate pollen stage were excised and pretreated at 8 deg. C for 0-7 days. For callus induction anthers were incubated at 25 +- 1 deg. C in dark in a modified Chu's N(6) medium containing 2.5 mg/l NAA, 0.5 mg/l Kn, 0.5 mg/l 2, 4-D. For plantlet regeneration calli were transferred to a modified MS medium supplemented with l mg/l each of Kn, NAA and BAP and incubated at 25 +- 1 deg. C in light. Frequency for callus induction and plantlet regeneration depended upon the genotype, number of days of cold pretreatment and the duration of anther- incubation period. In Jumli Marshi cold pretreatment for 4-5 days yielded the highest percentage of calli; no such effect was observed in Chhomrong. In Jumli Marshi cold pretreatment affected callus induction, but not its regeneration ability. Maximum yield of green plantlets per 100 anthers for Jumli Marshi and Chhomrong was 14.7 and 0.7, respectively.
