The influence of growing conditions on the in vitro development of sour cherry microplants

One method of vegetative propagation of fruit trees is micropropagation. In Estonia the meristem method is the means of micropropagation for plant propagation. That is most popular. A complicated problem in the meristem method is how to get high quality plant material with a stable capacity of rooting in vitro. In 1993 I arranged an experiment to find out how the light and temperature affect the proliferation and elongation of adventitious shoots in the multiplication phase and the subsequent rooting process. The experiment was arranged in the Estonian Plant Biotechnical Research Centre EVIKA. Sour cherry species "LaetiLeedu madalkirss" were used in the trials. Four combinations of light and temperature were explored. During observation and measurements significant differences between the four tested combinations of light and temperature appeared. Proliferation and elongation of adventitious shoots were better in the II variant as well as the quality of rooting. In that combination of conditions rooting of microplantlets was more-stable, the size and shape of the roots were better and the number of roots per microplant was higher compared with other tested combinations of light and temperature regimes. Summing up, the development of adventitious shoots and the subsequent rooting of microplantlets are tight dependent on growing conditions of plant material during multiplication phase
