Electroporation of potato (Solanum tuberosum L. cv. Desiree) protoplasts

The parameters for electroporation of the potato Solanum tuberosum L. cv. Desiree protoplasts were studied. At the same time, an improved protocol for regeneration of plants from leaf mesophyll protoplasts was established. The liquid ST medium B (Shepared and Totten, 1977) supplemented with 2.3 microM ZEA, 0.9 microM 2,4-D and 5.4 microM NAA was found to be the best for induction of cell divisions and formation of calli. The procedure described by Bokelmann and Roest (1983) proved to be the most suitable for further callus growth and shoot induction. The effect of different pulse duration times, number of pulses and voltages applied to protoplast suspensions were studied. A 20 micro sec pulse duration and two consequent pulses were chosen for further experimental conditions.
