Induction of multiple adventitious buds of maize [Zea mays] through vegetative organ culture in vitro

Effect of surface sterilized timing of node segments and shoot tip were 25 and 20 minutes respectively in 20 percent chlorox. Node segments and shoot tips from the maize plants 20-25 days old of Suwan 1 and Suwan 3 cultivars were brought to culture on the modified MS (1962) medium supplemented with various concentrations of BAP and IBA. Young shoot formation of Suwan 1, the highest number was eight shoots per one bud segment on the medium supplement with BAP 60 micro M. While in Suwan 3 cultivar, the highest number was 11.5 shoots when using BAP 70 micro M 60 micro M BAP combined with 0.005 or 0.05 micro M IBA. Both combinations produced 5.5 shoots in Suwan 1 cultivars, which were the highest number. In Suwan 3 the average of the highest number of 6.7 shoots was found in the medium with the sole BAP 80 micro M. Induction root system from the previous young shoot culture borne no root formation. The single shoot isolation of Suwan 3 cultivar on the MS (1962) medium varied with 4 concentration levels IBA with 3 concentration levels NAA. The suitable concentration was 0.5 micro M IBA with 25 micro M NAA.
