Phenotypic variability of Digitalis purpurea L. in vitro regenerated plants

A comparative study of in vitro-clones, regenerated through axillary bud multiplication, meristem culture or adventitious bud regeneration from callus, has been performed. Some morphological and biological indices, such as number of rosette leaves per plant, stem height, leaf length and width, plant weight, number of floriferous stems, flower colour, frost resistance and survival rate have been analysed during three consecutive years of vegetation in order to evaluate phenotypical differences and field performance of the forementioned populations. Plants regenerated from pre-existing organs are more homogeneous compared to callus regenerated plants, whose phenotypical variability was very high. Our results prove that "in vitro" techniques allow to regenerate rejuvenated plants. Micropropagated plants exceeded the control for all analysed parameters: they flowered earlier and even fruited in the third year of vegetation, while seedlings ceased their growth after two years in culture.
