Acute oral toxicity of the organophosphorus compound diethylphosphiric ester of dicyclopropyloketoxime and efficacy of antidotes on white rats

The acute oral toxicity of the new organophosphorus (OP) compound with insecticidal action, diethylphosphoric ester of dicyclopropyloketoxime, was studied on male and female Wistar rats, after single oral administration in three different carriers, namely corn oil, paraffin oil and Berol-948 plus water. Under the same experimental conditions, the efficacy of the antidotes atropine sulphate and 2-PAM (pyridine 2-aldoxime methiodine) was studied after intramuscular administration on the intoxicated animals. The experimental compound showed high acute oral toxicity on white rats. The antidote treatment increased LD50 values and affected the toxic symptoms a well. The combination of atropine sulphate plus 2-PAM controlled both the typical OP compound induced toxic symptoms and the more general toxic symptoms induced by almost all toxic substances, giving at the same time the best protection against the acute poisoning (LD50, males=85.9, females=47.5 mg/kg b.w.).
