Pathological changes in liver due to the toxicity of Ageratum conyzoides (babadotan)

英语. Liver injury is a consistent pathological change found intoxicated with Ageratum conyzoides. Adult Wistar rats showed a persistent reduction of body weight gain for 28 days of feeding on 20 percent A. conyzoides diet. Macroscopically liver appeared to have mottling on the capsular surface, and this lesion was the only pathological change found in rats. Megalocytosis and proliferation of bile suct cell were noted in liver microscopically. Similar clinical signs were also seen in rats fed on 10 percent of A. conyzoides diet for six weeks of feeding. Pathological changes did not appear in the liver during the first week of feeding. The pathological changes started to develop in the second week of feeding including anisokaryocytosis, proliferation of bile duct cells and infiltration of mononuclear cells inside and/or outside the veins and arteries of liver in the second week of feeding. Megalocytosis appeared in the third week followed by marked anisokaryocytosis and bile duct cells proliferation. On week-4, focal hepatic necrosis with bile duct cells proliferation were seen in the liver. Bile duct cells proliferation became more extensive in the portal tract where the interstitium of portal tracts were thickened and infiltrated by bile duct cells in the fifth week. The liver lesions were more prominent in sixth week of feeding. Supplementation of diets with normal diet led to liver regeneration in rats. Hepatic cells regeneration was seen in day-2 after the supplementation as being more basophilic and contained with granular materials in the cytoplasms followed by the infiltration of Kupffer cells and lymphocytes in sinusoids and portal tracts. Liver appeared to recover on day -5, but proliferation of bile duct cells were still present until this study was terminated at day-14

印度尼西亚. Kerusakan jaringan hati merupakan perubahan patologis yang konsisten ditemukan pada hewan yang mengalami keracunan tanaman Ageratum conyzoides (babadotan). Tikus putih jantan dewasa dari strain Wistar menunjukkan penurunan berat badan setelah pemberian 20 persen daun babadotan setiap hari selama 28 hari berturut-turut. Secara makroskopik hati memperlihatkan kelainan seperti bintik-bintik putih (mottling) pada permukaan kapsular, sedangkan organ lain tidak ditemukan kelainan patologis yang spesifik. Pengamatan mikroskopis dijumpai megalositosis sel hati dan proliferasi sel-sel saluran empedu. Gejala klinis yang sama juga ditemui pada tikus yang diberi pakan 10 persen daun babadotan. Kelainan patologis belum terbentuk pada minggu pertama; sedang anisokaryositosis, proliferasi sel-sel saluran empedu dan infiltrasi sel monokultur di dalam dan/atau di luar vena porta dan vena centralis terjadi dua minggu setelah pemberian daun babadotan. Megalositosis mulai terbentuk pada minggu ke-3 yang diikuti oleh anisokaryositosis yang nyata serta proliferasi sel-sel saluran empedu.
