A survey of canine parvovirus (CPV) antibody in dogs of Miyazaki city [Japan] and application of the double sandwich ELISA for detection of anti-CPV antibody

One hundred and thirty two dog sera collected from 127 dogs were tested for anticanine parvovirus (CPV) antibodies by haemagglutinin inhibition (HI) test. HI titers varied from 1:4 to 1:4,096 and approximately 10% of sera tested revealed high titers (1:1,000) indicating prevalence of CPV infection among dogs in Miyazaki city. We also applied a sensitive ELISA (Sandwich ELISA) where antigens were fixed with rabbit anit-CPV antibody to the plastic plates beforehand. High and specific responses were obtained by sandwich ELISA when compared with indirect ELISA developed by others. Furthermore a slight increase in IgM (anti-CPV antibody) level was easily detected by the sandwich ELISA even when HI antibody had not fully increased yet. This seems to be a useful method for clinical diagnosis of CPV infection
