Development of polymorphic DNA pattern analysis method using multiple or repetitive genes in the silkworm, Bombyx mori

We examined whether DNA polymorphism among commercial silkworm parent races, Asa, Hi, Fu, Yoh, Toh, Kai, Kouhaku and Hei, could be detected by using transposon, mag and k 1.4, as probes. The result showed that many polymorphic bands could be appeared when the parent silkworm DNAs were digested with the restriction enzymes recognizing nucleotide sequences which are absent or present at only one site within these transposon sequences. We showed that these patterns could be illustrated and analyzed by using Image analyzer (Millipore Co.). The genetic similarity among the races and phylogenetic relationships were also calculated. The genetic relationships obtained in these calculations agreed very well with the pedigree of these races. We concluded that this method is very sensitive to detect the polymorphic DNA patterns and that the analysis of these patterns gives the values of genetic similarity, showing useful relationships among commercial parent races
