Plantlet regeneration from immature coconut inflorescence in Laguna Tall

Explants from immature inflorescences of Laguna Tall with outer spathe lengths ranging from 70-150 mm produced pre-embryogenic nodules on Y3 callus induction medium after 3-4 months in culture. Depending on the physiological stage of development of the inflorescence, the optimum concentration of 2,4-dichlorophenoxyacetic acid (2,4-D) varied from 10 to the minus 4th power to 2 x 10 to the minus 4M in the presence of 0.25 percent activated charcoal (AC) which played a key role in minimizing browning. As cytokinin source, N to the 6th power - (2-isopentenyl) adenine (21 P) was added at 10 to the minus 6th power M to further alleviate the browning problem, although it has been shown that pre-embryogenic nodules can be produced in the absence of any cytokinin. A protocol was developed at Albay Research Center [Philippines] for embryoid maturation and germination consisting of a reduction of the 2,4-D concentration and a simultaneous increase in the level of 2i P. Root formation was enhanced with the addition of 7 x 10 to the minus 5th power M indole-3 butyric acid (IBA) in the last subculture. A number of complete plantlets with single and multiple shoots have been obtained using this protocol
