Production of vincristine-like alkaloid in callus culture of Catharanthus roseus BCR-1

A unique culture line of Catharanthus roseus callus which was designated as BCR-1 was established, and routinely subcultured in a hormone-free B5 medium. Preliminary alkaloid analysis following the thin-layer chromatography procedures of Farnsworth et.al. (1964) showed the presence of several indole alkaloids, and one of which is the blue spot suspected as vincristine. This vincristine-like alkaloid was isolated and subjected to a cytotoxicity assay employing Vero cells. Results revealed that it is highly cytotoxic, similar to that of the authentic vincristine. By HPLC, vincristine-like alkaloid was estimated to constitute 38 percent of the crude extract i.e. several times higher than in intact plant. On the basis of the present results, it can be concluded that C. roseus BCR-1 callus is a potential source of clinically-important alkaloids, including the anti-tumor alkaloid vincristine
